      • Increased expression of ICAM-3 is associated with radiation resistance in cervical cancer

        <P>To search for a marker that predicts the efficacy of radiation therapy in human cervical cancer, gene expression profiles between parental SiHa cervical cancer cells and radiation-resistant SiHa/R cells have been compared by the microarray technique. Microarray and Northern blot analyses demonstrated that the ICAM-3 expression was upregulated in SiHa/R cells. This increased expression of ICAM-3 in SiHa cells enhanced cell survival by about 34.3% after a 2 Gy dosage of radiation. In addition, SiHa/ICAM-3 cells showed a 2.45-fold higher level of FAK phosphorylation than that of the control cells. In tumor specimens, ICAM-3 staining was restricted to tumor stromal endothelial cells and lymphocytes. The overexpression of ICAM-3 was significantly more frequent in radiation-resistant cervical cancer specimens when compared with radiation-sensitive specimens (83.3% vs. 35.3%; p = 0.015). With these observations, we can suggest that an increased expression of ICAM-3 is associated with radiation resistance in cervical cancer cells and the expression of ICAM-3 can be used as a valuable biomarker to predict the radiation resistance in cervical cancer that occurs during radiotherapy. © 2005 Wiley-Liss, Inc.</P>

        <P><B>Summary</B></P> <P>The extremely low efficiency of human embryonic stem cell (hESC) derivation using somatic cell nuclear transfer (SCNT) limits its potential application. Blastocyst formation from human SCNT embryos occurs at a low rate and with only some oocyte donors. We previously showed in mice that reduction of histone H3 lysine 9 trimethylation (H3K9me3) through ectopic expression of the H3K9me3 demethylase Kdm4d greatly improves SCNT embryo development. Here we show that overexpression of a related H3K9me3 demethylase KDM4A improves human SCNT, and that, as in mice, H3K9me3 in the human somatic cell genome is an SCNT reprogramming barrier. Overexpression of KDM4A significantly improves the blastocyst formation rate in human SCNT embryos by facilitating transcriptional reprogramming, allowing efficient derivation of SCNT-derived ESCs using adult Age-related Macular Degeneration (AMD) patient somatic nuclei donors.         It has been well known that several kinds of intracranial lesions can be associated with cerebral aneurysms. Among them, each case of cerebral arteriovenous malformation, moyamoya disease and pituitary adenoma has been respectively reported as associated lesions with cerebral aneurysms. And we have added the other two cases of sylvian arachnoid cyst and nodular enhancing lesion, which might be coincidental association with cerebral aneurysms. We have reviewed literatures and discussed associated lesions with cerebral aneurysms.

        Advantages of laparoscopy in gynecologic surgery in elderly patients

        Objective Geriatric patients requiring gynecological surgery is increasing worldwide. However, older patients are at higher risk of postoperative morbidity and mortality, particularly cardiopulmonary complications. Laparoscopic surgery is widely used as a minimally invasive method for reducing postoperative morbidities. We compared the outcomes of open and laparoscopic gynecologic surgeries in patients older than 55 years.Methods We included patients aged >55 years who underwent gynecological surgery at a single tertiary center between 2010 and 2020, excluding vaginal or ovarian cancer surgeries were excluded. Surgical outcomes were compared between the open surgery and laparoscopic groups, with age cutoff was set at 65 years for optimal discriminative power. We performed linear or logistic regression analyses to compare the surgical outcomes according to age and operation type.Results Among 2,983 patients, 28.6% underwent open surgery and 71.4% underwent laparoscopic surgery. Perioperative outcomes of laparoscopic surgery were better than those of open surgery in all groups. In both the open and laparoscopic surgery groups, the older patients showed worse overall surgical outcomes. However, age-related differences in perioperative outcomes were less severe in the laparoscopic group. In the linear regression analysis, the differences in estimated blood loss, transfusion, and hospital stay between the age groups were smaller in the laparoscopy group. Similar results were observed in cancer-only and benign-only cohorts.Conclusion Although the surgical outcomes were worse in the older patients, the difference between age groups was smaller for laparoscopic surgery.         This study recorded the process of recovery from damage. Therefore this study was aimed to get a basic data of vegetation restoration through tracking survey of recovering process after reform construction. In early days after forest road was constructed, there were bare or almost bare ground. Recently vegetation status was surveyed 50 years after it was constructed. Part of forest in the site of this survey is hardly avoiding damage because road improvement is being expected. Therefore damage of vegetation is expected. After 50 years in bare ground, it was restored as total 168 species of 62 families such as 15 species of 7 families in pterophyta, 5 species of 2 families in gymnosperms, and 148 species of 53 families in angiosperms were occurred. Mixed forests of coniferous and broad leaves near forest road were recovered as 31 species such as Abies sachalinensis and Tilia japonica were occurred. If already existed forest roads were reconstructed, damage could be occurred again in very high possibility. Therefore environmentally-friendly construction is required through long-term monitoring.

        본 연구는 개방형혁신과 공유가치창출간의 관계를 규명하여 공유가치창출에 있어 개방형혁신의 중요성을 인식하는데 그 목적을 두고 연구의 진행을 하였다. 본 연구의 결과는 다음과 같다. 첫째, 개방형 혁신 요소 중 정보원천의 활용, 사용자 참여의 결과요인으로 예측한 공유가치창출이 유의한 결과를 나타냈다. 이로써 다양한 정보원천과 사용자 참여혁신을 활용한 개방형 혁신이 이루어질수록 경제적, 사회적 가치를 동시에 창출하는 공유가치창출 성과도 높을 것으로 예측할 수 있다. 둘째, 개방형 혁신 요소 중 하나인 협력 파트너 다양성은 공유가치창출과 유의하지 않은 관계로 나타났다. 특히, 기업의 공유가치창출을 위하여 다양한 정보원천의 활용을 장려하는데 시사점을 제공할 수 있다. 기업은 기업활동의 가치사슬에 대한 근원적 변화를 통하여 공유가치를 창출할 수 있다(Porter and Kramer,2011). 가치사슬은 환경에 대한 영향, 직원들의 건강과 안전에 대한 보장 등 다양한 사회적 이슈들과 연결되어 있다. 다양한 정보원천은 이러한 가치사슬 속의 사회적 이슈를 해결해나갈 수 있는 통합적인 시각을 갖는데 필요할 것이다.

        As the result realized the main component analysis by extracting the 31 basic geomagnetic bodies, which can be divided in forestry zones of basic geometric body of Fukuoka prefecture, it was divided in 7 kinds in forestry space. With the character appeared according to each form, the first factor is eternal pace management type of the 8 basic geomagnetic bodies like Kurogi; the second factor is asset maintenance of the 8 geomagnetic bodies like Nakagawa; the third factor is suburban type of the 5 geomagnetic bodies like Chikusino; the fourth factor is forestry mountain village of the 4 geomagnetic bodies like Hoshino; the fifth factor is material production zone type of the 4 geomagnetic bodies like Amagi; the sixth factor is the interior zone production type like Yabe; and the seventh factor is the own region type like Ooshima. Therefore, in this paper it was desirably decided that forestry advancement was induced from the first and second factor, and the rests are connected among them.

      • CDK7 is a reliable prognostic factor and novel therapeutic target in epithelial ovarian cancer

        <P><B>Abstract</B></P> <P><B>Objective</B></P> <P>Cyclin-dependent kinase 7 (CDK7) engages tumor growth by acting as a direct link between the regulation of transcription and the cell cycle. Here, we investigated the clinical significance of CDK7 expression and its potential as a therapeutic target in epithelial ovarian cancer (EOC).</P> <P><B>Methods</B></P> <P>CDK7 expression was examined in 436 ovarian tissues including normal to metastatic ovarian tumors using immunohistochemistry, and its clinical implications were analyzed. Furthermore, we performed <I>in vitro</I> and <I>in vivo</I> experiments using CDK7 siRNA or a covalent CDK7 inhibitor (THZ1) to elucidate the effect of CDK7 inhibition on tumorigenesis in EOC cells.</P> <P><B>Results</B></P> <P>The patient incidence of high CDK7 expression (CDK7<SUP>High</SUP>) gradually increased from normal ovarian epithelium to EOC (<I>P</I> < 0.001). Moreover, CDK7<SUP>High</SUP> was associated with an advanced stage and high-grade histology (<I>P</I> = 0.035 and <I>P</I> = 0.011, respectively) in EOC patients and had an independent prognostic significance in EOC recurrence (<I>P</I> = 0.034). CDK7 inhibition with siRNA or THZ1 decreased cell proliferation and migration, and increased apoptosis in EOC cells, and this anti-cancer mechanism is caused by G0/G1 cell cycle arrest. In <I>in vivo</I> therapeutic experiments using cell-line xenograft and PDX models, CDK7 inhibition significantly decreased the tumor weight, which was mediated by cell proliferation and apoptosis.</P> <P><B>Conclusion</B></P> <P>Mechanistic interrogation of CDK7 revealed that it is significantly associated with an aggressive phenotype of EOC, and it has independent prognostic power for EOC recurrence. Furthermore, CDK7 may be a potential therapeutic target for patients with EOC, whether platinum sensitive or resistant.</P> <P><B>Highlights</B></P> <P> <UL> <LI> CDK7 inhibition has an anti-cancer effect on platinum-sensitive EOC. </LI> <LI> CDK7 inhibition could induce responsiveness to platinum chemotherapy in platinum-resistant EOC. </LI> <LI> CDK7 overexpression had independent negative prognostic value for disease recurrence of EOC. </LI> <LI> CDK7 might play a critical role in EOC tumorigenesis, and it also serves as a possible therapeutic target in EOC. </LI> </UL> </P>
