Gamete Biotechnology is a science that studies on developing peer techniques for manipulating mammalian gametes and embryos. This is an applied science related to various fields of research area such as embryology, reproductive physiology and medicine, animal science, and biology. Developed technologies in this scientific area greatly contributes to improving the efficacy of animal husbandary and to establishing human reproductive medicine. Moreover such techniques will mostly accelerate to develop peer medical therapeutics such as cell therapy and tissue transplantation.
S.D. Rats를 이용 1, 1-Dichloro-1-fluoroethane의 아민성 흡입독성 연구

There were no specific effects for test materials on Sprague-Dawley (S.D.) rats in clinical symptoms, amounts of food intakes, weight changes, laboratory findings, and pathology after whole body l, l-Dichloro-1 -fluoroethane (used as coolant, metal cleaner and solvents) exposure(0, 1,500, 3,000, and 6,000 ppm) for 13 weeks (6 hour/day, 5 days/week). However, the loss of capillary vessels in eyeball (pupil) was observed in a female rat among 6,000 ppm group. Though there was a tendency for MCHC (Mean Corpuscular Hemoglobin Concentration) in rat to be decreased (p<0.05), it was not regarded as abnormal because the values were within normal limits. In asthma-stimulation related evaluations, there was also a tendency for inflammatory cell counts in bronchoalveolar lavages to be increased. But it had no statistical significance, and also no dependency on sex and the exposed concentration. Based on this result, the non observed effect level (NOEL) induced by 1, 1-Dichloro-l -fluoroethene inhalation was evaluated in groups with 3,000 ppm below (S .D. Rats, 13 weeks). Finally, it was concluded that the short term exposal of 1, 1-Dichloro-l-fluoroethane is not considered as a asthma stimulant by inhalation despite of some study limitations such as test animals use and short-term exposure .
Stem cell maintenance in a different niche

To overcome the difficulty of controlling stem cell fate and function in applications to regenerative medicine, a number of alternative approaches have been made. Recent reports demonstrate that a non-cellular niche modulating the biophysical microenvironment with chemical factors can support stem cell self-renewal. In our previous studies, early establishment was executed to optimize biophysical factors and it was subsequently found that the microgeometry of the extracellular matrix made huge differences in stem cell behavior and phenotype. We review here a three-dimensional, non-cellular niche designed to support stem cell self-renewal. The characteristics of stem cells under the designed system are further discussed.
One-step synthesis of gene carrier via gamma irradiation and its application in tumor gene therapy

<P><B>Introduction</B></P><P>Although numerous studies have been conducted with the aim of developing drug-delivery systems, chemically synthesized gene carriers have shown limited applications in the biomedical fields due to several problems, such as low-grafting yields, undesirable reactions, difficulties in controlling the reactions, and high-cost production owing to multi-step manufacturing processes.</P><P><B>Materials and methods</B></P><P>We developed a 1-step synthesis process to produce 2-aminoethyl methacrylate-grafted water-soluble chitosan (AEMA-g-WSC) as a gene carrier, using gamma irradiation for simultaneous synthesis and sterilization, but no catalysts or photoinitiators. We analyzed the AEMA graft site on WSC using 2-dimensional nuclear magnetic resonance spectroscopy (2D NMR; 1H and 13C NMR), and assayed gene transfection effects in vitro and in vivo.</P><P><B>Results</B></P><P>We revealed selective grafting of AEMA onto C6-OH groups of WSC. AEMA-g-WSC effectively condensed plasmid DNA to form polyplexes in the size range of 170 to 282 nm. AEMA-g-WSC polyplexes in combination with psi-hBCL2 (a vector expressing short hairpin RNA against <I>BCL2</I> mRNA) inhibited tumor cell proliferation and tumor growth in vitro and in vivo, respectively, by inducing apoptosis.</P><P><B>Conclusion</B></P><P>The simple grafting process mediated via gamma irradiation is a promising method for synthesizing gene carriers.</P>
