Deep Geothermal-water Characteristics of Tertiary Pohang in South Korea

Pohang area displays a larger potential of geothermal energy with the highest heat flow of ~83 mWm<sup>-2</sup> in South Korea. This study aims to reveal geothermal characteristics of four wells(BH-1 to BH-4 wells) of ~2,000 m in depth in Pohang area, using geophysical logs. The geology of the study area is composed of tertiary mudstone of 200 - 359.1 m, tuff of 73 - 240 m, sandstone/mudstone of 46 - 907 m, rhyolite of 259 - 375 m, and andesitic volcanic breccia of 8 34 m in thicknesses from the surface, with granodiorite at bottom. By the result, temperature and maximum electrical conductivity are 69.5°C at 1,502.6 m and 1,162 μS/cm at BH-2, 44.4°C at 912.3 m and 1,105 μS/cm at BH-3, and 82.5°C at 1,981.3 m and 3,412 μS/cm at BH-4. Thermal conductivity values at saturated state are 2.14 - 3.95 W/m-K at BH-1 and 2.36 - 3.61 W/m-K at BH-4. ß (determining heat flow rate and up/down direction) values were estimated by using 1-D steady-state heat transfer equation and were determined as -0.77 - 0.99 with the geothermal gradients (Ks) of 42.5 - 46.3°C/km at BH-1, -3.15 - 3.05 with the Ks of 25.0 - 29.1°C/km at BH-2, -1.80 - 2.09 with the Ks of 20.0 - 23.0°C/km at BH-3, and -4.10 - 5.18 with the Ks of 30.2 - 39.0°C/km at BH-4. Most depths of all the wells showed upward heat transfer. Based on the geophysical logs, the main aquifer is located between 200 and 300 meters.

<P>Drug development using marine bioresources is limited even though the ocean occupies about 70% of the earth and contains a large number of biological materials. From the screening test of the marine sponge extracts, we found <I>Hyrtios</I> sp. sponge collected from Chuuk island, Micronesia. In this study, the <I>Hyrtios</I> sp. extract was examined for anticancer activity against human colorectal carcinoma RKO cells that are wildtype for p53 and RKO-E6 that are p53 defective. The <I>Hyrtios</I> sp. extract dose-dependently inhibited viability in both cell lines. Multinucleation as an indication of mitotic catastrophe was also observed. Cytotoxicity tests gave significantly different results for RKO and RKO-E6 cells after 48 h exposure to <I>Hyrtios</I> sp. extract. In RKO cells treated with <I>Hyrtios</I> sp. extract, cell death occurred by induction of p53 and p21 proteins. In p53-defective RKO-E6 cells, <I>Hyrtios</I> sp. extract decreased expression of JNK protein and increased p21 protein. These results indicate that <I>Hyrtios</I> sp. extract induced apoptosis <I>via</I> different pathways depending on p53 status and could be a good natural product for developing new anticancer drugs. </P>
Retrospective analysis of transbronchial cryobiopsy: the first report in Korea

Background: Interstitial lung diseases(ILD) are heterogeneous group of lung diseases that require a multidisciplinary approach through discussion with clinicians, radiologists, and pathologists. Surgical lung biopsy is the gold standard for pathologic diagnosis of ILD. Recently, the usefulness of transbronchial cryobiopsy (TBC) has increased in the assessment of ILD. As far as we know, there is no study in Korea on the role of TBC in ILD diagnosis, yet. This study first evaluated the significance of TBC in suspected cases of ILD in Korea. Methods: The medical records of 17 patients suspected of ILD who were performed TBC at Ulsan University Hospital from August 2016 to June 2019 were retrospectively evaluated. Results: Seventeen patients with a mean age of 67 years were included in this analysis and there were 47%(eight patients) of women. The 1.9 mm sized probe was used for three patients and 2.4 mm sized probe was used for 14 patients. The median number of specimens per procedure was four (range; 1-9). The mean diameter of the specimens was 0.5 cm(range; 0.2-1.2cm). The diagnostic yield of TBC was 47.1%(eight patients). The frequent histopathologic results included usual interstitial pneumonia(two patients), organizing pneumonia(two patients) and adenocarcinoma(two patients). In the multidisciplinary diagnosis, they were diagnosed with idiopathic pulmonary fibrosis(two patients), cryptogenic organizing pneumonia(two patients) and adenocarcinoma(two patients). Four of the nine patients with non-diagnostic histopathologic patterns were able to be diagnosed after a multidisciplinary review. As a result, the number of patients who could be diagnosed by TBC increased to 12 patients(70.6%). The complication occurred in eight patients including mild bleeding (three patients) and pneumothorax(four patients). Conclusions: The diagnostic yield of TBC in this study was 47.1% and increased to 70.6% after a multidisciplinary diagnosis. The value of TBC as the alternative methods of surgical lung biopsy should be evaluated through further study.

Staphylococcus aureus is an important foodborne pathogen that causes diverse diseases ranging from minor infections to life-threatening conditions in humans and animals. To further understand its pathogenesis, the genome of the strain S. aureus FORC_001 was isolated from a contaminated food. Its genome consists of 2,886,017 bp double-stranded DNA with a GC content of 32.8%. It is predicted to contain 2,728 open reading frames, 57 tRNAs, and 6 rRNA operons, including 1 additional 5S rRNA gene. Comparative phylogenetic tree analysis of 40 complete S. aureus genome sequences using average nucleotide identity (ANI) revealed that strain FORC_001 belonged to Group I. The closest phylogenetic match was S. aureus MRSA252, according to a whole-genome ANI (99.87%), suggesting that they might share a common ancestor. Comparative genome analysis of FORC_001 and MRSA252 revealed two non-homologous regions: Regions I and II. The presence of various antibiotic resistance genes, including the SCCmec cluster in Region I of MRSA252, suggests that this strain might have acquired the SCCmec cluster to adapt to specific environments containing methicillin. Region II of both genomes contains prophage regions but their DNA sequence identity is very low, suggesting that the prophages might differ. This is the first report of the complete genome sequence of S. aureus isolated from a real foodborne outbreak in South Korea. This report would be helpful to extend our understanding about the genome, general characteristics, and virulence factors of S. aureus for further studies of pathogenesis, rapid detection, and epidemiological investigation in foodborne outbreak.

<P>Although various anticancer drugs have been developed for the treatment of nonsmall cell lung cancer, chemotherapeutic efficacy is still limited. Natural products such as phytochemicals have been screened as novel alternative materials, but alternative funds such as marine bioresources remain largely untapped. Of these resources, marine sponges have undergone the most scrutiny for their biological activities, including antiinflammatory, antiviral, and anticancer properties. However, the biological mechanisms of the activities of these marine sponges are still unclear. We investigated the anticancer activity of marine sponges collected from Kosrae in Micronesia and examined their mechanisms of action using nonsmall cell lung cancer A549 cells as a model system. Of 20 specimens, the <I>Haliclona</I> sp. (KO1304-328) showed both dose- and time-dependent cytotoxicity. Further, methanol extracts of <I>Haliclona</I> sp. significantly inhibited cell proliferation and cell viability. A549 cells treated with <I>Haliclona</I> sp. demonstrated induced expression of c-Jun N-terminal kinase (JNK), p53, p21, caspase-8, and caspase-3. The percentage of apoptotic cells significantly increased in A549 cultures treated with <I>Haliclona</I> sp. These results indicate that <I>Haliclona</I> sp. induces apoptosis via the JNK-p53 pathway and caspase-8, suggesting that this marine sponge is a good resource for the development of drugs for treatment of nonsmall cell lung cancer.</P>

TNFα is an inflammatory cytokine required for immune responses, playing a crucial role in the pathogenesis of inflammatory autoimmune diseases via interactions with its cognate receptors, TNFR1 and TNFR2. Therapeutic antibodies targeting TNFα, including infliximab, adalimumab, certolizumab pegol, and golimumab, are clinically used for the treatment of the inflammatory diseases such as rheumatoid arthritis, psoriatic arthritis, Crohn’s disease, and inflammatory bowel disease. Here, the Fab fragment of golimumab was overexpressed in the periplasmic region of Escherichia coli and purified by affinity and gel-filtration chromatography. The purified protein was crystallized by hanging-drop vapor diffusion in the presence of 100 mM Tris, pH 8.5, 0.1 M ammonium phosphate, and 12% (w/v) PEG 6,000 at 20°C. The crystal diffracted X-ray to 2.60 Å resolution and belonged to the orthogonal space group P2 1 2 1 2, with unit cell parameters a = 72.74, b = 130.29, and c = 93.35 Å. An asymmetric unit of the crystal contains two molecules of the Fab fragment of golimumab with a V M of 2.14 Å 3 Da -1 and a solvent content of 42.48%.

In this study, a selecvively block barrier was developed to prevent the spread of contaminants (TPH, Phenol) in the ground only when contamination occurs. The materials were used Jumunjin sand, bentonite, polyolefin elastomer and spill hound marine. First, the properties and environmental hazards characteristics of materials were analyzed for evaluated their usability. Then, the possibility of use as a barrier material was confirmed by analyzing the water permeability characteristics that change after 24 hours of contact with contaminants. As a result of the analysis, the pH of each component was similar to the general groundwater pH range. In addition, the toxicity characteristics and the possibility of dissolution of hazardous substances, it was determined that there was no environmental hazard as the content was below the regulation value. Lastly, when comparing the permeability coefficient before and after contact with the contaminant, the permeability coefficient of approximately α × 10<sup>-3</sup>cm/sec before contact was reduced to α × 10<sup>-6</sup>cm/sec after contact with the contaminant.
