      • HPLC를 이용한 Alnus속 식물에서의 diarylheptanoids의 정량분석

        Quantitative determination of diarylheptanoids (oregonin and hirsutanonol), which were characteristic components of Alnus spp. has been conducted by High Performance Liquid Chromatography. The HPLC analysis was carried out using an YMC J'sphere ODS-H80 column [150×4.6mm, S-4μm. 80A(particle)] and a combination of acetonitrile and H2O was used as mobile phase in gradient solvent system. The results showed differnce in the species (Alnus japonica, A hirsuta and A. hirsuta var. sibirica)

        백서 Paraquat 두여에 의한 간과 폐 조직의 산화성 손상에서 Vitamin C와 Deferoxamine의 항산화 효과에 관한 연구

        Background: The toxicity of paraquat has been known to be caused by oxygen free radicals which leads to the lipid peroxidation and multiple organ failure. Although vitamin C has been known to be a potent antioxidant, recently there are numerous data which have shown that a low dose of vitamin C may act as a prooxidant due to the stimulation of the Fenton reaction with metal ions, which produces hydroxyl radicals. It has been reported that a deferoxamine in paraquat intoxication could reduce the production of the hydroxyl radicals by the inhibition of the Fenton reaction through the reduction of iron ion in tissue. The aim of this study was to evaluate the effect of the high and low dose of vitamin C and deferoxamine on lipid peroxidation and plasma TNF-α in paraquat intoxication. Methods: Female Sprague -Dawley rats were divided into seven groups: control group which was not given paraquat(20 mg/kg), P group which was given paraquat only, PVH group given paraquat and high dose of vitamin C(100 mg/kg), PVL group given paraquat and low dose of vitamin C(10 mg/kg), PVHD given paraquat, high dose of vitamine C and deferoxamine(100 mg/kg), PVLD given paraquat, low dose of vitamin C and deferoxamine, and PD given paraquat and deferoxamine. Animals were killed at 6 and 24 hours after treatment. Malondialdehyde(MDA), superoxide dismutase(SOD) and glutathione(GSH) contents, catalase activity, plasma TNF-α, and histologic changes in the lung and liver tissue were measured. Results: The lung histology in the PVH and PD or PVHD groups showed the significant decreases in the alveolar edema and interstitial thickness compared to the P group. The liver histololgy in the PVH and PVHD groups demonstrated marked differences in the central venous and sinusoidal dilatation compared to that of the P group. While the MDA levels of the lung and liver in the PVH and PD groups showed the significant reduction compared to that of the P group at 6 hours after treatment, all groups showed the significant changes compared to the P group at 24 hours. There was no significant change of the SOD levels of the lung and liver at 6 hours among all groups. At 24 hours, the SOD levels of the lung in PVH, PVL, and PVHD groups showed the significant increases compared to the P group. The increase of the SOD level in groups combined with deferoxamine, however, revealed a little reduction. The SOD level of the liver in PVH group only significantly increased compared to the P group at 24 hours. There was no significant change of the GSH level of the lung and liver among all groups at 6 hours. At 24 hours, the GSH level of the lung and liver were significantly increased in both PVH and PD group and PVH group, respectively, compared to the P group. Although the catalase activity of the lung was not significantly increased, that of liver was signiflcantly increased in both PVHD and PD groups compared to the P group at 6 hours. The catalase activities of the lung and liver were significantly increased in PVH, PD, and PVHD at 24 hours. The concentrations of the plasma TNF-α were slightly decreased at 6 hours and slightly increased at 24 hours compared to that of the P group, but they were not significant. Conclusion: This study showed that although the low dose of vitamin C had no effect, the high dose of vitamin C revealed a decrease of the MDA level and an increase of SOD, GSH, and catalase activity in the lung and liver tissues, and the effect of the high dose of vitamin C increased with time. The administration of the deferoxamine with or without high dose of vitamin C, however, significantly showed the inhibition of the lipid peroxidation and antioxidant effect and low dose vitamin C decreased the effect of deferoxamine. The effects of the vitamin C and deferoxamine on plasma TNP-α were not clearly shown.

      • 정신분열병 환자의 주관적 증상과 객관적 정신병리와의 관련성

        연구목적 : 주관적 증상평가도구인 K-FBF(Kyung Hee-Frankfurter Beschwerde-Fragebogen)가 정신분열병 환자의 어떤 증상 영역을 반영하는지, 특히 정신분열병의 병태생리와 밀접한 관련을 가지며 객관적 평가도구로는 파악이 어려운 음성 증상 또는 인지 장애의 측면들을 반영하는지 조사하고자, 정신분열병 환자의 K-FBF 결과를 객관적 정신병리 평가도구인 PANSS(Positive and Negative Syndrome Scale for Schizophrenia), 대표적인 자기보고식 다차원 증상목록(multidimensional self-report symptom inventory)인 간이정신진단검사(SCL-90-R)의 평가 결과와 비교 분석하였다. 방 법 : 고졸이상의 학력으로 DSM-IV진단기준에 의한 정신분열병 환자 중 급성기가 경과한 66명을 대사으로 연구를 수행하였다. K-FBF, SCL-90-R을 시행한 날, 두 명의 정신과 의사가 PANSS를 수행하였다. K-FBF의 질문항목에 대한 반응양상을 조사하고, 10개 소척도에 대하여 요인분석을 시행하였다. K-FBF와 연령, 성별, 병식과의 상관관계를 조사하였다. 또한, K-FBF, PANSS, SCL-90-R제반 척도간에 상관관계 분석을 시행하였다. 결 과 : K-FBF는 높은 연령일수록 호소하는 항목의 수나 그 정도가 감소하였으며, 유병기간을 조절변수로 삼았을 때도 동일한 결과를 보였다. K-FBF의 소척도를 요인분석하여 두개의 합산척도를 추출, 각각 자각된 고통(RD, Recognized Distress)과 지각왜곡(PA, Perceptual Alteration)으로 명명하였다. 위의 합산척도와 PANSS의 상관관계에서, K-FBF 표준화 총점을 조절변수로 하여 편상관을 조사한 결과 PA척도와 PANSS 음성척도가 상관관계를 보였다. 이는 Huber의 기본증상이란 정신분열병의 음성증상을 주관적 측면에서 접근한 것이라는 평가와 관련된다. 한편, K-FBF표준화 총점과 SCL-90-R총점은 강한 상관관계를 나타낸 반면, SCL-90-R총점과 PANSS총점은 비록 통계적으로는 유의하지만 매우 미미한 상관관계만을 보였다. 따라서 평가척도의 상관성은 그 내용보다는 평가하는 주체에 더 큰 영향을 받는다고 판단되었다. 결 론 : 본 연구결과 K-FBF로 측정된 Huber의 기본증상은 정신분열병의 음성증상을 보다 더 반영하는 것으로 평가되었으며, 또 객관적 평가도구인 PANSS보다는 자기보고형 도구인 SCL-90-R과 더 높은 상관관계를 보인 것으로 나타났다. 그러나, 본 연구결과의 일반화를 위해서는 다른 질병단계, 다른 인구학적 변인을 가진 환자들을 대상으로 한 연구가 계속되어야 할 것이다. Objectives : Kyung Hee-Frankfurter Beschwerde-Fragebogen(K-FBF) is a representative subjective symptom measuring tool of schizophrenic patients. We performed psychopathology measurements using K-FBF, Positive and Negative Syndrome Scale(PANSS) and Symptom Checklist-90-Revised(SCL-90-R) in patients with schizophrenia and then compared the results of these three examinations one another, in order to examine which psychopathologic aspects K-FBF might reflect and to measure the possibility whether this instrument reflect the negative symptoms or cognitive impairments, which are difficult to measure by conventional objective instruments measuring psychopathology. Methods : All 66 patients satisfying DSM-IV criteria for schizophrenia, were approached. Their education level was more than high school and they were not in the acute stage of schizophrenia. They performed K-FBF and SCL-90-R, and at the same day, they were interviewed by one of both psychiatrists using PANSS. We examined the patterns of K-FBF response and performed facor analysis about 10 subscales. We examined the correlation between K-FBF and age, sex and insight. Also, we performed correlation analysis among the K-FBF, PANSS and SCL-90-R. Results : As for K-FBF, with increasing age, the number of positive response items and the symptom severity were decreased. Even when controlling prevalence period as control variable, the correlation remained unchanged. Performing factor analysis to the K-FBF subscales, we created 2 factors, ie summation subscales. We named those recognized distress(RD) and perceptual alteration(PA) each. Using K-FBF standardized total scores as controlling variable, we performed partial correlation between PA and PANSS negative symptom scale. The result was PA and PANSS have statistically significant correlation. This correspons with the view that Huber's basic symptoms are reflecting subjective approach of negative symptoms of schizophrenia. While K-FBF standardized total scores and PANSS total scores had statistically strong correlations, SCL-90-R total scores and PANSS total scores had statistically significant, but very mild correlations. So, we had the judgement that the correlations among the psychopathology measurement tools are more influenced by the rater than the contents of tools. Conclusion : We concluded that Huber's basic symptoms, measured by K-FBF, reflect more negative symptoms of schizophrenia than other symptoms. And K-FBF has more correlation with the PANSS, objective psychopathology. Measuring tool, than the SCL-90-R, self-report quetionnaire. But, more studies using patients with another disease stages and another demographic variables are needed in order to generalize the results of this study.

      • SiH_2Cl_2/SiH_4 혼합가스를 사용하여 유도결합형 플라즈마 증착법으로 제작된 다결정 실리콘 박막의 제조 연구

        Polycrystalline silicon thin film have been deposited by an inductively coupled plasma chemical vapor deposition using SiH₂Cl₂/SiH₄/H₂ mixtures. The ICP source offers some advantages which include high electron density, simple design, good uniformity and lower ion bombardment on the growing surface. The poly-Si quality and deposition rate can be improved significantly by adding SiH₂Cl₂ into SiH₄ plasma. But an optimum flow rate ratio of SiH₂Cl₂to SiH₄seems to be ∼1. The Cl radicals appear to enhance the etching effect and promote the growth of crystalline phase. When both SiH₂Cl₂ and SiH₄ were used as the source gas, the deposition rate had a maximum value of 7.2 A˚/sec. The poly-Si film exhibited the volume fraction from Raman scattering of 87% and the SEM average grain size of ∼3000 A˚

      • ZnSe 박막의 성장온도에 따른 특성연구

        We have investigated the AnSe thin-film properties with growth temperature. The ZnSe thin-film samples were grown on GaAs substrates by molecular beam epitaxy system. The grown samples were characterized by double crystal X-ray diffractometry and photoluminescence. As increasing growth temperature, the grown ZnSe thin-film thickness was decreased with -1.5 nm/hr℃ by re-evaporation of An and Se molecules on the surface. From the result of photoluminescence measurements, the emission intensity ratio of I₂/X_hh peak was increased, which was explained in terms of the increased density of donor impurity from the Ga inter-diffusion at the interface.

        Chlorpromazine이 백서 해마에서 전기경련 충격에 의한 MAPK 타이로신 인산화에 미치는 영향

        Object : In order to examine the interaction mechanisms of electroconvulsive shock(ECS) and antipsychotic drug at the level of molecular biology, we observed the effect of chlorpromazine pre-treatment on the activation of mitogen activated protein kinase(MAPK) induced by electroconvulsive shock(ECS) in rat hippocampus Method : Male Sprague-Dawley rats were divided into 2 groups. To the experimental group chlorpromazine(20㎎/㎏) was given intraperitoneally, and to the control distilled water was given instead. Thirty minutes later, ECS was given and the hippocampus was dissected out 2 minutes thereafter. Immunoblotting with antiphosphotyrosine antibody was carried out, and the signal intensity at 42kDa band was quantitized using densitometer. The obtained result was compared by student t-test between the experimental and the control group. The absolute amount of MAPK was measured by immunoblotting with anti-MAPK antibody. Result : The tyrosine phosphorylation of MAPK reached peak at 2 minutes after ECS. However, in the chlorpromazine pre-treated group, the peak level of MAPK tyrosine phosphorylation was significantly attenuated(t=-3.12, df=14, p=0.008) compared to the control. In contrast to this, the absolute amount of MAPK did not differ between the pretreated and the control group. Conclusion : Chlorpromazine attenuated the tyrosine phosphorylation of MAPK by ECS. This finding seems to be related to the fact that chlorpromazine pre-treatment changed the cfos expression by ECS in rat brain. Antipsychotic drug and ECS might interact at the level of MAPK signal transduction system, and this might explained the observed synergistic effect of two treatment modality.

      • ZnSe/GaAs 이종접합 구조의 photoreflectance 연구

        We have studied the interface characteristics of undoped ZnSe/GaAs heterostructure, resulting from the strain between ZnSe and GaAs, using photoreflectance (PR) spectroscopy. For PR spectra measured at room temperature, the two peaks were observed at about 1.4 eV GaAs bandgap region originating from ZnSe/GaAs and GaAs/SI-GaAs interfaces and the shoulder peak was observed at about 2.7 eV ZnSe bandgap region originating from the separation between heavy hole and light hole. we could be determined the origin of two different features observed in the PR spectra by combining in-phase and out-phase measurements of Lock-in amplifier. From PR result, we knew that ZnSe epilayer was partially relaxed due to thick layer thickness over than the critical layer thickness (CLT) and the stress effect caused by these layer relaxation affected the ZnSe/GaAs heterointerface more than the interface of GaAs buffer/SI-GaAs substrate.

        The optical and electrical properties of Cl-doped ZnSe alloys have been studied by Hall effect and photoluminescence(PL) measurements. By using ZnCl_2 as dopant source, the high quality n-type ZnSe layers with electron concentration up to 1.4×10^19 ㎤ have been achieved. Hall mobility for an epilayer with n = 6.5×10^16 ㎤ are ∼400 ㎠/V.s at 300 K and ∼900㎠/V.s at 80 K. For heavily doped samples, Cl donors are partially compensated. The PL measurements were performed at 11 K and 300 K. The excitonic emission associated with a neutral donor is dominant in the samples doped with Cl concentration below ∼10^19 ㎤. The carrier compensation was discussed with V^Zn-Cl^Se complex as reported origin of self-activated(SA) emission in previous reports.

        불완전 전뇌 허혈후 소생의 백서 모델을 이용한 재관류 뇌혈류량과 허혈성 뇌조직 손상도와의 관계

        Background: Experimental data indicate that low-flow reperfusion following prolonged cardiocirculatory arrest may aggravate early cerebral microcirculatory reperfusion disorders. We investigated the influence of cerebral reperfusion flow change to the ischemic histopathologic damage of brain tissue after incomplete forebrain ischemia in rats. Materials and method: Anesthetized Sprague-Dawley rats were undergone ligation of both internal carotid artery by microvascular clamp for 10 minutes. After release of the clamp, reperfusion was started with several different flow levels (0, 10, 20, 30, 50, and 100%) of internal carotid artery comparing to pre-clamping phase using flowmeter. After 15minutes of reperfusion, rat brains were prepared by perfusion-fixation with 3% formaldehyde. Under light microscopic examination of Hematoxylin-Eosin stained tissue slide, histopathologic damage was examined at cortex, putamen, and hippocampus regions. Categorical hisotopathologic damage scores were derived in each regions by manual counts of ischemic neurons. Result: The histopathologic damage scores were 0, 10.2±0.5, 7.6±1.5, 5.9±1.4, 5.0±2.8, 3.5±0.7, and 1.0±0.0 in control, 0, 10,20, 30, 50, and 100% reperfusion groups, respectively(p<0.05). Conclusion: Our results showed significant increment of brain histopathologic damage scores along with decreasing amount of cerebral reperfusion flow after incomplete forebrain ischemia. We believe restoration of reperfusion flow to pre-ischemic level would be a critical component in attenuation of brain ischemic damage.
