A report of 46 unrecorded bacterial species in Korea belonging to the classes Alphaproteobacteria, Betaproteobacteria, Deltaproteobacteria and Epsilonproteobacteria

        Jung, Hye Su,Yoon, Jung-Hoon,Kim, Seung-Bum,Yi, Hana,Cho, Jang-Cheon,Joh, Kiseong,Cha, Chang-Jun,Seong, Chi-Nam,Bae, Jin-Woo,Im, Wan-Taek,Kim, Myung Kyum,Lee, Soon Dong,Jeon, Che Ok The National Institute of Biological Resources 2019 Journal of species research Vol.8 No.2

During a comprehensive investigation of indigenous prokaryotic species in Korea, a total of 46 bacterial strains assigned to the classes Alphaproteobacteria, Betaproteobacteria, Deltaproteobacteria, and Epsilonproteobacteria were isolated from a diversity of habitats including freshwater, seawater, brackish water, ginseng soil, plant roots, natural caves, and tidal flats. Based on their high 16S rRNA gene sequence similarities (>98.7%) and formation of strongly-supported phylogenetic clades with the closest type species, each strain was assigned to an independent, predefined bacterial species. Since there were no published or official reports regarding the isolation of these 46 species in Korea, here we report them as new species to Korea: 34 species in 14 families in the five orders of Alphaproteobacteria, 10 species in five families in the three orders of Betaproteobacteria, one species of Deltaproteobacteria and one species of Epsilonproteobacteria. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are described in the species description section.

Differential Toxicokinetics Determines the Sensitivity of Two Marine Embryonic Fish Exposed to Iranian Heavy Crude Oil

        Jung, Jee-Hyun,Kim, Moonkoo,Yim, Un Hyuk,Ha, Sung Yong,Shim, Won Joon,Chae, Young Sun,Kim, Hana,Incardona, John P.,Linbo, Tiffany L.,Kwon, Jung-Hwan American Chemical Society 2015 Environmental science & technology Vol.49 No.22

<P>Interspecific difference in the developmental toxicity of crude oil to embryonic fish allows the prediction of injury extent to a number of resident fish species in oil spill sites. This study clarifies the comparative developmental effects of Iranian heavy crude oil (IHCO) on the differences of biouptake and toxic sensitivity between embryonic spotted sea bass (<I>Lateolabrax maculates</I>) and olive flounder (<I>Paralichthys olivaceus</I>). From 24 h after exposure to IHCO, several morphological defects were observed in both species of embryonic fish, including pericardial edema, dorsal curvature of the trunk, developmental delay, and reduced finfolds. The severity of defects was greater in flounder compared to that in sea bass. While flounder embryos accumulated higher embryo PAH concentrations than sea bass, the former showed significantly lower levels of CYP1A expression. Although bioconcentration ratios were similar between the two species for some PAHs, phenanthrenes and dibenzothiophenes showed selectively higher bioconcentration ratios in flounder, suggesting that this species has a reduced metabolic capacity for these compounds. While consistent with a conserved cardiotoxic mechanism for petrogenic PAHs across diverse marine and freshwater species, these findings indicate that species-specific differences in toxicokinetics can be an important factor underlying species’ sensitivity to crude oil.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/esthag/2015/esthag.2015.49.issue-22/acs.est.5b03729/production/images/medium/es-2015-03729u_0004.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/es5b03729'>ACS Electronic Supporting Info</A></P>

      • The Methoxyflavonoid Isosakuranetin Suppresses UV-B-Induced Matrix Metalloproteinase-1 Expression and Collagen Degradation Relevant for Skin Photoaging

        Jung, Hana,Lee, Eunjoo H.,Lee, Tae Hoon,Cho, Man-Ho MDPI 2016 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.17 No.9

<P>Solar ultraviolet (UV) radiation is a main extrinsic factor for skin aging. Chronic exposure of the skin to UV radiation causes the induction of matrix metalloproteinases (MMPs), such as MMP-1, and consequently results in alterations of the extracellular matrix (ECM) and skin photoaging. Flavonoids are considered as potent anti-photoaging agents due to their UV-absorbing and antioxidant properties and inhibitory activity against UV-mediated MMP induction. To identify anti-photoaging agents, in the present study we examined the preventative effect of methoxyflavonoids, such as sakuranetin, isosakuranetin, homoeriodictyol, genkwanin, chrysoeriol and syringetin, on UV-B-induced skin photo-damage. Of the examined methoxyflavonoids, pretreatment with isosakuranetin strongly suppressed the UV-B-mediated induction of MMP-1 in human keratinocytes in a concentration-dependent manner. Isosakuranetin inhibited UV-B-induced phosphorylation of mitogen-activated protein kinase (MAPK) signaling components, ERK1/2, JNK1/2 and p38 proteins. This result suggests that the ERK1/2 kinase pathways likely contribute to the inhibitory effects of isosakuranetin on UV-induced MMP-1 production in human keratinocytes. Isosakuranetin also prevented UV-B-induced degradation of type-1 collagen in human dermal fibroblast cells. Taken together, our findings suggest that isosakuranetin has the potential for development as a protective agent for skin photoaging through the inhibition of UV-induced MMP-1 production and collagen degradation.</P>

      • Clinical Impact of CD4+CD25+Foxp3+ Regulatory T Cell in Hepatocellular Carcinoma Patients Undergoing Tran-sarterial Chemoembolization

( Hana Park ),( Jae Hyung Jung ),( Min Kyung Jung ),( Jun Yong Park ),( Do Young Kim ),( Sang Hoon Ahn ),( Simon Weonsang Ro ),( Jeon Han Park ),( Eui-cheol Shin ),( Kwang-hyub Han ) 대한간학회 2018 춘·추계 학술대회 (KASL) Vol.2018 No.1

Aims: Regulatory T cell (Treg) play an essential role in regulation of antitumor immunity. Although several studies regarding Treg in hepatocellular carcinoma (HCC) have been reported, knowledge of Treg after transarterial chemoembolization (TACE) is still unrevealed. The aim of this study was to investigate clinical impact of Treg in HCC patients who treated with TACE. Methods: We measured the frequency of peripheral blood Tregs in 23 healthy controls and 27 HCC patients undergoing TACE between 2010 and 2012 at Severance Hospital. The patients were divided into two groups according to tumor response; complete response group and incomplete response group. Results: The frequency of Treg in HCC patients was significantly higher than in healthy controls (7.52±2.62% vs. 4.99±1.4%, P<0.001). In a Treg subpopulation, the frequency of Treg (II) was significantly higher in HCC patients than in healthy controls (2.51±1.08% in HCC vs. 0.60±0.28% in healthy controls, P<0.001). In comparison of Tregs numbers at baseline and post-TACE by tumor response, the change of Treg (III) in complete response group from before to after TACE was statistically significant (63.8±42.2/mm3→53.2±31.9/mm3, P=0.050). The complete response group contained more patients with low post-TACE Treg (III) numbers, and the incomplete response group more patients with high post-TACE Treg (III) numbers, with statistical significance (70.9% vs. 29.4% in complete response group; 30.0% vs. 70.0% in incomplete response group, P=0.040). Patients with high post-TACE Treg (III) exhibited a significantly shorter time to progression than those with high post-TACE Treg (III) (3.8 months vs. 11.6 months, P=0.038). On multivariate analysis, hypoalbuminemia (HR 3.324; 95% CI (1.098-10.063), P=0.034) and high post-TACE Treg (III) (HR 3.080; 95% CI (1.091-8.696), P=0.034) were independent sig-nificant prognostic factors in terms of progression. Conclusions: High peripheral blood Treg (III) levels at 1 month after TACE were independently prognostic of poor progression-free survival.

Analysis of the Cytotoxicity of Green Pigment Produced on the Surface of Roasted and Retorted Chestnuts Using Immune Cells and Gastrointestinal Cancer Cells

        Hana Jung,Ji-Hyun Jeong,Hee Soon Cheon,Jun-Bong Choi,Hyunnho Cho,Changho Jhin,Keum Taek Hwang 한국식품영양과학회 2011 Preventive Nutrition and Food Science Vol.16 No.3

Roasted and retorted (RR) chestnuts develop green pigment spots on their surface during storage. The purpose of this study was to evaluate the cytotoxicity of the green pigment using RAW 264.7, MOLT-4, KATOⅢ and HT-29 cells. The pigment scraped from RR chestnuts (GP), whole RR chestnuts with green pigment spots (GC), whole RR chestnuts without green pigment (WC) and roasted and frozen stored chestnuts (FC) were extracted in 10% DMSO. MOLT-4 cells were less resistant to the cytotoxicity of the chestnut extracts than the RAW 264.7 cells. The GP extracts did not show different responses against H2O2-induced oxidative stress and LPS-induced NO production compared to the other extracts. The chestnut extracts did not have proliferative activity on either of the KATOⅢ or HT-29 cells (p>0.05). Our results from the comparison of the green pigment produced on the surface of the RR chestnuts to chestnuts that do not develop the green pigment suggest that the pigment may not be harmful in terms of either cytotoxicity towards immune cells or proliferation of gastrointestinal cancer cells.

      • P142 : A comparative split-face study of low-fluence 1064nm Q-switched Nd:YAG alone and combined pulsed-dye laser for the treatment of melasma

( Hana Jung ),( Woong Suk Chae ),( Jun Young Seong ),( Sookhyun Kong ),( Yusung Choi ),( Hoseok Suh ) 대한피부과학회 2014 대한피부과학회 학술발표대회집 Vol.66 No.2

Background: The pathogenesis of melasma has not yet been cleary demonstrated. Recent evidence suggests that vascular abnormalities are involved in the pathogenesis of melasma. Objectives: The objective of this pilot study was to evaluate the dual treatment of 1,064-nm Q-switched Nd:YAG(QSNY) laser and pulsed-dye laser in the treatment of melasma. Methods: Seventeen Korean women were recruited for this study. All patients were treated on the entire face using the QSNY weekly for nine sessions. Three sessions of Pulsed-dye laser were performed at 4-week intervals on the half face that was randomly assigned. Clinical parameters included physician and patient assessment, gross erythema score and Melasma Area and Severity Index score(MASI). Results: The mean MASI score on both sides was decreased at 9 and 16 weeks compared with baseline(p<0.001 for both), while no significant differences between both sides were reported(p=0.164). Difference in physician and patient assessment between both sides were not statistically significant. Gross erythema score in combined-treated side was statistically significantly lower than those for QSNY alone(p<0.001). Conclusion: Although our study results show that both dual treatment and QSNY alone are capable of improving melasma, antiangiogenetic laser for treatment of melasma specially accompanied by pronounced gross erythema in Asian skin is a possible treatment option.

      • Changes in Immunologic Function after Transarterial Chemoembolization in Patients with Hepatocellular Carcinoma

( Hana Park ),( Ji-ye Song ),( Hee Jung An ),( Yun Bin Lee ),( Joo Ho Lee ),( Mi Na Kim ),( Young Eun Chon ),( Yeon Jung Ha ),( Seong Gyu Hwang ),( Kyu Sung Rim ) 대한간학회 2016 춘·추계 학술대회 (KASL) Vol.2016 No.1

Aims: Transarterial chemoembolization (TACE) is widely used as a treatment modality for intermediate stage of hepatocellular carcinoma (HCC). Although several studies showed enhanced natural killer (NK) cell response after local treatment for HCC have been reported, knowledge of impact on NK cell after TACE is still unrevealed. The aim of this research was to investigate immunologic changes after TACE in HCC patients. Methods: Total 23 patients undergoing TACE for hepatocellular carcinoma were enrolled. Absolute counts of peripheral blood lymphocytes followed by phenotypic and functional characterization of NK cell population were carried out the day before, 1 and 4 weeks after TACE. Results: Peripheral blood lymphocytes kinetics revealed decrease of NK cell population at 1 week after TACE from the baseline (4.18% at 1 week vs. 9.54% at baseline, P=0.047), and restoration of NK cell population at 4 week after TACE from 1 week after TACE (12.24% at 4 week vs. 4.18% at baseline, P=0.040). This was along with significantly increased level of inhibitory NK receptor, NKG2A, at both time points of 1 week and 4 week after TACE from the baseline (20.24% at 1 week from 4.26% at baseline, P<0.001; 23.92% at 4 week from 4.26% at baseline, P<0.001), while there were no significant changes in activating NK receptors. Anti-K562 cell cytotoxicity appeared consistently decreased in terms of absolute activity at 1 week after TACE as compared to baseline, and showed a tendency to restore at 4 weeks after TACE. Conclusions: Unlike previously reported immunologic changes in patients with local treatment for HCC, immunologic function seems to be compromised shortly after TACE. This result suggests that various therapeutic strategies can have different effects on body immune function, and better understanding of the changes in the immune system in HCC patients will promise better tumor control.
