Late Mesozoic granites are extensively distributed in the Luanchuan district of eastern Qinling and can be divided into two types: Late Jurassic to Early Cretaceous granites (ore-related plutons) and Late Cretaceous granites (Laojunshan batholith). This study presents new geochemical and zircon U-Pb-Hf isotopic data from the Shibaogou and Yuku plutons to provide robust constraints on the petrogenesis and tectonic significance of the late Mesozoic granites in the Luanchuan district. Zircon U-Pb dating results yielded weighted mean 206Pb/238U ages of 149.1 ± 0.8 Ma and 150.5 ± 0.8 Ma, which were interpreted as the crystallization ages of the Shibaogou and Yuku plutons, respectively. We propose that the late Mesozoic granites contain high concentrations of SiO2 and alkali elements (Na2O + K2O) and feature metaluminous to weakly peraluminous characteristics. Enrichment in light rare earth elements and large ion lithophile elements and depletion in high field strength elements are observed. Mineralogical and geochemical evidence reveal that the late Mesozoic granites are highly fractionated I-type granites with fractional crystallization of feldspar, plagioclase and accessory minerals (e.g., apatite and titanite or magnetite). Based on the Hf composition, we suggest that the parental magmas of the ore-related plutons were derived from remelting of the Taihua and Xiong’er groups with minor contributions of mantle-derived materials and that the Laojunshan batholith was generated by the hybridization of ancient crust- (Kuanping group) and mantle-derived components. Collectively, the above arguments indicates a tectonic transition from compression to post-collisional extension during the late Mesozoic, that was likely triggered by the continental collision of the North China Block and the Yangtze Block, which generated numerous contemporaneous granites and Mo-W-Pb-Zn-Ag-Au poly-metallic deposits.
Mutation of the lbp-5 gene alters metabolic output in Caenorhabditis elegans

Intracellular lipid-binding proteins (LBPs) impact fatty acid homeostasis in various ways, including fatty acid transport into mitochondria. However, the physiological consequences caused by mutations in genes encoding LBPs remain largely uncharacterized. Here, we explore the metabolic consequences of lbp-5 gene deficiency in terms of energy homeostasis in Caenorhabditis elegans. In addition to increased fat storage, which has previously been reported, deletion of lbp-5 attenuated mitochondrial membrane potential and increased reactive oxygen species levels. Biochemical measurement coupled to proteomic analysis of the lbp-5(tm1618) mutant revealed highly increased rates of glycolysis in this mutant. These differential expression profile data support a novel metabolic adaptation of C. elegans, in which glycolysis is activated to compensate for the energy shortage due to the insufficient mitochondrial -oxidation of fatty acids in lbp-5 mutant worms. This report marks the first demonstration of a unique metabolic adaptation that is a consequence of LBP-5 deficiency in C. elegans. [BMB Reports 2014; 47(1): 15-20]

Purpose: the study aimed to compare the quality of life (QOL) and radiotherapy complications among Chinese nasopharyngeal carcinoma (NPC) patients at different 3-dimensional conformal radiotherapy (3DCRT) stages adjusting for other variables. Methods: 511 NPC patients at different 3DCRT stages were enrolled. They were interviewed regarding SF-36, complications and socio-demographic variables and cancer- or treatment-related variables. Analysis of covariance (ANCOVA) based on SF-36, complications scores as dependent variables, 3DCRT stages as independent variables, and other variables as covariate were established. Results: The influencing factors of PCS included 3DCRT stages and age group. The influencing factors of MCS included 3DCRT stages and income. Most QOL scores of NPC patients were significantly associated with 3DCRT stage, after accounting for other variables. QOL scores of the patients receiving 3DCRT were the lowest, QOL scores of people after 3DCRT gradually increased. PCS scores of people greater than 5 years after 3DCRT was improved to or even better than the level before 3DCRT. The complications with significantly different scores of patients at different 3DCRT status included xerostomia, throat ache, hypogeusia, caries, hearing loss, snuffles. Conclusions: Clinicians should pay more attention to older NPC patients and patients with lower income. When patients receive 3DCRT, measures should be taken to reduce radiation injury to improve the patients' QOL.

Inhibition of acetylcholinesterase (AChE) is one of the important modes of action available for the control of insects. An assessment was made of the electric eel AChE inhibitory activity of 55 endophytic fungi isolated from the leaves of Huperzia serrata collected in Eunshi, Hubei Province, China. Of 55 mycelial extracts and 55 ethyl acetate-soluble fractions from liquid culture of the fungi, the ethyl acetate-soluble fraction of strain ES056 exhibited strong AChE inhibitory activity (88%) at a concentration of 100 μg/mL. Based on morphological characteristics and nuclear ribosomal DNA ITS sequence analysis, this fungus showed greatest similarity to members of the order Eurotiales and was shown to be most closely related to members of the Aspergillus aculeatus group. It was designated Aspergillus sp. MY056. This strain merits further study as a potential anti-AChE product.

In this study, the nematode-trapping fungus, Monacrosporium sphaeroides, was transformed with a plasmid harboring the hygromycin B phosphotransferase gene, via restriction enzyme-mediated integration (REMI). Frequencies of up to 94 transformants ${\mu}g^{-1}$ per linearized plasmid DNA were obtained by optimizing the PEG concentration, as well as the category and quantity of the added restriction enzyme. $90\%$ of the transformants were determined to be stable for drug resistance when 20 randomly selected transformants were tested. Southern analyses revealed that the transforming DNA was integrated into the M. sphaeroides genome either with or without rearrangement. Five mitotic stable mutant strains were obtained using this approach, all of which had been altered with regard to sporulation capacity and pathogenicity toward nematodes. Southern blot analyses of the five mutants revealed that foreign plasmid DNA had integrated into the genome. Three of the mutants, Tms2316, Tms3583 and Tms1536, exhibited integration at a single location, whereas the remaining two, Tms32 and Tms1913, manifested integration at double or multiple locations. Our results suggest that the transformation of M. sphaeroides via REMI will facilitate insertional mutagenesis, the functional analysis of a variety of genes, and the tagging or cloning of genes of interest.

<P>The potential benefits of generating and using transgenic cattle range from improvements in agriculture to the production of large quantities of pharmaceutically relevant proteins. Previous studies have attempted to produce transgenic cattle and other livestock by pronuclear injection and somatic cell nuclear transfer, but these approaches have been largely ineffective; however, a third approach, lentivirus-mediated transgenesis, has successfully produced transgenic livestock. In this study, we generated transgenic (TG) Korean native cattle using perivitelline space injection of viral vectors, which expressed enhanced green fluorescent protein (EGFP) systemically. Two different types of lentiviral vectors derived from feline immunodeficiency virus (FIV) and human immunodeficiency virus (HIV) carrying EGFP were injected into the perivitelline space of MII oocytes. EGFP expression at 8-cell stage was significantly higher in the FIV group compared to the HIV group (47.5%2.2% v.s. 22.9%2.9%). Eight-cell embryos that expressed EGFP were cultured into blastocysts and then transferred into 40 heifers. Ten heifers were successfully impregnated and delivered 10 healthy calves. All of these calves expressed EGFP as detected by in?vivo imaging, PCR and Southern blotting. In addition, we established an EGFP-expressing cell line from TG calves, which was followed by nuclear transfer (NT). Recloned 8-cell embryos also expressed EGFP, and there were no differences in the rates of fusion, cleavage and development between cells derived from TG and non-TG calves, which were subsequently used for NT. These results illustrate that FIV-based lentiviruses are useful for the production of TG cattle. Moreover, our established EGFP cell line can be used for additional studies that involve induced pluripotent stem cells.</P>

야생 균주 Xanthophyllomyces dendrorhous으로부터 biomass와 astaxanthin 산물을 증가시키기 위해 shake flask배양 조건에서 astaxanthin산물에 영향을 미치는 물리적 인자, 영양 인자와 Carotenogenesis의 생성 자극 인자에 대한 연구를 HPLC분석 방법을 이용하여 수행하였다. 최적에 온도, 초기 pH값, 탄소와 질소 공급 조건에서 Carotenogenesis의 선구 물질인 acetic 산, mevalonic 산, 토마토 추출물과 당근 추출물은 astaxanthin 생산을 상당히 증가시킬 수 있었다. To improve biomass and astaxanthin production by wild-type Xanthophyllomyces dendrorhous simultaneously in shake flask culture, physical factors, nutritional factors and carotenogenesis stimulating factors affecting astaxanthin production were studied on base of HPLC quantitative analysis. The results suggested that carotenogenesis precursor composition acetic acid, mevalonic acid, tomato extract, and carrot extract could increase the productivity of astaxanthin markedly based on the optimized temperature, initial pH value, carbon and nitrogen sources conditions.
