      • Medicinal Chemistry : EVALUATION OF CADMIUM-INDUCED NEPHROTOXICITY USING URINARY METABOLOMIC PROFILES IN SPRAGUE-DAWLEY MALE RATS

        The aim of this study was to investigate urinary metabolomics profiles associated with cadmium (Cd)-induced nephrotoxicity and their potential mechanisms. Metabolomic profiles were measured by high-resolution 1H-nuclear magnetic resonance (NMR) spectroscopy in the urine of rats after oral exposure to CdCl2 (1, 5, or 25 mg/kg) for 6 wk. The spectral data were further analyzed by a multivariate analysis to identify specific urinary metabolites. Urinary excretion levels of protein biomarkers were also measured and CdCl2 accumulated dose-dependently in the kidney. High-dose (25 mg/kg) CdCl2 exposure significantly increased serum blood urea nitrogen (BUN), but serum creatinine (sCr) levels were unchanged. High-dose CdCl2 (25 mg/kg) exposure also significantly elevated protein-based urinary biomarkers including osteopontin, monocyte chemoattractant protein-1 (MCP-1), kidney injury molecules-1 (Kim-1), and selenium-bingding protein 1 (SBP1) in rat urine. Under these conditions, six urinary metabolites (citarate, serine, 3-hydroxyisovalerate, 4-hydroxyphentllactate, dimethylamine, and betaine) were involved in mitochondrial energy metabolism. In addition, a few number of amino acids such as glycine, glutamate, tyrosine, proline, or phentlalanine and carbohydrate (glucose) were altered in urine afrer CdCl2 exposure. In particular, the metabolites involved in the glutathione biosynthesis pathway, including cysteine, serine, methionine, and glutamate, were markedly decreased compared to the control. Thus, these metabolites are potential biomarkers for detection of Cd-induced nephrotoxicity. Our results further indicate that redox metabolomics pathways may be associated with Cd-mediated chronic kidney injury. These findings provide a biochemical pathway for better understanding of cellulat mechanism underlying Cd-induced renal injury in hunmans.

      • S. cerevisiae와 A. pullulans를 이용한 수용액 중의 납 제거에서 온도와 pH의 영향

        The environmental factors, temperature and pH, were controlled to investigate the effects on the removal of lead in the solution by S. cerevisiae and A. pullulans. The range of experimental temperature was 20℃∼50℃, and that of Ph was 2.5∼6.5 and 2.5∼12, respectively. The optimum temperature of S. cerevisiae and A. pullulans in the removal of lead was 30℃ and 40℃, respectively. And the initial removal rate of lead by, S. cerevisiae was increased according to the increase of temperatue. The variation of the amounts of lead removal by S. cerevisiae according to the variation of temperature was steeper that by A. pullulans. The optimum pH condition was existed on both of the microorganisms, S. cerevisiae and A. pullulans, in our experimental range and that is 4.5 and 9.0, respectively.

        The experiments were performed in a high temperature furnace to examine the effects of the temperature difference between the single round jet and the surrounding fluid on the characteristics of impinging heat transfer. It was found that the average heat transfer coefficient increased in proportion to 1.5th power of the jet velocity, which was higher than that of previous measurements in which the jet temperature was the same as the ambient temperature. since the jet is thermally mixed with its surroundings, the heat transfer increases as the nozzle-to-surface distance decreases. The maximum average heat transfer occurred at the nozzle-to-surface distance normalized by the nozzle diameter.

        Bacillus sphaericus ts-D1290 단백질의 전기영동적 분석

        Bacillus sphaericus ts-D1290 치사돌연변이주의 특성을 연구하기 위하여 제한온도와 허용온도에서 생산하는 단백질의 차이점을 연구하였고, 모기유충에 치사력을 가진 단백질을 분리하여 모기유충에 치사되는지를 조사하였다. ts-D1290 균주를 42℃에서 배양했을 때는 30℃배양보다 균체의 단백질 함량이 약 1/5로 감소하였고, 42℃에서 4시간 배양 후 30℃로 배양하였을 때는 약 1/3로 감소하였다. 그리고 단백질의 패턴은 ts-D1290 균주를 42℃에서 전배양 후 30℃로 하강 배양하였을 때는 30℃에서 전배양하여 42℃로 상승배양하였을 때와는 달리 분자량 221kd 단백질이 나타나지 않았지만 분자량 150kd 단백질은 30℃ 전 배양하였을 때와 같이 현저하게 나타났다. 두 균주에 80㎍/㎖ 농도의 chloramphenlcol을 첨가하여 42℃에서 배양하였을 때 정상균주는 30℃에서 배양한 대조군보다 단백질 함량이 1/2로 감소하였고, ts-D1290 균주는 30℃에서 배양하였을 때는 첨가하지 않았을 때보다 분자량 150kd 단백질이 약하게 나타났다. ts-D1290과 정상균주가 생산한 150kd단백질은 모기유충에 치사성을 나타냈다. Bacillus sphaericus ts-D1290 was characterized by SDS-PAGE produced by the mutant at 30℃ and 42℃. The total amount of proteins produced by the mutant at 42℃ decreased to one-fifth of those at 30℃; however, when the culture was shifted down from 42℃ after 4 to 30℃, the total amount of protein decreased to one-third and the 221 kd protein did not appear, but the 155 kd appeared remarkably. When the mutant and the wild type strain were cultured in the media containing 80㎍ per ㎖ of chloramphenicol at 42℃, the wild type strain synthesized half amounts of the total proteins than those at 30℃, and the mutant produced one-tenth of the total protein amounts. When the both strains were cultured in the media containing chloramphenicol, the 155 kd protein was produced was produced in lesser amounts than those without chloramphenicol. The 150 kd protein showed lethal activity to Culex pipiens 3rd instar larvae.

      • 디바이스 전압제어를 통한 동적 소모 전력 관리

        배터리의 용량이 한정된 이동기기에서는 배터리의 효율을 높이기 위한 전력 절감 기법에 관련한 기술이 중요시된다. 배터리의 용량을 증대시키기 위한 기존의 방법으로 시스템 전체의 평균 동작 전압을 감소시킴으로써 전력 소비량을 줄이는 방법이 제시되어 왔다. 이는 프로세서의 저전력 정책으로 시스템의 상태에 따라 프로세서의 주파수와 공급 전압을 조절함으로써 시스템 전체의 전력 소비를 감소시켜 결과적으로 배터리의 사용량을 증대시킨다. 본 연구에서는 현재 프로세서에만 사용되어 오던 주파수와 공급 전압의 조절을 배터리의 잔량에 따라 각 디바이스에 적용한 DPM(Dynamic Power Management)을 제시함으로써, 프로세서의 전력 관리보다 시스템 차원에서 배터리의 특성을 고려한 부가적 전력 절감 기법을 제시한다.

        Choanal polyps are a separate clinical entity. While the etiology of choanal polyps is unclear, they present a fairly uniform clinical picture. Maxillary sinus is the usual site of origin, but in rare cases, a choanal polyp may originate from a different site, e.g, turbinate sinus, anteroinferior surface of the ethmoidal bulla, sphenoethmoidal recess, or sphenoid sinus. A 37 year-old woman was referred with 6 months history of nasal obstruction. Anterior rhinoscopic examination revealed the large polyp on the posterior portion of nasal cavity extending to the choana. Endoscopic and radiologic evaluation was taken, and FESS was performed. Polyp was removed completely. We report this case with review of the literature.

        In case of the occurrence of osteonecrosis at three or more anatomical location, it is defined as multifocal osteonecrosis, but still any king of study with references hasn't been reported in Korea. The patient who had admitted due to the pain in both knee joints, both hip joints and left ankle joint and we diagnosed as multifocal osteonecrosis by using bone scan and magnetic resonance imaging. To relieve right knee joint and left hip joint pain, we perfomed arthroplasty. Here we present one case report of multifocal osteonecrosis through analysis by patient's history and hematologic examination and provide a review of the literature.

        Bacterial strains showing the fibrinolytic activity were screened from Doenjang (Korean traditional soybean paste). The strain 5,6, 13 and 15 isolated from Doenjang showed a high level of caseinolytic and fibrinolytic activity. The fibrinolytic activities of strain 5, 6, 13, and 15 were 32.9, 31.0, 22.3, and 23.2 unit, respectively. A strain 5 among the four strains showed the highset fibrinolytic activity (32 unit), and subsequently identified as Bacillus subtilis. The fibrinolytic strain was designated as Bacillus subtilis KS-5.

      • 미생물 유래 β-glucan 분리 · 정제 및 이를 이용한 오염물질 정화용 복합 미생물제재 개발

        β-glucan occuring mainly in mushrooms and fungi are attractive candidates for numerous industrial application due to their pharmacological activity such as antitumor activity via immunomodulating behavior and excellent rheological properties such as emulsification, viscosity, jellation, etc. Screening of β-glucan producing microorganism was performed by congo red dye binding assay of ethanol-precipitates from each culture filtrate. The β-glucan produced form the selected strains was purified mainly by directly diluting the culture broth with the same volume of water, removal of cells by centrifugation at 4,000×g for 20 mim, and precipitation with 3 volumes of ethanol. The monosaccharide composition was analyzed by either TLC or HPLC afer trifuoroacetic acid hydrolysis. The molecular mass of the β-glucan of HH-71 strain was estimated to be about 4,500,000 dalton on a GFC-HPLC chromatogram. A Bacillus protease coated with β-glucan showed 3-times more increased thermal stability comparing to the uncoated control when stored at 30℃ for 1 week. In a similar manner coating of wastewater (containing chlorinated organic compounds) treatment bacteria with β-glucan resulted in significant increase of thermal stability. Although more detailed experimental data need to be accumulated, these preliminary data clearly suggest that β-glucan is applicable as a coating material to stabilize activity or viability of industrial enzymes and microorganisms for treatment of wastewater. Based on these results, β-glucans obtained from HH-71 and HH-87 strains are currently investigated for coating ablility of microbial consortia for the treatment of wastewater.
