        Metagenomic insight of nitrogen metabolism in a tannery wastewater treatment plant bioaugmented with the microbial consortium BM-S-1

        <P>Nitrogen (N) removal in a tannery wastewater treatment plant was significantly enhanced by the bioaugmentation of the novel consortium BM-S-1. In order to identify dominant taxa responsible for N metabolisms in the different stages of the treatment process, Illumina MiSeq Sequencer was used to conduct metagenome sequencing of the microbial communities in the different stages of treatment system, including influent (I), buffering (B), primary aeration (PA), secondary aeration (SA) and sludge digestion (SD). Based on MG-RAST analysis, the dominant phyla were Proteobacteria, Bacteroidetes and Firmicutes in B, PA, SA and SD, whereas Firmicutes was the most dominant in I before augmentation. The augmentation increased the abundance of the denitrification genes found in the genera such as Ralstonia (nirS, norB and nosZ), Pseudomonas (narG, nirS and norB) and Escherichia (narG) in B and PA. In addition, Bacteroides, Geobacter, Porphyromonasand Wolinella carrying nrfA gene encoding dissimilatory nitrate reduction to ammonium were abundantly present in B and PA. This was corroborated with the higher total N removal in these two stages. Thus, metagenomic analysis was able to identify the dominant taxa responsible for dissimilatory N metabolisms in the tannery wastewater treatment system undergoing bioaugmentation. This metagenomic insight into the nitrogen metabolism will contribute to a successful monitoring and operation of the eco-friendly tannery wastewater treatment system.</P>

        DNA-Stable Isotope Probing Integrated with Metagenomics for Retrieval of Biphenyl Dioxygenase Genes from Polychlorinated Biphenyl-Contaminated River Sediment

        <B>ABSTRACT</B><P>Stable isotope probing with [<SUP>13</SUP>C]biphenyl was used to explore the genetic properties of indigenous bacteria able to grow on biphenyl in PCB-contaminated River Raisin sediment. A bacterial 16S rRNA gene clone library generated from [<SUP>13</SUP>C]DNA after a 14-day incubation with [<SUP>13</SUP>C]biphenyl revealed the dominant organisms to be members of the genera <I>Achromobacter</I> and <I>Pseudomonas</I>. A library built from PCR amplification of genes for aromatic-ring-hydroxylating dioxygenases from the [<SUP>13</SUP>C]DNA fraction revealed two sequence groups similar to <I>bphA</I> (encoding biphenyl dioxygenase) of <I>Comamonas testosteroni</I> strain B-356 and of <I>Rhodococcus</I> sp. RHA1. A library of 1,568 cosmid clones was produced from the [<SUP>13</SUP>C]DNA fraction. A 31.8-kb cosmid clone, detected by aromatic dioxygenase primers, contained genes of biphenyl dioxygenase subunits <I>bphAE</I>, while the rest of the clone's sequence was similar to that of an unknown member of the <I>Gammaproteobacteria</I>. A discrepancy in G+C content near the <I>bphAE</I> genes implies their recent acquisition, possibly by horizontal transfer. The biphenyl dioxygenase from the cosmid clone oxidized biphenyl and unsubstituted and <I>para</I>-only-substituted rings of polychlorinated biphenyl (PCB) congeners. A DNA-stable isotope probing-based cosmid library enabled the retrieval of functional genes from an uncultivated organism capable of PCB metabolism and suggest dispersed dioxygenase gene organization in nature.</P>

      • Bacterial community comparisons by taxonomy-supervised analysis independent of sequence alignment and clustering.

        <P>High-throughput sequencing of 16S rRNA genes has increased our understanding of microbial community structure, but now even higher-throughput methods to the Illumina scale allow the creation of much larger datasets with more samples and orders-of-magnitude more sequences that swamp current analytic methods. We developed a method capable of handling these larger datasets on the basis of assignment of sequences into an existing taxonomy using a supervised learning approach (taxonomy-supervised analysis). We compared this method with a commonly used clustering approach based on sequence similarity (taxonomy-unsupervised analysis). We sampled 211 different bacterial communities from various habitats and obtained 1.3 million 16S rRNA sequences spanning the V4 hypervariable region by pyrosequencing. Both methodologies gave similar ecological conclusions in that β-diversity measures calculated by using these two types of matrices were significantly correlated to each other, as were the ordination configurations and hierarchical clustering dendrograms. In addition, our taxonomy-supervised analyses were also highly correlated with phylogenetic methods, such as UniFrac. The taxonomy-supervised analysis has the advantages that it is not limited by the exhaustive computation required for the alignment and clustering necessary for the taxonomy-unsupervised analysis, is more tolerant of sequencing errors, and allows comparisons when sequences are from different regions of the 16S rRNA gene. With the tremendous expansion in 16S rRNA data acquisition underway, the taxonomy-supervised approach offers the potential to provide more rapid and extensive community comparisons across habitats and samples.</P>

        Human skin accommodates diverse bacteria and fungi that influence both healthy and diseased skin. Increasing evidence indicates that interactions between the skin microbiota, diet, and the host contribute to the development of types of skin diseases. Here, I will show the case studies to identify the structural signatures of microbiomes related to on acne vulgaris, dandruff/seborrhoeic dermatitis, skin sensitivity, and etc. Also, we applied microbial ecological theory to provide a framework for understanding crucial health-associated features of the human microbiome. Furthermore, we will identify and characterize an essential factor that plays a critical role in polymicrobial interactions in the skin microbiota and contributes to cutaneous health.

      • A Driving Method of Pixel Circuit Using a-IGZO TFT for Suppression of Threshold Voltage Shift in AMLED Displays

        <P>A driving method of pixel circuit using amorphous indium gallium zinc oxide (a-IGZO) thin-film transistor (TFT) is proposed to improve the image quality of active matrix light-emitting diode displays. The proposed pixel circuit employs a diode-connected structure to compensate for variation in threshold voltage (V-th) of the a-IGZO TFT. In addition, the proposed driving method adopts negative bias annealing to suppress the V-th shift. The annealing time is optimized based on the experimental observation of the minimum V-th shift. After a stress time of 30 000 s, the measurement results showthat the (Vth) shift is reducedby 29.6%, using an optimized annealing time of 5% of one frame time. In addition, the maximum deviation in the emission current using the proposed driving method wasmeasured to be less than 4.32% after a stress time of 30 000 s.</P>

        의료 환경의 수어통역 콘텐츠 개발 과정에 관한 연구

        본 연구는 전문적이면서 정확한 의료수어통역이 미흡한 상황에서 농인의 의료 서비스 접근성을 원활하게 하는 의료 환경의 수어통역 콘텐츠를 개발하는 것에 목적을 둔다. 본 연구의 목적을 달성하기 위해 문헌고찰, 농인 대상 개별 면접, 수어통역사 및 수어 전문가 대상 현장 요구도 조사, 의료 및 수어 전문가 자문을 진행하였으며, 이를 바탕으로 주된 진료 상황별 시나리오와 기본 의학용어, 의료용어 설명자료 등의 수어통역 콘텐츠를 개발하였다. 본 연구를 통해 농인의 상황과 의료적 중요성을 고려한 의료수어 콘텐츠를 개발하여 의료수어 영역의 전문성을 고취시켰으며, 의료용어 및 의료 정보 학습 주체인 농인들과 수어통역사들에게 효과적이며 효율적으로 정보를 전달하는 의학용어 수어사전 반응형 홈페이지를 개발하였고, 농인 당사자가 주체가 되는 수어번역의 필요성과 중요성을 깨닫게 되었다. The purpose of this study is to develop sign language interpreting content in the medical setting that facilitates Deaf people’s access to medical services in situations where professional and accurate medical sign language interpreting is insufficient. To achieve the purpose of this study, we conducted a literature review, individual interviews for Deaf people, on-site requirement surveys of sign language interpreters and sign language experts, and medical and sign language expert consultations. Based on this, we developed sign language interpreting content such as main care contextual scenarios, basic medical terms, and medical term descriptions. Through this study, we developed medical sign language content considering the situation and medical importance of Deaf people to promote expertise in the medial sign language area and developed a responsive website of sign language medical dictionary that effectively and efficiently delivers information to Deaf people and sign language interpreters; we realized the need and importance of sign language translation for Deaf people to be the main bodies.

        노인복지관 이용 노인들의 돌봄 욕구 분석

        본 연구는 통합연구방법을 활용하여 노인복지 현장에서 인식하는 돌봄을 서비스로 실천하기 위한 노인복지관의 역할을 모색하고자 한 것이다. 노인복지관에서 어떤 수준과 범위에서 돌봄을 서비스로 제공해야 하는지에 대한 접근을 수행하기가 쉽지 않기에 지역사회 돌봄 체계에서 노인복지관의 역할과 정체성이 재정립될 필요가 있다. 이러한 현실에 근거하여 본 연구는 B노인종합복지관을 이용하는 당사자들이 인식하는 돌봄은 무엇이며, 노인의 욕구에 부합하는 실제적인 돌봄의 내용을 살펴보고자 질적연구와 양적연구를 혼합하여 진행하였다. 질적연구는 FGI를 진행하여 근거이론 방법으로 범주화 분석하였으며, 양적연구는 B노인종합복지관 이용자 300명을 대상으로 설문조사 후 통계 분석하였다. 연구결과, 질적분석에서는 노인의 활기찬 삶과 건강관리, 노인의 활기찬 삶과 B노인복지관, 노인의 활기찬 삶과 사회참여, 노인의 활기찬 삶과 평생교육, 노인의 활기찬 삶의 마무리가 주요 쟁점으로 제시되었다. 양적분석에서는 돌봄서비스의 우선순위와 고려되어야 할 돌봄의 구체적 내용이 도출되었다. 양적분석 결과를 토대로 질적연구의 결과와 내재시켜 노인 당사자가 필요로 하는 돌봄의 패러다임을 살펴보고 사회문화적 돌봄을 실천하기 위한 노인복지관의 역할을 제언하였다. This study used integrated research methods to explore the role of senior welfare centers in implementing the care recognized in the senior welfare field as a service. Because it is not easy to determine what level and scope of care should be provided as a service at senior welfare centers, the role and identity of senior welfare centers need to be redefined in the community care system. Based on this reality, this study was conducted through a mixture of qualitative and quantitative research to examine what care is perceived by those using Senior Welfare Center B and the content of actual care that meets the needs of the elderly. The qualitative study was conducted through FGI and categorized and analyzed using the grounded theory method, and the quantitative study was conducted through statistical analysis after surveying 300 users of B Senior Welfare Center. As a result of the FGI analysis, the active life and health care of the elderly, the active life of the elderly and B elderly welfare center, the active life and social participation of the elderly, the active life and lifelong education of the elderly, and the conclusion of the active life of the elderly were presented as major issues. In the quantitative analysis, the priorities of care services and specific contents of care to be considered were derived. Based on the results of quantitative and qualitative research, the paradigm of care required by the elderly party was examined, and the role of elderly welfare center for the elderly to practice social and cultural care was suggested.
