본 연구에서 Herpes simplex 1형과 2형 바이러스를 세포배양하여 숙주세포에서 클론을 분리하여 전자현미경에 의한 바이러스의 복제과정을 관찰하였으며, 제놈 DNA를 분리하여 제한효소 패턴을 분석하고 클로닝하여 제놈 library를 만들었다. Glycoprotein-B, D와 H 유전자가 포함되어 있는 재조합 플라스미드와 thymidine kinase 유전자를 밝히었고, 동시에 Herpes simplex 2형 바이러스에서도 이들 유전자들에 대한 상동성을 조사하였다. Glycoproteins의 면역학적 특성을 알아보기 위하여 단백질 발현을 대장균에서 immunoblotting으로 조사하였으며, baculovirus 발현체계를 이용한 Spodoptera frugiperda 21세포에서의 유전자 발현을 immunoblotting과 간접면역형광법에 의해 조사분석한 결과 다음과 같은 성적을 얻었다.
1) H. simplex 1형 바이러스에 대한 플라크 모양은 원형으로 작은 크기였고, 직경이 약 2.O~2.5 mm이었다. H. simplex 2형 바이러스에 대한 플라크 모양은 H. simplex 1형 바이러스 플라크 보다는 크고 불규칙한 원형이었으며 직경은 약 3.O~3.5 mm이었다.
2) H. simplex 1형과 2형 바이러스를 Vero 세포에 감염시켜 전자현미경으로 관찰하였을때 핵내에서 직경이 100~110 nm인 뉴클레오캡시드와 세포질에서 150 ~200 nm이상의 성숙된 바이러스를 관찰 할 수 있었으며, 이들 바이러스가 증식되는 과정을 관찰할 수 있었다.
3) H. simplex 1형과 2형 바이러스 제놈 DNA를 BamHI 등 18종의 제한효소로 절단한 결과 분자크기와 절단된 패턴은 차이가 있었으나 비슷한 양상을 보였다.
4) H. simplex 1형과 2형 바이러스 제놈 DNA의 BamHI 재조합 플라스미드는 BamHI 단편의 크기에 의해 각각 pHLA-l~27과 pHLB2-l~27로 명명하였으며, 분자크기는 1.13~13.87과 0.70~15.08 kb이었고, 바이러스 제놈 DNA의 BamHI 제한효소패턴과 비교하여 100%의 제놈 library를 얻었다. H. simplex 1형 바이러스 제놈 DNA의 BglⅡ 재조합 플라스미드 클론들의 분자크기는 4.55~20.1 kb이었고, pHLB-l~l5로 명명하였으며, H. simplex 1형 바이러스 제놈 DNA의 BglⅡ 제한 효소 패턴과 비교하여 하나의 단편만 재조합 플라스미드를 얻지 못했다. H. simplex 2형 바이러스 제놈 DNA의 EcoRI 재조합 플라스미드 클론들의 분자크기는 1.59~7.26 kb이었으며, pHLE2-1~5라 명명하였다.
5) H. simplex 1형과 2형 바이러스 제놈 DNA의 제한효소 단편에서 gB, gD 와 gH 유전자 위치를 밝히기 위해 southern blot에 의해 조사한 결과, gB와 gD 유전자 단편의 위치는 상이함을 알 수 있었고, gH 유전자는 동정되지 않았다. H. simplex 1형 바이러스 gB 유전자에 대한 상동성을 보인 H. simplex 2형 바이러스의 재조합체는 pHLA2-1과 25이었으며, 분자크기는 0.7과 12.7 kb 이었고, gD 유전자에 대한 상동성을 보인 pHLA2-6, 7, 10, 21과 22의 재조합체는 1.65, 1.77, 2.40, 5.85과 6.07, kb의 분자크기의 단편이었다.
6) H. simplex 1형 바이러스의 thymidine kinase 유전자는 pHLB-4와 pHLA-12에서 동정되었으며, 분자 크기는 각각 6.41, 3.74 kb이었고, H. simplex 2 형 바이러스에서는 pHLA2-11로 3.66 kb의 분자크기의 단편이었다.
7) 재조합 플라스미드 pHLA-21은 gB 유전자를 포함하는 단편으로 분자크기는 7.82 kb, pHLA-17은 gD 유전자를 포함하는 단편으로 6.43 kb, pHLB-4는 gH 유전자를 포함하는 단편으로 6.41 kb로 밝혀졌으며, H. simplex 1형 바이러스의 gB와 gH 유전자의 대장균내에서의 발현은 pHLA-21과 pHLB-4 재조합체로부터 pGEX-GB와 pET-GH를 만들어 SDS-PAGE상에서 나타난 단백질 패턴을 immunoblotting을 하였을때 분자량 121 kDa과 91 kDa에서 blot이 나타났다.
8) H. simplex 1형 바이러스의 gH 유전자에 대한 재조합 바이러스인 플라크와 바이러스 클론을 분리하여 BacPAK6-AcGH로 명명하였고, 이 재조합 바이러스를 발현시켜 SDS-PAGE상에서 나타난 단백질 패턴을 anti-human과 mouse의 serum을 사용하여 immunoblotting하였을때 분자량이 92 kDa에서 같은 blots이 나타났으며, Sf21세포내에서 발현된 단백질을 확인하기 위하여 Sf21세포와 초음파로 파쇄한 세포에서 형광항체를 뚜렷하게 관찰할 수 있었다.
이상의 결과를 종합하여 보면 H. simplex 1형 바이러스 gH 유전자가 발현한 단백질에 대한 면역성 연구와 동시에 H. simplex 2형 바이러스에서의 gH 유전자도 동정되어야 하며, 재조합 백신으로 gB 유전자는 그 유용성이 밝혀져 있어 이에 대한 연구와 최근에 H. simplex 2형 바이러스의 gG의 유전자가 H. simplex 1형 바이러스의 gD 유전자의 같은 ORF에 존재하는 유전자로 밝혀져 있어, gD 유전자와 병행하여 이에 대한 연구도 해야 한다는 결론을 얻었다.

In order to facilitate the molecular characterization of the Herpes simplex virus types 1(HSV-1) and 2(HSV-2) genome DNAs, a library of cloned restriction fragments have been produced. The glycoprotein-B(gB), the glycoprotein-D(gD) and the glycoprotein-H(gH) genes containing HSV-1 DNA were studied in an attempt to develop a recombinant vaccines.
The Vero cells were infected with HSV-1 and HSV-2. Forty-eight hours after infection, the infected cells were lysed, and multinucleated giant cells were observed approximately at seventy-two hours postinfection. The isolated plaques of HSV-1 and HSV-2 were designated as HL-1, HL-2, respectively.
The multiplications of HL-1 and HL-2 were observed in Vero cells using electromicroscopy. The nucleocapsids in nuclei were observed, and the assembled virions were budded out through the vacuole, and the virons were released from the cells.
HSV-1 and HSV-2 were analyzed by digestion of their genome DNAs with restriction enzymes. HSV-1 and HSV-2 genome DNAs were digested with BamHⅠ, BglⅡ, EcoRⅠ, respectively and cloned in Escherichia coli(E. coli) using the plasmid vector pBluescript SK(+) and pBacPAK9. The BamHI restriction fragments of HSV-1 and HSV-2 genome DNAs were cloned, and the same twenty-seven recombinant fragments resulted with the molecular sizes ranging from 1.13 ~ 13.87, 0.70~ 15.08 kilobases, respectively. These fragments were labelled in consecutive alphabetical orders, by other reaseacher, but I have named this series in numerical orders. And, after using BglⅡ restriction enzyme on HSV-1 genome DNA, fifteen fragments were cloned. And the sizes of these fragments ranged from 4.55~20.10 kilobases. Next, using EcoRI restriction enzyme on HSV-2 genome DNA, five fragments were cloned with the molecular sizes ranging from 1.59~7.26 kilobases. The cloned fragments were identified by the digestion of the recombinant DNAs with restriction enzymes and comparing by their electrophoretic mobilities in agarose gel with that of similarly digested uncloned DNAs. The presence of HSV-1 and HSV-2 sequences in the recombinant plasmids were confirmed by Southern blot analysis of plasmid DNA to HSV-1 and HSV-2 genome DNAs.
In the hybridization of the probe DNA blots, the gB, gD and gH genes of HSV-1 genome DNA were located on the fragments of BamHI-g, BamHI-jki and BglⅡ-m, respectively. The BamHI-g, BamHI-jki and BglⅡ-m fragments of HSV-1 genome DNA were cloned only into the pBluescript SK(+) BamHI site and pBacPAK9 BglⅡ site which was confirmed with Southern blotting. And the recombinant plasmids containing gB, gD and gH genes of HSV-1 genome DNA were identified as pHLA-21, pHLA-17 and pHLB-4, respectively, and the molecular sizes of there genes were 7.82, 6.43 and 6.41 kilobases, respectively. The gB gene containing HSV-2 recombinant plasmids were identified as pHLA2-1, pHLA2-25, respectively. And five recombinant plasmids containing HSV-2 gD gene were identified, using Southern blotting.
The pHLA-12, pHLB-4 and pHLA-11 recombinant plasmids containing thymidine kinase(TK) genes of HSV-1 and HSV-2 genome DNAs were identified by Southern blot analysis, the molecular sizes were 3.74, 6.41 and 3.66 kilobases, respectively. The genome location of the TK genes were found to be in a smilar position to that of HSV-1 and HSV-2 genome DNAs.
The gB and gH genes encoding the HSV-1 DNA were cloned into E. coli insertion vectors, pGEX-3X, pET-22b, respectively. The gB and gH were detected in E. coli cells, as polypeptides with molecular sizes of 121, and 92 kDa, respectively. And as expected, the gene for gH of HSV-1 was expressed in Spodoptera frugiperda 21 cells from recombinant baculovirus(BacPAC6-GH), constructed using a baculovirus transfer vector, pBacPAKg. The gH protein, 92 kDa were detected in immunoblot analysis and were shown by indirect immunofluorescence assay using anti-mouse polyclonal antibodies, and human serum on both techniques.
With, continued reseach, these resultes may continued toward ultimately developing a recombinant vaccines for HSV-1 and HSV-2.
