        Modal parameter estimation of civil structures based on improved variational mode decomposition

        This paper proposes an improved variational mode decomposition (IVMD) algorithm for structural modal parameter estimation based on non-stationary responses. In this improved VMD, the mean envelope entropy (MEE) and particle swarm optimization (PSO) are first employed to determine the optimal decomposition parameters for the subsequent VMD analysis. Then the VMD algorithm is used to decompose the non-stationary data into a number of intrinsic mode functions (IMFs). After obtaining the IMFs based on the IVMD, structural modal parameters such as natural frequencies and damping ratios of civil structures can be determined by using Natural Excitation Technique (NExT) and Direct Interpolating approach (DI). The feasibility and accuracy of the proposed procedure are evaluated by both numerical and full-scale examples. The natural frequencies and damping ratios are successfully identified from the vibration responses with high noise and nonstationary characteristics. The results of this study illustrate that the proposed procedure provides a powerful approach to identify the modal parameters of civil structures using non-stationary responses.

        Rapid Detection of Clostridium tetani by Recombinase Polymerase Amplification Using an Exo Probe

        Tetanus is a potentially fatal public health illness resulted from the neurotoxins generated by Clostridium tetani. C. tetani is not easily culturable and culturing the relevant bacteria from infected wounds has rarely been useful in diagnosis; PCR-based assays can only be conducted at highly sophisticated laboratories. Therefore, a real-time recombinase polymerase amplification assay (Exo-RPA) was constructed to identify the fragments of the neurotoxin gene of C. tetani. Primers and the exo probe targeting the conserved region were designed, and the resulting amplicons could be detected in less than 20 min, with a detection limit of 20 copies/reaction. The RPA assay displayed good selectivity, and there were no cross-reactions with other infectious bacteria common in penetrating wounds. Tests of target-spiked serum and pus extract revealed that RPA is robust to interfering factors and has great potential for further development for biological sample analysis. This method has been confirmed to be reliable for discriminating between toxic and nontoxic C. tetani strains. The RPA assay dramatically improves the diagnostic efficacy with simplified device architecture and is a promising alternative to real-time PCR for tetanus detection.

        Enhancement of transglutaminase production in Streptomyces mobaraensis DSM 40587 by non-nutritional stress conditions: Effects of heat shock, alcohols, and salt treatments

        Stress-mediated bioprocess is a strategy designed to enhance biological target productivity. In this study,an attempt was made to enhance transglutaminase (TGase) production by Streptomyces mobaraensis by using different stress conditions including heat shock, alcohols and salt stress. Results showed that the effects of stress on TGase production depended on the type applied. For heat shock, TGase production (1.32 U/ml) was recorded maximum in the culture treated at 48 h post inoculation in water bath at 60 oC for 1 min. For alcohols treatment, the maximum activity of TGase (1.77 and 1.75 U/ml) was obtained when 3% methanol was added into the medium at 0 or 24 h of fermentation. However, a 3.5-fold increased production of TGase (3.8 U/ml) was observed in the medium supplemented with 0.2mol/l MgCl2 compared with the basic medium at the beginning of fermentation. In conclusion, TGase production from S. mobaraensis was improved by heat shock, methanol and salt stress treatments, MgCl2 stress was the most effective.

        A New Triterpene and an Antiarrhythmic Liriodendrin from Pittosporum brevicalyx

        A new triterpene, 21-O-senecioyl-R1-barrigenol (1) and 13 known compounds were isolated from the ethanol extracts of the leaves and bark of Pittosporum brevicalyx (Oliv.) Gagnep. Their structures were elucidated based on spectral data. The antiarrhythmic action of one furofuran lignan, liriodendrin (2), was tested on a model of CaCl2-induced arrhythmia and compared with the effect of verapamil. The prophylactic administration of liriodendrin (2) was effective in prolonging latency of arrhythmia and reducing the occurrence of ventricular fibrillation from 75% to 25%. The overall mortality rate was significantly reduced by the prophylactic administration of liriodendrin from 87.5% to 25%. The antiarrhythmic effect of liriodendrin (5.0 mg/kg) was similar to that of verapamil (1.05 mg/kg). Thus, liriodendrin may be a potent suppressor of CaCl2-induced arrhythmias.
