In order to investigate the effects of Bangpungtongseoungsan(here in after referred to BPTS) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BPTS extract) and fed for 13weeks.
Items of this experimental study are as follows.
Body weight change, diet intake change, final increse of body weight, the weight change of the adipocytes in body, the level change of ALT, AST, total cholesterol, LDL-Cholesterol, triglyceride, glucose, free fatty acid, total protein, albumin, A/G ratio, T-bilirubin and creatinine, the expression of β3AR and leptin gene in primary adipocytes, the expression of UCP-2, GLUT1, GLUT4, 5-HtP and LPL in primary adipocytes, the production change of TNF-α and leptin in primary adipocytes, the expression of leptin and β3AR in adipocytes tissue.
The following results have been obtained:
1. The change of body weight and the final increase of body weight were decreased significantly, and diet intake was a little decreased.
2. The amount of the adipocyte in body weight was decreased significantly.
3. The levels of ALT, total cholesterol, LDL-Cholesterol, glucose, free fatty acid were decreased significantly.
4. The levels of triglyceride, creatinine were decreased but did not show significance.
5. There were little change in levels of AST, total protein, albumin, A/G ratio, T-bilirubin.
6. In primary adipocytes, β3AR gene expression was increased significantly and leptin gene expression was decreased in 100 ㎍/㎖ density significantly.
7. In primary adipocytes, the expression of GLUT4, 5-HtP were decreased but did not show significance and there were little change in the expression of UCP-2, GLUT1, LPL.
8. In primary adipocytes, the amount of TNF-α was decreased in 100 ㎍/㎖ density significantly and the amount of leptin was decreased but did not show significance.
9. In adipocytes tissue, the expression of leptin was decreased and the expression of β3AR was increased.
