Detection of Near-membrane Calcium Ions in Live Tissues with a Two-Photon Fluorescent Probe

A two-photon fluorescent probe (ACaCL) that can detect near-membrane $Ca^{2+}$ is reported. This probe can be excited by 780 nm fs pulses, shows high photostability and negligible toxicity, and can visualize near-membrane $Ca^{2+}$ in live cells and deep inside live tissues by two-photon microscopy.
異常環境下의 營養問題 硏究(第2報) : 振動이 Serum Cholesterol 量에 미치는 影響

혈청 cholesterol량은 비단 식사의 성분에 의하여 영향 받을 뿐만이 아니라 개체의 주위환경조건에 따라서도 많은 영향을 받을 것으로 사려 되기 때문에 진동이 혈청 cholesterol량에 미치는 영향과 또한 제1보 때와 마찬가지로 Caesin, 비타민류, α-Tocopherol, 인삼분말 등의 방호효과도 아울러 검토하였기에 결과를 다음과 같이 결론한다. 1. Basal diet군(백미식군)은 각 첨가군에 비하여 Serum cholesterol량이 낮다. 그러나 인삼첨가군의 Serum cholesterol량이 타군에 비하여 가장 낮은 것은 흥미로운 점이다. 2. 단백질첨가군, 비타민류 첨가군 및 α-Tocopherol 첨가군의 Serum cholesterol량은 Basal diet군의 그것보다 현저히 높다. 3. α-Tocopherol 첨가군을 제외하고 다른 모든 군의 경우 진동군이 비진동군에 비해서 Serum cholesterol량이 현저히 높다. 4. α-Tocopherol 첨가군의 경우에는 진동군과 비진동군 간에 별 차이가 없는 것으로 보아 α-Tocopherol은 진동으로부터의 Serum cholesterol량 변화를 막아주는 것으로 사려 된다.

<P><B>Background</B></P><P>Clinical sequencing data should be shared in order to achieve the sufficient scale and diversity required to provide strong evidence for improving patient care. A distributed research network allows researchers to share this evidence rather than the patient-level data across centers, thereby avoiding privacy issues. The Observational Medical Outcomes Partnership (OMOP) common data model (CDM) used in distributed research networks has low coverage of sequencing data and does not reflect the latest trends of precision medicine.</P><P><B>Objective</B></P><P>The aim of this study was to develop and evaluate the feasibility of a genomic CDM (G-CDM), as an extension of the OMOP-CDM, for application of genomic data in clinical practice.</P><P><B>Methods</B></P><P>Existing genomic data models and sequencing reports were reviewed to extend the OMOP-CDM to cover genomic data. The Human Genome Organisation Gene Nomenclature Committee and Human Genome Variation Society nomenclature were adopted to standardize the terminology in the model. Sequencing data of 114 and 1060 patients with lung cancer were obtained from the Ajou University School of Medicine database of Ajou University Hospital and The Cancer Genome Atlas, respectively, which were transformed to a format appropriate for the G-CDM. The data were compared with respect to gene name, variant type, and actionable mutations.</P><P><B>Results</B></P><P>The G-CDM was extended into four tables linked to tables of the OMOP-CDM. Upon comparison with The Cancer Genome Atlas data, a clinically actionable mutation, p.Leu858Arg, in the <I>EGFR</I> gene was 6.64 times more frequent in the Ajou University School of Medicine database, while the p.Gly12Xaa mutation in the <I>KRAS</I> gene was 2.02 times more frequent in The Cancer Genome Atlas dataset. The data-exploring tool GeneProfiler was further developed to conduct descriptive analyses automatically using the G-CDM, which provides the proportions of genes, variant types, and actionable mutations. GeneProfiler also allows for querying the specific gene name and Human Genome Variation Society nomenclature to calculate the proportion of patients with a given mutation.</P><P><B>Conclusions</B></P><P>We developed the G-CDM for effective integration of genomic data with standardized clinical data, allowing for data sharing across institutes. The feasibility of the G-CDM was validated by assessing the differences in data characteristics between two different genomic databases through the proposed data-exploring tool GeneProfiler. The G-CDM may facilitate analyses of interoperating clinical and genomic datasets across multiple institutions, minimizing privacy issues and enabling researchers to better understand the characteristics of patients and promote personalized medicine in clinical practice.</P>

Diets rich in flavonoid-containing foods are sometimes associated with cancer, neurodegenerative and cardiovascular disease prevention. β-glucan, one of the most important bioactive compounds in medicinal mushrooms, is used to boost the immune system, diabetes and cancer. In this study, we aimed to determine the content of two flavonoid compounds (naringenin and naringin) and β-glucan in Flammulina velutipes and Flammulina velutipe-gold strain. The naringenin, naringin, and β-glucan components from the Flammulina velutipes and Flammulina velutipe-gold strain were measured by the HPLC analysis. As a result, the contents of naringenin, naringin, and β-glucan were found to be much higher in Flammulina velutipe-gold strain than Flammulina velutipe. From the above results, we may suggest that Flammulina velutipe-gold strain might have useful as a material for functional food and pharmaceutics fir the pathological process of various human diseases.
수삼양경 락혈의 깊이별 침자가 백서의 nNOS, NO 및 Norepinephrine의 변화에 미치는 영향

Objectives: This study was intended to observe the changes in the expression of neurotransmitters, such as nNOS, NO and NE upon the needle insertion at varying depths at the connecting point. Methods : Needles were inserted into rats, on both left and right sides of the connecting point, including the LI6, SI7 and TE5 acupoints which are three yang meridians of the hand. After insertion, needles were retained for five minutes. Each acupuncture groups were treated acupuncture at each acupoint and at the depths of superficial, middle and deep layer. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessels were extracted to examine the changes in the expression of nNOS, NO and NE. Results : In terms of the effect in nNO production, there was a significant increase only in the middle and deep layer at SI7 acupoint, but there was no significant change in the expression of NO. Regarding the formation of norepinephrine within tissues, the middle layer on LI6 acupoint, the middle layer and the deep layer on TE5 acupoint showed a significant increase, while production of plasma norepinephrine was significantly decreased at the middle layer and the deep layer on LI6 acupoint and the deep layer on SI7 acupoint. Conclusions : The effect of needles applied at the connecting point of three yang meridians on the activities of nNOS, and NE could be observed, and it can be induced that the effect of needle stimulation on disrupted nervous system can be examined through additionalresearches based on this one.

Due to the shortage of donor organs, complicated medical problems, or for economic reasons, the number of end-stage renal failure(ESRF) patients requiring maintenance dialysis in Korea has been consistently increasing. Despite the rapid growth of renal transplanation(RT), less than 20% of patients have had the privilege of RT. Therefore, the importance of HD as a maintenance or bridging measure until RT should not be overlooked even in the era of chimera. To improve patient survival and quality of life, comfortably located, well functioning, potentially permanent angioaccess is mandatory. This review summarizes our 14 year experience with internal vascular access procedures, especially focusing on the importance of early failure(EF). For the successful vascular access for maintenance HD, selection of types of surgery according to the size or status of vein is important to reduce EF, which is one of the major factors influencing long-term patency. In every sense, side to end radiocephalic fistula(SERCF) at wrist level should be tried initially. But if the cephalic vein is not suitable, there is no reason to explore the wrist, because, even after successful anastomosis, venous outflow might be inadequate to prevent cannulation. In this situation, the brachio cephalic fistula(BCF) at the antecubital fossa have to be preferred as the primary choice. We could achieve more than 70% of 3- year success rates with the RCF or BCF if the vein was good. More perfect techniques and attentions are necessary to reduce early complications such as bleeding or wound problems.
