        Heteroexpression and Functional Characterization of Glucose 6-Phosphate Dehydrogenase from Industrial Aspergillus oryzae

        The engineered Aspergillus oryzae has a high NADPH demand for xylose utilization and overproduction of target metabolites. Glucose-6-phosphate dehydrogenase (G6PDH, E.C. 1.1.1.49) is one of two key enzymes in the oxidative part of the pentose phosphate pathway, and is also the main enzyme involved in NADPH regeneration. The open reading frame and cDNA of the putative A. oryzae G6PDH (AoG6PDH) were obtained, followed by heterogeneous expression in Escherichia coli and purification as a his6-tagged protein. The purified protein was characterized to be in possession of G6PDH activity with a molecular mass of 118.0 kDa. The enzyme displayed maximal activity at pH 7.5 and the optimal temperature was 50°C. This enzyme also had a half-life of 33.3 min at 40°C. Kinetics assay showed that AoG6PDH was strictly dependent on NADP+ (K<sub>m</sub> = 6.3 μM, k<sub>cat</sub> = 1000.0 s<sup>-1</sup>, k<sub>cat</sub>/K<sub>m</sub> =158.7 s<sup>-1</sup>·μM<sup>-1</sup>) as cofactor. The K<sub>m</sub> and k<sub>cat</sub>/K<sub>m</sub> values of glucose-6-phosphate were 109.7 s<sup>-1</sup>·μM<sup>-1</sup> and 9.1 s<sup>-1</sup>·μM<sup>-1</sup> respectively. Initial velocity and product inhibition analyses indicated the catalytic reaction followed a two-substrate, steady-state, ordered BiBi mechanism, where NADP<sup>+</sup> was the first substrate bound to the enzyme and NADPH was the second product released from the catalytic complex. The established kinetic model could be applied in further regulation of the pentose phosphate pathway and NADPH regeneration of A. oryzae to improve its xylose utilization and yields of valued metabolites.

        The effects of MCM-41’s calcination temperature on the structure and hydrodenitrogenation over NiW catalysts

        MCM-41 was calcined at 500, 560, 600 or 650 oC. It was used as support for NiW catalysts of hydrodenitro-genation (HDN) for quinoline in order to investigate the influences of the MCM-41’s calcination temperature on thestructure and the HDN performance of NiW catalysts. The NiW catalysts were characterized by XRD, N2 adsorption-desorption, XPS, Raman, HRTEM and Py-IR techniques. The results showed that the surface area (SBET), the averagepore diameter (Dp) and the pore volume (Vp) of the MCM-41 increased with increase of the MCM-41’s calcinationtemperature. The high SBET , Dp and Vp were beneficial for the high dispersion of W species, the formation of appropriatenature of W species and acid sites on the catalysts. The HDN activity followed the order of NiW-650≈NiW-600>NiW-560>NiW-500, while the relative selectivity of HDN pathways was similar for all the catalysts.

        Acupuncture Therapy for the Treatment of Myelosuppression after Chemotherapy: A Literature Review over the Past 10 Years

        The aim of this study is to review current studies on the effect of acupuncture therapy on bone marrow suppression after chemotherapy. The authors of the present paper have searched related literature over the past 10 years at home and abroad, analyzing the features and the effects of acupuncture therapy (including acupuncture, moxibustion, point injection, point application, etc.) for treating myelosuppression after tumor chemotherapy. We also discuss the year of publication, document type, acupuncture therapy, acupoint selection, and adverse effects with figures. We analyzed 159 articles related to acupuncture therapy from 2004 to 2013, and the analysis revealed that point injection was the most frequently used therapy for clinical applications, and that Zusanli (ST36) was the most frequently used acupoint. The results showed that some problems regarding the design method, acupoint selection, and acupuncture intervention measure existed in those research studies. We hope to provide readers with an overall and objective understanding of acupuncture and moxibustion therapy for treating myelosuppression after tumor chemotherapy.

Analyze the model by using the average cycle method. The Computer simulation results verify the correctness of the theory. Has some practical significance.

        Working Conditions on the Afterglow Characteristics of Rare-earth Luminous Fibers

        To test and clarify the stability of afterglow performance of luminescent fibers can accelerate the step of the commercialized application on luminescent textiles. This work investigated the possible effects of a number of working conditions on the afterglow characteristics of luminous fibers. The fibers in our studies did not show any significant change in their afterglow brightness and duration after storage for 12 months under conditions of constant temperature and humidity, after 5 hours of light exposure, or soaking in water for 4 hours. The insignificant decay appears to follow the same mechanism. Thermal perturbation seemed to cause some changes to the initial brightness and decay time with the best performance being observed at about 80 oC. Moreover, contact with acid or base for 5 minutes only resulted in slight reduction of the afterglow brightness. Our studies thus indicate a high degree of stability of the afterglow performance of the luminous fibers used.

        Extraction of microbial transglutaminase from Amycolatopsis sp. fermentation broth using aqueous two-phase system

        Partitioning of microbial transglutaminase (MTG) from Amycolatopsis sp. in the polyethylene glycol (PEG)/salt-based ATPS was investigated for the first time. The key parameters such as the molecular weight of PEG (PEG 600-6000), the type and concentration of phase-forming salt (ammonium sulfate or phosphates), the pH of system (pH 5.0-8.5), and the concentration of neutral salt (0-6% NaCl, w/w) were determined. The partition coefficient of the enzyme was not linear with PEG molecular weight; PEG1000 gave better yield than others. The concentration of PEG1000, ammonium sulfate and NaCl, and the system pH showed effects with different extents on specific activity (SA) and yield of the enzyme. In the ATPS of 26% w/w PEG 1000 and 19% w/w ammonium sulfate in the presence of 5% w/w NaCl and at pH 6.0, MTG was partitioned into the PEG-rich phase with a maximum yield of 86.51% and SA was increased to 0.83. The results of SDS-PAGE showed the MTG produced by the test strain differed from the enzymes reported before. Thus, this study proves that ATPS can be used as a preliminary step for partial purification of MTG from Amycolatopsis sp. fermentation broth.

        Herbal medicine Radix Scutellariae quality improved by exposure of the fresh root to high temperature

        Approximately 1200 years ago, the close association between the quality of medicine and the producing area was well known in China. Recent work has shown conclusively that the quality of herbal medicine is influenced by unfavorable conditions, but the exact cause remains unclear and is a difficult problem in research on herb resources. Fresh roots of S. baicalensis were exposed to temperatures of 35 °C ~ 55 °C, and the H2O2 level remained relatively inflexible. The superoxide dismutase (SOD) and catalase (CAT) activities remained stable at 35 °C and 45 °C but decreased on day 1 at 55 °C. The excess ROS due to the high temperatures must be removed by another pathway. Our study reported that the phenylalanine ammonia-lyase (PAL) activities rose. The fresh roots were exposed to 55 °C for 2 days, the baicalin increased by 38.5%, the wogonoside by 24.1%, and the baicalein by 37.0%. This method can substantially improve the quality of Radix Scutellariae.

        Numerical Simulation and Analysis Procedure for Model-based Digital Driving Dependability in Intelligent Transport System

        Based on numerical simulation and analysis procedure for measurement of digital driving shaping behaviour, this paper presents a methodology for model-based digital driving dependability analysis in intelligent transport system. First of all, the new role of driver in intelligent traffic information network environment has been described in enough detail, and then an algorithm for measuring dependability for both digital driving and normal driving has been developed for understanding digital driving error mechanisms. The emphasis of this paper is placed on modelling of digital driving reliability and simulating of digital driving safety by considering the influence of digital driving behaviour shaping factors. The objective of model-based driving dependability analysis is to provide a new solution for enhancing traffic safety.

        Analysis of the repeatability of a deployable space tri-prism mast based on the Monte Carlo method

        Space deployable mast, which is a supporting mechanism, especially for a space telescope, requires high repeatability. In this study, a deployed space tri-prism mast was taken as the research object, and the change of a tensional cable’s tension force was equivalent to an external load. Based on the accurate stiffness model of the mast, the repeatability of the tri-prism mast was calculated by using the Monte Carlo method. Then, the influences of the number of units, the change in the tension force, the thickness of the linkage, and the radius of the tensional cable on the repeatability of the tri-prism mast were studied. Moreover, the correctness of the analytical model was verified by the repeatability experiments. Results demonstrated that the proposed method could be used to analyze the repeatability of tri-prism mast. This study provided references and suggestions for the design of high-precision space deployable mast.
