        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         혈관질환 정보관리 시스템

        As medical technology progresses rapidly, there is a rise in the average age along with the Korean dietary lifestyle becoming more westernized, which leads to an increase in the number of vascular disease patients in Korea. Thus, we need to manage the medical information of a disease systematically in order to diagnose and treat constructively. However, since there has been no standardized method of man agement to date, a great deal of information could not be properly utilized nor studied. Therefore, the departments of Cardiology, Radiology and Neurology of Samsung Seoul Hospital recently got together to develop an information management system called the Vascular Data System. This program was developed to be run on win98 O/S, upper Pentium Ⅲ, and upper 128 MB Memory, and its source code is Dephi 4.0. It was configured for the user to set the configurations as well as do a variety of search and analysis. If this program were to be updated continuously, it may be used extensively as well as in various parts of clinical research activities.

      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

      • 만성 견비통에 대한 동씨침 치료의 무작위 대조군의 임상 연구

        Objectives : To observe the effect of acupuncture treatment in chronic shoulder pain patients. Methods : 36 voluntary patients were randomly assigned to an acupuncture treatment group(E GrouP, n=18) and a control group(C GrouP, n=18). The E Group patients received acupuncture treatment on LI_(15), TE_(14), GB_(21) and Master Dong's acupuncture points, Shin-gwan and Gyun-joong, twice a week for four weeks. The C Group patients received no treatment. All patients in both groups were instructed to practise self exercise in their daily lives. Evaluations were made at baseline and after four weeks of study. The Constant Shoulder Assessment(CSA), Shoulder Pain and Disability Index(SPADI) and the patient's subjective pain was measured by Visual Analogue Scale(VAS). The obtained data was analyzed. Results '. The E Group showed significant(p<0.05) improvement in CSA, SPADI and VAS after four weeks of treatment. The C Group showed significant(p<0.05) improvement in CSA, but the change of SPADI and VAS was insignificant(P>0.05). CSA and SPADI of E Group significantly(p<0.05) improved compared to the C GrouP, but the difference of VAS change in the two groups was insignificant(p>0.05). Conclusions : Four weeks of acupuncture treatment significantly improved CSA, SPADI and VAS. The improvement of CSA and SPADI was significant(p<0.05) compared to untreated patients.

        2급 와동 수복 시 한국 치과 의사들의 복합레진 사용 실태 연구

        본 연구는 2008년 1월 현재 한국 치과의사들의 복합레진 사용 실태에 관한 설문 조사로서, 2급 와동을 수복하는 증례에서 치과의사의 면허 년도, 교육 배경 및 진료환경 등에 따라 수복재료의 선택, 복합레진의 사용여부 및 사용방법, 문제점 등을 비교하였다. 한국 치과의사들을 대상으로 2급 와동 수복에 관한 17 문항의 설문지를 제작하였다. 이 설문지는 시술자 정보 2급 와동의 수복으로 크게 2부분으로 나누어진다. 이 설문지는 대한치과의사협회를 통해 12.193 명의 치과의사들에게 E-메일로 발송되었다. 이 중 2.612개의 메일이 수신 확인되었고 840 개의 설문지가 작성되어 회신되었다. 수신 확인된 메일 수에 대한 회신된 메일 수의 비율 (회신율)은 32.2%이었고, 이 자료는 SPSS프로그램에서 카이제곱 분석을 이용하여 교차 분석하였다. 답변자의 비율은 면허년도를 기준으로 1998 ~ 2007년에 면허를 취득한 그룹 (한국 전체 치과의사의 33 3%, 추정 년령 26-35세)이 60.3%로 1997년 이전에 면허 취득한 그룹 (한국 전체 치과의사의 66.7%.추정 년령 36세 이상)의 39.7%보다 높은 것으로 나타났다. 또한 이들이 근무하는 병원은 개인의원 (77%)이 가장 많았고 남자 치과의사(79%)가 많았다. 복합레진 수복에 대한 지식은 학생 때 수업이나 학회, 세미나를 통하여 (83.4%) 얻은 것으로 나타났다. 2급 와동 수복 시 재료 선호도를 살펴보면 금인레이가 65.7%를 차지하고 있고 복합레진 직접 수복은 12.1%로 낮게 나타났다. 2급 와동의 복합레진 직접 수복 시 시술 방법에 있어서는 러버댐을 사용하지 않거나 잘 사용하지 않는 그룹의 비율(74.4%)이 더 많았고. 격벽법으로는 mylar strip (53.4%)이나 metal matrix (33.8%), Palodent svstem (6.5%)를 사용하였다. 충전은 적층법 (99.6%)을 사용하는 것으로 나타났으며, 시술 시 인접면 형성을 가장 어려워하였다.(57.2%). 2-step 접착시스템 (76%)이 3-step 접착시스템 (16%)보다 많이 사용되고 있었다. 사용하는 레진 제품으로는 Z250(20%)이 가장 많이 사용되고 있었다. The purpose of this study was to assess the current materials, methods and difficulties according to the year of licence and educational background of Korean dentists in Class Ⅱ direct composite resin restorations. Total 17 questions were included in the questionnaire. Questions were broadly divided into two parts first, operator's information and second the materials and methods used in Class Ⅱ posterior composite restoration. The questionnaire was sent to dentists enrolled in Korean Dental Association via e-mail. Total 12,193 e-mails were distributed to dentists. 2,612 e-mails were opened and 840 mails (32.2%) were received from respondents. The data was statically analyzed by chi-square test using SPSS(v 120.1 SPSS Inc. Chicago. IL USA). Male dentists among respondents was 79% 60.3% of the respondents acquired their licences recently (1998-2007) and 77% practiced in private offices 83.4% have acquired their knowledge through school lectures conferences and seminars. For the Class Ⅱ restorations gold inlays were preferred by 65.7% of respondents while direct composite resin restorations were used by 12.1% amalgam users were only 4.4% of respondents. For the restorative technique 74.4% of respondents didn't use rubber dam as needed. For the matrix. mylar strip (53.4%), metal matrix (33.8%) and Palodent system (6.5%) were used 99.6% of respondents restored the Class Ⅱ cavity by incremental layering. Obtaining of the tight interproximal contact was considered as the most difficult procedure (57.2%) followed by field isolation (21%). Among various bonding systems, 22. 6% of respondents preferred SE Bond and 20.2% used Single Bond. Z-250 was used most frequently among a variety of composite resins.

      • LCA 기법을 적용할 제품의 우선순위 선정 방법론

        본 연구의 목적은 일정한 논리적 절차를 거쳐서 객관적인 전과정평가의 우선순위를 선정하기 위한 일반적인 방법론의 개발이다. 우선 전과정평가 대상이 되는 제품(산업군)의 우선순위를 평가하기 위해서 제품의 환경성과 관련된 환경규제, 국내외 전과정평가 수행사례, 유해물질 등을 평가항목으로 설정하였다. 설정된 평가항목들에 대한 자료조사 및 분석을 통하여 세분화한 데이터베이스를 생성하였으며, 또한 학계 및 산업계의 의견을 반영하기 위해 전과정평가 전문가 및 산업계 관계자들을 대상으로 설문조사를 실시하였다. 설문조사결과를 바탕으로 각각의 평가항목 및 세부평가항목의 가중치를 부여하였고, 각 평가항목별로 점수를 설정하였다. 본 연구에서 개발한 방법론은 우선순위를 비교해야하는 제품(산업군)들이 각각의 평가항목에서 설정해놓은 해당 점수를 부여받게 된다. 부여받은 각 평가항목의 점수들을 모두 합산하면 평가대상이 되는 제품(산업군)에 대해 하나의 우선순위 평가 점수를 갖게 된다. 이와 같은 과정으로 방법론이 구축되었으며, 사례연구를 통하여 방법론의 타당성을 검증하였다. 이렇게 산출된 우선순위 평가점수를 평가하고자하는 제품(산업군)들과 비교하여 높은 점수를 갖는 제품(산업군)이 전과정평가를 먼저 실시하는 것이 바람직하다는 결론을 얻을 수 있다. The purpose of this study is to develop a general method to decide the priority of LCA application through a logical procedure. First of all, evaluation categories were developed to decide priority of subject products(industrial groups). The evaluation categories consist of environment regulations related to subject product, LCA case studies, hazardous substances, etc. The subdivided database was constructed through researching and analyzing information about the selected evaluation categories. In addition, a questionnaire was conducted to reflect the opinions from academic and industrial circles. Based on the result of the questionnaire, each evaluation category and sub-evaluation category were weighed differently, and certain points were assigned to them. According to the method developed in this study, comparing industrial groups or products for prioriting will have its total score that is a sum of points assigned to each evaluation category. This results that an industrial group or product having higher score than the others becomes the subject to apply LCA.

        For many years and primarily for economical reasons, Dissimilar Metal Welds have been used as transition joints in a variety of equipment and applications. But Dissimilar Metal Welds have several fabrication and metallurgical drawbacks that can often lead to in-service failures. In this paper, the laser weldability of STS304 stainless steel and SM45C at dissimilar metal welds using a continuous wave Nd:YAG laser are experimentally investigated. An experimental study was conducted to determine effects of welding parameters, on eliminating or reducing the extent welding zone formation at dissimilar metal welds and to optimize those parameters that have the most influence parameters such as focus length, power, beam speed, shielding gas, and wave length of laser were tested.

      • 질량분석법에 의한 항암제 BLM 금속착물의 정량제목

        BLM혼합물인 Bleocin으로부터 BLM-A2를 고순도로 대량 분리, 정제할 수 있는HPLC조건을 확립하였다. 질량분석법을 이용한 다양한 금속의 BLM 착물의 합성과정을 추적하는 연구기법을 개발할 수 있었고 BLM의 금속착물 형성에 의한 새로운 BLM 정량법을 개발하였다. 이러한 방법을 이용하여 CoBLM-A2(성분 1, 2, 3), FeBLM-A2, CuBLM-A2, PtBLM-A2 (성분 1, 2),ZnBLM-A2, CoBLM-32(성분 1, 2)등의 착물을 합성하고 질량분석법을 이용한 다양한 착물합성에 정도를 확인할 수 있었다. The highly purified and large scaled BLM-A2 was obtained from the anticancer drug(Bleocin) containing BLM mixture by establishing HPLC condition. The complex formations with various metaI ions were monitored by Mass spectrometry. CoBLM-A2(component 1,2,3), FeBLM-A2, CuBLM-A2, PtBLM-A2(component 1,2), CoBLM-B2 [component 1,2] were synthesized and quantitatively characterized by Mass spectrometry.
