Enhanced Lovastatin Production by Solid State Fermentation of Monascus ruber

The purpose of this study was to optimize the solid state cultivation of Monascus ruber on sterile rice. A single-level-multiple-factor and a single-factor-multiple-level experimental design were employed to determine the optimal medium constituents and to optimize carbon and nitrogen source concentrations for lovastatin production. Simultaneous quantitative analyses of the ${\beta}$-hydroxyacid form and ${\beta}$-hydroxylactone for of lovastatin were performed by the high performance liquid chromatography (HPLC) method with a UV photodiode-array (PDA) detector. The total lovastatin yield ($4{\sim}6\;mg/g$, average of five repeats) was achieved by adding soybean powder, glycerol, sodium nitrate, and acetic acid at optimized levels after 14 days of fermentation. The maximal yield of lovastatin under the optimal composition of the medium increased by almost 2 times the yield observed prior to optimization. The experimental results also indicated that the ${\beta}$-hydroxylactone form of lovastatin (LFL) and the ${\beta}$-hydroxyacid form of lovastatin (AFL) simultaneously existed in solid state cultures of Monascus ruber. while the latter was the dominant form in the middle-late stage of continued fermentation. These results indicate that optimized culture conditions can be used for industrial production of lovastatin to obtain high yields.
In Vitro Inhibitory Effect of Triterpenoidal Saponins from Platycodi Radix on Pancreatic Lipase

In the process of investigating anti-obesity effect of Platycodi Radix, we found that aqueous extract of Platycodi Radix might inhibit intestinal absorption of dietary fat by inhibiting pancreatic lipase (PL) activity. In order to clarify the anti-obesity mechanism of Platycodi Radix, activity-guided isolation was performed to find active components. The total saponin fraction of Platycodi Radix appeared to have a potent inhibitory activity against the hydrolysis of triolein emulsified with phosphatidycholine by pancreatic lipase in vitro. Based on these results, further purification of active components yielded 10 known triterpenoidal saponins, among these compounds, platycodin A, C, D, and deapioplatycodin D exhibited significant inhibitory effects on PL at the concentration of $500\;{\mu}g/mL$ with 3.3, 5.2, 34.8, and $11.67\%$ pancreatic lipase activity vs control, respectively. Platycodin D was found to inhibit the PL activity in a dose-dependent manner. Therefore, the anti-obesity effect of Platycodi Radix might be due to the inhibition of pancreatic lipase by its saponins.

Real-time Fourier transform infrared spectroscopy and optical microscopy demon-strated that helical segments formed along with the spherulite growth. Atomic force microscopy revealed the details of growth,twisting and branching of lamellar crystals. Cooperative packing of these twisting lamellae led to regular banded spherulites observedunder polarized light microscopy. Real-time observation on the crystallization process provided richer information than the characterization of the final structures; consequently, it provides deeper insight into the organization mechanism of the hierarchical structures.

야생 균주 Xanthophyllomyces dendrorhous으로부터 biomass와 astaxanthin 산물을 증가시키기 위해 shake flask배양 조건에서 astaxanthin산물에 영향을 미치는 물리적 인자, 영양 인자와 Carotenogenesis의 생성 자극 인자에 대한 연구를 HPLC분석 방법을 이용하여 수행하였다. 최적에 온도, 초기 pH값, 탄소와 질소 공급 조건에서 Carotenogenesis의 선구 물질인 acetic 산, mevalonic 산, 토마토 추출물과 당근 추출물은 astaxanthin 생산을 상당히 증가시킬 수 있었다. To improve biomass and astaxanthin production by wild-type Xanthophyllomyces dendrorhous simultaneously in shake flask culture, physical factors, nutritional factors and carotenogenesis stimulating factors affecting astaxanthin production were studied on base of HPLC quantitative analysis. The results suggested that carotenogenesis precursor composition acetic acid, mevalonic acid, tomato extract, and carrot extract could increase the productivity of astaxanthin markedly based on the optimized temperature, initial pH value, carbon and nitrogen sources conditions.
