야생동물로부터 유래되는 감염병이 공중보건학적으로 주된 위협이 되고 있다. 야생동물들이 감염병의 보균자로서의 역할에 대한 인지나 위험성은 증가하고 있지만 이에 대한 연구나 이를 감시하기 위한 기법 혹은 체계는 만들어져 있지 않다. 감시가 필요한 감염병의 수가 증가하고 있고 이에 따라 이들 각각을 모두 검사하는 데는 높은 비용, 많은 시간 그리고 대용량의 샘플이 필요하다. 대용량 고효율 pPCR 플랫폼은 많은 수의 감염병을 빠르고 효율적으로 진단할 수 있는 좋은 방법이다. 이 연구는 대용량 고효율 플랫폼을 적용하여 야생동물 구조센터로 구조된 야생동물이 가지고 있는 감염병에 대해서 조사한 것이다.
전북야생동물구조센터에 2018년부터 2019년에 구조된 야생동물 중 혈액 및 기관, 폐, 대장(분변), 비장에 대하여 시료를 채취하였다. 총 121 마리의 환자에 대한 시료가 채취되었으면 그 중 조류는 55마리, 포유류는 66마리였다. 각 환자의 sample은 하나로 합친 후 DNA와 RNA를 동시에 추출하였다. 진단에 사용한 플랫폼은 19 종류의 병원체를 2번씩 검사하도록 제작되었고 한번에 48개의 샘플을 검사할 수 있게 하였다. 병원체가 검출된 검체에 대해서는 각각의 혈액과 기관에 대해 singleplex qPCR을 진행하여 어떤 혈액 혹은 기관에서 병원체가 검출된 것인지 확인하였다.
대용량 고효율 qPCR 플랫폼을 이용한 결과에서 Campylobacter jejuni, Campylobacter coli, Mycoplasma spp., Plasmodium spp. 총 4가지 병원체에서 양성이 나왔다. 각각 병원체의 유병률은Campylobacter jejuni (10/121, 8.3%), Campylobacter coli (1/121, 0.8%). Mycoplasma spp. (78/121, 64.5%) and Plasmodium spp. (15/121, 12.4 %)이었다. Singleplex qPCR을 통해서는 대부분 병원체는 대장(분변)에서 많이 검출되었음을 보였으며 특히 Campylobacter spp. 인경우엔 양성인 개체 모두 대장에서 병원체가 검출되었다.
이 연구는 구조 야생동물이 감염병 보균자로서의 역할을 할 수 있다는 위험성을 확인하였다.

Infectious disease derived from wild animals is a major threat to public health. While the risk of the role of wildlife as a reservoir of infectious disease is increasing, effective and efficient systems for monitoring for infectious disease have not been developed yet in Korea. The surveillance of wildlife-mediated infectious diseases is challenging because of high cost, long turnaround time and large sample volume. High-throughput real-time PCR (qPCR) platform can be a simple alternative for detecting pathogens in large numbers of samples. This is a pilot study to survey on the potential of rescued wild animals as reservoirs of infectious disease based on the high-throughput qPCR platform in the Republic of Korea. From 2018 to 2019, blood and tissue samples (trachea, lung, large intestine [including stool] and spleen) collected from wildlife rescued by the Jeonbuk wildlife center were included in this study. A total of 121 patients consisting of 55 birds and 66 mammals were included. The high-throughput qPCR platform was designed to diagnose 19 pathogens in duplicate against 48 samples simultaneously. Each tissue or blood was grinded and mixed one and then, total nucleic acid isolated from each tissue or blood simultaneously. The results showed that the samples were positive to Campylobacter jejuni (10/121, 8.3%), Campylobacter coli (1/121, 0.8%). Mycoplasma spp. (78/121, 64.5%) and Plasmodium spp. (15/121, 12.4 %). Singleplex qPCR confirmed that most pathogens were detected in the large intestine (including stool) and especially Campylobacter spp. was detected 100 % in the large intestine. This study suggested that wild animals have potential as infectious disease reservoirs. In a further study, phylogenetic analysis of each pathogen will be conducted to confirm relationship between wild animals and others.
