The objective of this study was to observe the effects of water extracts of Rehmannia glutinosa on the glutamate-induced inward current that heterologously express human NMDA receptor. The N-methyl-D-aspartate (NMDA) receptor is one of three types of ionotropic glutamate receptors, the others being the AMPA and kainate receptors. It is activated when glutamate and glycine (or D-serine) bind to it, and when activated it allows positively charged ions to flow through the cell membrane. In the present study, we investigated the regulatory effects of the water extracts of Rehmannia glutinosa on the glutamate-induced inward current that heterologously express human NMDA receptor. The current was measured using the two-electrode voltage clamp technique. In this study, we found the water extracts of Rehmannia glutinosa inhibited glutamate induced current in a reversible and concentration-dependent manner. The IC50 was about 51.2 μg/ml. The water extracts of Rehmannia glutinosa action on glutamate induced current was attenuated by increasing glutamate concentration, and was membrane holding potential-dependent. These results show a possibility that the water extracts of Rehmannia glutinosa interacts with NMDA receptor, which was heterologously expressed and that the water extracts of Rehmannia glutinosa on glutamate induced current of NMDA receptor could be one of contributions of Rehmannia glutinosa-mediated neuroprotections.
