Enantiomeric separation of naproxen is necessary in drug evaluation processes as the pharmacological activity of naproxen resides mainly in the (S)-enantiomer, The enantioseparation of naproxen was carried out with three different stationary phases. The methods were validated with the terms of linearity, intraday and interday precision, accuracy, selectivity and recovery. Among the selected stationary phases, ChiroSil DMB column with hexane : t-butylmethylether : acetic acid (60:40:0.1, v/v/v) as mobile phase showed the best enantioseparation of the naproxen in the commercial preparations. The wavelength for detection was 270 nm and flow rate was 2.0 mL/min. The contents of (S)-naproxen in the commercial preparations were 92.7 - 98.3%, indicating that all tested preparations were satisfactory to current regulation. However, the contents of (R)-naproxen as impurity in the commercial preparations were varied from 0.08% to 1.08%.
PREPARATION OF CINOBUFAGIN-LOADED BOVINE SERUM ALBUMIN NANOPARTICLES FOR HEPATOCARCINOMA THERAPY

Cinobufagin-loaded bovine serum albumin nanoparticles were prepared for treating hepatocellular carcinoma. In this report, cinobufagin-bovine serum albumin-nanoparticles (Cino-BSA-NP) were prepared by an aqueous desolvation process. The physicochemical properties, toxicity, and cancer-related applications of Cino-BSA-NP were investigated. Cino-BSA-NP had a uniform spherical morphology with a particle size in the range of 50–240 nm and an average size of 86.3 nm. The zeta potential of the nanoparticles was -49 mV. The overall embedding ratio was 79.5% and the drug loading was 24.1%. Cino-BSA-NP gave cinobufagin release of up to 53.5% within 3 h, followed by slower controlled release. Cino-BSA-NP inhibited growth of hepatocarcinoma cells in vitro to a similar extent as free cinobufagin, but with a much higher median lethal dose (LD50). Hepatic histomorphological changes indicated that hepatic damage was much less severe with Cino-BSA-NP than with free cinobufagin (2.19 mg/kg). The survival time of nude mice with orthotopic transplantation tumors treated with Cino-BSA-NP was prolonged significantly. The results confirm that Cino-BSA-NP renders cinobufagin completely dispersible in aqueous media, meeting the key requirements for intravenous injection, and show controlled release, thus significantly improving cinobufagin's antitumor activity while reducing its side effects.

Insulin, transferrin and selenium (ITS) complex is reported to improve in vitro development of oocytes and embryos. This study was carried out to investigate the effects of ITS during in vitro culture (IVC) of porcine parthenogenetic and nuclear transfer (NT) embryos on subsequent developmental capacity in vitro. The electrically activated oocytes were cultured in Porcine Zygote Medium (PZM-3) with various concentrations (0, 0.1, 0.5, and 1.0%) of ITS for 7 days. Also, the electrically activated reconstructed embryos were cultured in PZM-3 with various concentrations (0, 0.1, 0.5, and 1.0%) of ITS for 6 days. Addition of ITS to culture medium did not affect development of porcine parthenogenetic embryos in vitro. To test the effect of ITS on the in vitro development of porcine NT embryos, factorial experiments were also performed for in vitro maturation (IVM) medium (TCM-199) with or without 1% ITS and culture medium (PZM-3) with or without 0.5% ITS. Addition of 0.5% ITS to culture medium increased (p<0.05) the proportion of NT blastocysts compared with non-treated group. In contrast, addition of 1% ITS to culture medium was ineffective or had a detrimental effect. Also, addition of ITS only to maturation medium increased (p<0.05) the percentage of NT blastocysts formation compared with the control group. In conclusion, addition of ITS to IVM or IVC medium could improve subsequent blastocyst development of porcine NT embryos.

Interspecies somatic cell nuclear transfer (iSCNT) is a valuable tool for studying the interactions between an oocyte and somatic nucleus. The object of this study was to investigate the developmental competence of in vitro-matured porcine oocytes after transfer of the somatic cell nuclei of 2 different species (goat and rabbit). Porcine cumulus oocytes were obtained from the follicles of ovaries and matured in TCM-199. The reconstructed embryos were electrically fused with 2 DC pulses of 1.1kV/cm for $30{\mu}s$ 0.3M mannitol medium. The activated cloned embryos were cultured in porcine zygote medium-3 (PZM-3), mSOF or RDH medium for 7 days. The blastocyst formation rate of the embryos reconstructed from goat or rabbit fetal fibroblasts was significantly lower than that of the embryos reconstructed from porcine fetal fibroblast cells. However, a significantly higher number of embryos reconstructed from goat or rabbit fetal fibroblasts cultured in mSOF or RDH, respectively, developed to the morular stage than those cultured in PZM-3. These results suggest that goat and bovine fetal fibroblasts were less efficacious than porcine-porcine cloned embryos and that culture condition could be an important factor in iSCNT. The lower developmental potential of goat-porcine and porcine-bovine cloned embryos may be due to incompatibility between the porcine oocyte cytoplasm and goat and bovine somatic nuclei.

A chiral mobile phase HPLC method is described for chiral separation and determination of chlorpheniramine (CP) enantiomers in various commercial preparations. Chromatographic separation was achieved on a conventional ODS column with a mixture of aqueous sodium phosphate (5 mM) containing 0.5 mM carboxymethyl-β-cyclodextrin, methanol and triethylamine (73:25:2, v/v/v, pH 4.3) as the mobile phase. The flow rate of isocratic elution was 0.24 mL/min and peaks were detected at 224 nm. The method was applied to nine commercial CP preparations in six dosage forms and CP enantiomers were well separated without any disturbance of other ingredients or impurities present. The results showed that only one preparation was d-CP and the others were dl-CP preparations. The contents of all the preparations were found to be in the range of 97%-104% of labeled contents. This method was economical and convenient, affording sufficient accuracy, precision and reproducibility, as well as sensitivity and selectivity.
Thiolated Eudragit-Coated Chitosan Microspheres as an Oral Drug Delivery System

<P>The aim of this study is to prepare mucoadhesive chitosan microspheres for protein drug to deliver to intestine through oral administration. The thiolated Eudragit was synthesized by reaction between L-cysteine hydrochloride and Eudragit® L-100. About 8 mol-% of cysteine was introduced to the Eudragit-cysteine conjugate. The conjugate was used to coat bovine serum albumin (BSA)-loaded chitosan microspheres. The average particle sizes of BSA-loaded thiolated Eudragit-coated chitsoan microspheres (TECMs) were 4.06±0.74 .m and the uniform sizedistribution was shown. The in vitro release of BSA from BSA-loaded TECMs was pH-dependent. Our results indicated that thiolated Eudragit might be a good candidate as a coating material for oral delivery of protein drug owing to mucoadhesive and pH-sensitive properties.</P>
Structure and property of a vertical cutting Ca0.4Sr0.6Bi4Ti4O15 ferroelectric ceramic

Ca0.4Sr0.6Bi4Ti4O15 ceramics with a-axis preferred orientation were prepared by a solid-state reaction method. The cylindrical Ca0.4Sr0.6Bi4Ti4O15 ceramics were cut into two types of plates: one was along the cylinder axis (A), and the other was perpendicular to the direction of the cylinder axis. The effects of sintering temperature on the structure and electrical properties of the Ca0.4Sr0.6Bi4Ti4O15 ceramics were investigated. When the sintering temperature was 1180 oC, sample A gave the highest a-axis (200) orientation with a value of about 27.1%. At this sintering temperature, sample A showed better dielectric properties and ferroelectric properties, the relative dielectric constant (εr), dielectric loss (tan δ), remnant polarization (Pr), and coercive field (Ec) were 217.6, 0.0027, 14.2 μC/cm2 and 49.6 kV/cm, respectively.
