      • Bacillus subtilis 接種이 土壤微生物相에 미치는 影響(Ⅰ) : 볏짚 및 크로버 添加區에서의 微生物相의 變動 Changes in Microflora by Application of Straw and Clover

        토양으로부터 분리한 B. subtilis를 有機物이 施用된 토양에 還元 接種하여 有機物 種類에 따른 B. subtilis의 토양중에서 適應·定着性 및 B.subtilis가 다른 土壤微生物相에 미치는 영향을 經時的으로 조사하였다. 또한 有機物이 시용된 토양중에서 B,subtilis와 植物 病原菌인 F. oxysporum을 동시에 接種하여 상호간의 拮抗的 關係를 검토한 결과는 다음과 같다. 1) B. subtilis菌數는 對照區에서보다 B. subtilis 接種區에서 增加를 보였고 단지 볏짚 및 크로버만을 사용한 區에서보다 볏짚 및 크로버 施用區에 B. subtilis를 접종한 區가 약 2배 정도 B. subtilis菌數의 증가를 보였다. 특히 B.subtilis를 有機物 施用區에 접종했을 때 볏짚 시용구가 크로버 시용구에서보다 B. subtilis菌數가 약 3배 정도 현저하게 증가하였다. 2) 細菌과 絲狀菌數는 대조구보다 B.subtilis 接種區에서 각각 23% 및 20%씩 감소하였으므로 또한 크로버 시용구에서보다 크로버+B. subtilis 접종구에서도 각각 33% 및 16%씩 감소하였고, 볏짚 시용구에서는 볏짚+B. subtilis 접종구에서 각각 29% 및 32%씩 감소경향을 나타내었다. 3) 胞子形成菌數는 대조구보다 B. subtilis 접종구에서 약 10배 정도 증가하였으며 단지 有機物만을 시용한 區보다도 크로버+B. subtilis 접종구와 +B. subtilis 접종구에서 각각 2배, 4배 정도의 증가경향을 보였다. 또한 gram(-)細菌도 胞子形成菌數와 비슷한 경향을 나타내었고, B. subtilis接種이 그람 음성 세균수의 변동에는 영향을 미치지 않았다. 4) F. oxysporum의 菌數는 크로버 시용구에서보다 크로버+F. oxysporum 접종구에서 25배 정도 증가하였으나 크로버+B. subtilis 접종구에서는 크로버+F. oxysporum 접종구보다 약 5배 정도의 감소경향을 나타내었다. 또한 F. oxysporum菌數는 볏짚+F. oxysporum區에서 볏짚 單獨施用區보다 3배 정도 증가하였으나 볏짚+B. subtilis+F. oxysporum접종구는 볏짚+F. oxysporum접종구보다 F. oxysporum의 數가 약 17배 정도 감소경향을 나타내었다. 더욱이 F. oxysporum菌數는 크로버+B. subtilis+F. oxysporum접종구보다 볏짚+B. subtilis+F. oxysporum접종구에서 약 40배 정도의 감소경향을 나타내었다. 5) 크로버 시용구의 토양 pH는 약 ph 9에 이르러 토양 ph가 alkali화되었으며 볏짚시용구의 토양 ph는 큰 변화 없이 약산성을 유지하였다. This study has been conducted to investigate the adaptation of Bacillus subtilis to the soil and the antagonistic interaction between B. subtilis and pathogen Fusarium oxysporum as well as the periodic change of soil microflora when B. subtilis was inoculated to the soil treated with organic matters. Some results obtained from this study are as follows : 1) The number of B. subtilis was more increased about 6 times in the plot inoculated with B.subtilis than in control plot. It was relatively double in the organic matter+ B. subtilis plot than in the only organic matter plot. B. subtilis was particuraly increased about 3 times in the straw +B. subtilis plot, compared with the clover +B. subtilis plot. 2) The number of total bacteria and fungi was decreased by 23% and 20% in the plot inoculated with B.subtilis respectively than in control plot. It was not only decreased by 33% and 16% in the clover+B. subtilis plot respectively than in the only clover plot but it was also decreased by 29% and 32% in the straw+B. subtilis plot respectively than in the only straw plot. 3) The number of spore-forming bacteria was increased about 10 times in the plot inoculated with B.subtilis than in the control plot. It was also increased 2 times and 4 times in the clover+B.subtilis plot and in the straw+B. subtilis plot respectively than in each organic matter plot. The number of gram-negative bacteria and the spore-forming bacteria were slightly increased in all the plot inoculated with B. subtilis. 4) The number of F. oxysporum was increased about 25 times in the clover+ F. oxysporum plot than in the only clover plot. But it was decreased about 5 times in the clover+B.subtilis+ F. oxysporum plot than in the clover +F. oxysporum plot. Furthermore, F. oxysporum was remarkably decreased about 40times in the straw+B. subtilis+ F. oxysporum plot than in the clover+b. subtilis+ F. oxysporum plot. It occured that B. subtilis had inhibited the growth of pathogen F. oxysporum when B. subtilis was inoculation to the soil treated with straw. 5) Soil pH was changed to pH 9 in the plot treated with clover to become the alkalification of experimental soil. Meanwhile it was not changed in the plot treated with straw.

        구면과 비구면 RGP 렌즈 장기 착용시 각막 상피, 내피에 미치는 영향-주사전자현미경적 관찰

        To investigate the comparative effect of spherical and aspherical RGP lens were worn during 3 weeks on rabbit's cornea. Four white rabbits were worn right eyes with spherical lens and 4 white rabbits were worn right eyes with aspherical RGP lens. Left eyes were served as control. The rabbits were sacrificed at 3 weeks after fitting and observed morphological changes by scanning electron microscopy and also investigate proliferation rate of the corneal epithelium with RGP wearing. After spherical RGP lens wearing, the epithet layer damaged compared to aspherical lens. The superficial cell layer strip off seriously, cell size significantly changed abnormal. Both spherical and aspherical RGP lens fitting group showed so many bacteria and back surface of lens was found like a fern shape. The aspherical RGP lens original material type was some formal than spherical lens. We thought that these pattern was significantly altered with spherical lens by prohibited transmitter oxygen from atmosphere therefore the epithelium shape was changed. This suggested wearing the aspherical lens might be less physiologic than shperical lens fitting. 구면과 비구면 RGP렌즈 장용시 효과를 비교하기 위하여 3주간 실험토끼 눈의 각막에 콘택트렌즈를 착용 시켜 조사하였다. 8마리의 흰 실험토끼 눈 중 4마리의 우안은 구면 RGP렌즈, 4마리의 우안은 비구면 RGP렌즈를 착용 하였다. 왼눈은 대조군으로 사용하였다. 콘택트렌즈착용 3주 후 안구를 적출하여 형태변화를 주사전자현미경으로 관찰하였으며 RGP렌즈를 착용시 각막 상피 세포증식률을 조사하였다. 구면 RGP렌즈를 착용 후, 상피층은 비구면에 비하여 손상을 받았고 상피층은 심하게 벗겨져 나갔으며 세포 크기는 비정상으로 변하였다. 구면과 비구면 렌즈 두 군 모두 많은 박테리아가 보였으며 렌즈의 후면은 양치류 모양의 형태를 발견하였다. 비구면 렌즈의 원재료는 구면렌즈보다는 다소 정형적으로 보였다. 이러한 형태는 대기로부터 산소 전달을 방해함으로 인하여 구면렌즈를 착용한 군에서 각막을 변화시킨 것으로 생각된다. 이 논문은 구면 렌즈보다는 비구면 렌즈가 생리적으로 각막 세포에 덜 영향을 미쳐 더 적합한 것을 제시한다.

      • 각망어구의 구조개량을 위한 기초적 연구

        In order to increase fishing efficiencies of the fyke net used in the coast of Cheju Island water tank experiment was caried out entering and escaping behavior using nets that were reduced to 1/20 of the size of fyke net and were improved to have antrance structure, and Mackerel Scomber japonicus as experimental fish. The results of measurement are as follows: 1. Fish school behavior in the main net was showed two different patterns: swimming in a circle in the right space of the main net and swimming back and forth in ellipse in the right and left space. 2. The swimming speed of mackerel school was 24.9. 12.6 and 32.0cm/sec in the center space. right space of main net and in the mouth. 3. The entering rate of fish school was 40% in net with 35cm length of the upper and funnel net in the mouth of fyke model net and 49% in conventional type fyke model net. 4. The escaping rate of fish school was 10% in net with 35cm length of the upper and funnel net in the mouth of fyke model net and 69% in conventional type fyke model net. 5. The remain rate of fish school was 90% in net with 35cm length of the upper and funnel net in the mouth of fyke model net and 31% in conventional type fyke model net.

      • 돈사폐수를 이용한 Polyhydroxyalkanoates의 생산

        돈사페수를 발효기질로 이용한 생분해성 고분자, polyhydroxyalkanoates(PHA)의 생산이 가능하였다. 균주로는 질소 혹은 인 등의 비탄소 영양원의 제한 없이도 PHA의 생산이 가능한 Azotobacter vinelandii UWD를 이용하였고 돈사폐수배지에서의 PHA 생산특성을 glucose 합성배지에서의 특성과 비교하였다. A. vinelandii uwd균주는 탄소원으로 glucose와 alkanoates가 함유된 배지에서 alkanoates를 우선적으로 소비하여 주로 생장을 하고, glucose 소비단계에서는 생장보다는 PHA를 생산하는 diauxic growth를 하였다. 돈사폐수 원액을 배지로 사용할 경우 UWD 균주의 건조균체량과 PHA함량이 낮았는데(0.91 g/L,37 wt%). 이는 돈사폐수 중에 함유되어 있는 acetate, propionate, butyrate 및 valerate 등과 같은 높은 농도의 alkanoates가 세포생장을 저해하기 때문이었다. 증류수로 50% 희석한 배지(총 생산 균체 건중량 2.02g/L)를 사용하는 것이 바람직한 것으로 밝혀졌다. 50% 희석 돈사폐수 배지에 glucose를 3% 첨가하면 건조균체량은 9.40g/L로 현저하게 증가하였으며, 균체내 PHA 함량은 58.3%, PHA 생산속도는 0.11g/(L·h)이었다. Glucose 합성배지에서 합성된 PHA는 poly-β-hydroxybutyrate(PHB)의 형태이었고, 돈사폐수 배지로부터는 copolymer인 poly-β-hydroxybutyrate-co-β-hydroxyvalerate, P(HB-co-HV)를 2-8mol% 합성할 수 있었다. 돈사폐수배지를 이용하여 PHA를 생산하는 동안 배지의 염도를 대략 50% 정도 제거가 가능하였다. Biodegradable polymer, polyhydroxyalkanoates(PHA), could be produced using swine wastewater (SW)as a fermentation substance. Azotobacter vinelandii UWD was able to produce PHA without limiting non-carbon nutrients such as nitrogen and phosphorous. Production characteristics of strain UWD in SW medium were compared with those in glucose sythetic medium In the synthetic medium containing glucose and alkanoates as carbon sources. A vinelandii UWD grew diauxically, first by using alkanoates and then by using glucose to produce biomass and PHA. Using raw SW medium, cell mass production was small(0.91 g/L)and PHA content was 37 wt% because cell growth was inhibited by alkanoates such as acetate, propionate, butyrate, and valerate contained in SW. Use of two-fold diluted SW as fermentation medium was desirable for PHA production. When 3% of glucose was added to the two-fold diluted SW, dry cell mass increased to 9.40 g/L, and the PHA content and PHA production rate were 58.3 wt% and 0.11g/(L·h), respectively. When grown in SW media A. vinelandii UWD formed PHA copolymers containing β-hydroxybutyrate and β-hydroxyvalerate(HV), and HV content in the copolymer was 2-8mol% whereas homopolymer of poly-β-hydroxybutyrate was produced in the glucose synthetic medium. Approximately 50% of salinity in the medium was reduced after PHA production using SW medium.

        Numerical simulation is performed by the commercial code FLUENT for individual air condition system to calculate the velocity and temperature profiles around worker in climate room. The main calculation region is the space of ㄱ shape between wall and worker in the individual air condition system. The fresh air is emitted from 3 inlet attached on the right, left and center wall. Because the comfort of worker is closely related with the velocity and temperature of wind, these data are key role for design of the individual air condition system. This work is previous study for evaluation of thermal comfort of workers in room without much experiments according to metabolism and heat emission of human body. Numerical simulation results are as followings, 1. There is an possibility of the thermal comfort of worker is gotten worse due to the presence of dramatic change of wind velocity around worker. 2. Temperature gap exists in worker's body between top and bottom according to fresh air inlet velocity and position which is right, left and center of wall. 3. Calculation results of the skin temperature of worker are key data for evaluation of thermal comfort with experimental data in climate room. 4. Optimum condition of temperature, direction and velocity of fresh air will be obtain by numerical simulation for energy effective individual air condition system.

        Sensitivity analysis of satellite-retrieved SST using IR data from COMS/MI

        Sea Surface Temperature (SST) is the temperature close to the ocean's surface and affects the Earth's atmosphere as an important parameter for the climate circulation and change. The SST from satellite still has biases from the error in specifying retrieval coefficients from either forward modeling or instrumental biases. So in this paper, we performed sensitivity analysis using input parameter of the SST to notice that the SST is most affected among the input parameter. We used Infrared (IR) data from the Communication, Ocean, and Meteorological Satellite (COMS)/Meteorological Imager (MI) from April 2011 to March 2012. We also used the Global Space-based Inter-Calibration System (GSICS) correction to quality of the IR data from COMS. SST was calculated by substituting the input parameters; IR data with or without the GSICS correction. The results of this sensitivity analysis, the SST was sensitive from -0.0403 to 0.2743 K when the IR data were changed by the GSICS corrections.

        Value of sonohysterography in preoperative assessment of myometrial invasion for patients with endometrial cancer.

        <P>OBJECTIVE: The aims of this study were to compare the diagnostic performance of sonohysterography (SH) with that of magnetic resonance imaging (MRI) in estimation of myometrial invasion and to evaluate the influence of SH on peritoneal cytologic results for patients with endometrial cancer. METHODS: Seventy-four patients with endometrial cancer were included. Sonohysterography and MRI were performed before surgery. All patients had complete staging procedures, including peritoneal cytologic analyses. The sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) were determined for SH and MRI. RESULTS: The concordance rates of myometrial invasion for SH and MRI were 82.4% and 81.1%, respectively. The sensitivity, specificity, PPV, and NPV for identification of deep myometrial invasion were 64.7%, 87.7%, 61.1%, and 89.3% on SH and 70.6%, 84.2%, 57.1%, and 90.6% on MRI. Two patients (2.7%) were found to have positive results for malignant cells on peritoneal cytologic analyses. CONCLUSIONS: Sonohysterography appears to be a useful preoperative method for predicting myometrial invasion, comparable to MRI.</P>
