Mutagenesis of Streptomyces kasugaensis for Kasugamycin Production

This study was performed to develop mutant strain using a combination of UV irradiation procedures with protoplast mutagenesis in order to achieve an effective kasugamycin production from Streptomyceskasugaensis. Whenlessthan 1.0g/lof Linoleic acid was used, the cell growth was not inhibited. On the other hand, the cell growth was greatly inhibited when more than 1.6 g/l of linoleic acid was used. Among the various mutant strains, SK-12 was obtained in medium containing 1.6g/l of linoleic acid, showing the highest rate of both cell growth and kasugamycin production. In order to compare kasugamycin production with the SK-12 and the parent strain using soybean oil, cultures were performed in a flask. The production of kasugamycin was increased with the increase time. The maximum kasugamycin concentration was 1.2g/l after 6 days of culture. The product yield from soybean oil was 0.05g/l/g consumed carbon source, which was roughly 5.0 fold higher than the parent strain. These results show that it was effective method for obtaining a mutant resistant to linoleic acid for the effective production of kasugamycin from soybean oil.

A novel series of benzylidenethiazolidine-2,4dione derivatives was synthesized and investigated for 15hydroxyprostaglandin dehydrogenase (15-PGDH)-scavenging activity, PGE2 release, and wound-healing activity. Among the tested derivatives, seven compounds (3, 9, 11, 12, 13, 14, and 25) resulted in a 50% inhibition of 15-PGDH at concentrations between 0.07 and 0.2 μM and increased PGE2 levels from 300 to over 600% in A549 cells treated with 5.0 and 10.0 µM of the compounds for 12 h. A scratch wound-healing assay using HaCaT cell line was conducted to verify the effects of 10 µM of these compounds on cell regeneration. The closure rate of the scratch wound healing showed that all compounds (3, 9 , 11, 12, 13, 14, and 25) had greater wound regeneration effects than the cell growth factor, TGF-β1, which was used as the positive control. In particular, (Z)-N-benzyl-4-((2,4-dioxothiazolidin5-ylidene)methyl)benzamide (compound 14) showed that the highest wound closure rate, which was 360%; this is about 3.6-fold higher than that of TGF-β1. Overall, these results show that compound 14 may be considered a promising candidate for the development of novel woundhealing agents.

The object of this research is to study effect of the waste vegetable oil on mycelium and exopolysaccharide production from Pleurotus sajor-caju PSC 14. Among various culture temperatures, the mycelium and exopolysaccharide production were highest at 29°C. However, they decreased over and below 29°C. When waste vegetable oil 2.0g/L was used for 5 days, the maximum mycelium production was obtained. On the other hand, in the case of exopolysaccharide production, it was obtained at waste vegetable oil of 1.0g/L. Among various surfactants, Tween 80 was the best one for effect on the mycelium and exopolysaccharide production from submerged liquid fermentation of Pleurotus sajor-caju PSC 14. Fed batch fermentations were carried out in 20L airlift bioreactor containing 10L working volume. When Tween 80 was used, the maximum mycelium and exopolysaccharide concentration were respectively 5.62 g/L and 0.67g/L after 8 days of culture. The product yield from oil was 0.12g/g consumed carbon source. These results show that Tween 80 can be useful for the mycelium and exopolysaccharide production from submerged liquid fermentation of Pleurotus sajor-caju PSC 14 using waste vegetable oil.

Of all the many types of foreign bodies which may enter the upper gastrointestinal tract, some may cause serious complication while they are spontaneously passed. But objects longer than 10.0 cm and thicker than 2.0 cm tend to lodge in the stomach, duodenojejunal flexure, ileocecal valve and anus. We experienced a case of spontaneously passed long esophageal stent (13.0 * 2.5 cm in size), which was inserted because of lower esophageal stricture by extension from cardiac cancer of the stomach, after chemotherapy. So, we report this case with the review of the literature.

This study was to suggest the method for wrinkle improvement using Mugil cephalus extract. In order to evaluate the cosmetic product made of M. cephalus extract, the experiments were conducted with subjects and skin states for 4 weeks. Effect of cosmetics containing M. cephalus extract on the water holding capacity and evaporation of the women skin were investigated. The water holding capacity was increased after 1 h of application. But after 2 h, it was decreased. In the case of the water evaporation, it was increased with the increase of time. On the clinical trial, it was also found that water, oil, and rough level on the women skin were increased and wrinkles were improved. Side effects were also not detected during the application and treatment of cosmetics. Based on this result, the application of M. cephalus extract had positive effects on the improvement of wrinkles and skin in women. Therefore, it will be used to develop the wrinkle improvement therapy for women.
Precision of the milled full-arch framework fabricated using presintered soft alloy: A pilot study

PURPOSE. This study aimed to evaluate the marginal discrepancy of full-arch frameworks in implant-supported prostheses fabricated using pre-sintered soft alloy (PSA). MATERIALS AND METHODS. Full-arch metal frameworks were fabricated on the edentulous implant model using casting alloy (CA), fully-sintered hard alloy (FHA), and PSA (n = 4 in each group). To evaluate the misfit of the framework to the abutments, the absolute marginal discrepancy (AMD) values of the frameworks were measured in cross-sectional images that had been drawn as part of the triple-scan protocol. The AMD values were compared among the tested alloy groups using the Kruskal-Wallis test, with a post hoc Mann-Whitney U test (α=.05). RESULTS. The FHA and PSA groups showed lower marginal discrepancies than the CA group (P<.001). However, the FHA group did not differ significantly from the PSA group. CONCLUSION. Soft alloy milling is comparable to hard alloy milling, and it is more precise than casting in terms of the marginal fit of implant-supported, full-arch prostheses.
