This study was performed to investigate the skin-whitening effect of Sabaeck-san. The inhibitory effect of methanolic extracts obtained from the Sabaeck-san & Mori Cortex, Glycyrrhizae Radix, Lycii Cortex Radicis which were consistant of Sabaek-san was tested on melanin synthesis. The inhibitory effect of Sabaeck-san, Mori Cortex, Glycyrrhizae Radix & Lycii Cortex Radicis on tyrosinase activity and on melanin production from dopachrome by autoxidation were measured. Ultimately, these exhibited skin-whitening effects, in vitro. The results were as follows: 1. Sabaeck-san & Mori Cortex exhibited strong inhibition. Especially, Sabaeck-san inhibited greater than Mori Cortex. Glycyrrhizae Radix also exhibited potent inhibition, but weaker than Sabaeck-san & Mori Cortex. Lycii Cortex Radicis had weak inhibition. 2. The amount of melanin synthesis was estimated by measurement of OD at 475㎚, and then the weight of standard melanin & the inhibitory percent of the test substance were determined. On inhibitory effects of the production of melanin, Sabaeck-san was the most potent, and the 2nd was Mori Cortex, the 3rd was Lycii Cortex Radicis and then Glycyrrhizae Radix. 3. In this experiment, Sabaeck-san showed much stronger anti-tyrosinase activity & inhibition of melanin production than each 3 herbs which were consistant of Sabaeck-san. These results suggest that Sabaeck-san & Mori Cortex showed strong inhibitory effect on tyrosinase & production of melanoma cell. Especially, Sabaeck-san extract showed relatively strong tyrosinase inhibitory activity & production of melanin compared to Mori Cortex in spite of small dosage of Mori Cortex in the Sabaecks-san than single extract of Mori Cortex.
