Background :
Hyangsayukgunja-tang is a traditional Korean medicine that is used to treat various gastrointestinal tract disorders such as anorexia, functional dyspepsia and gastric ulcer. Our purpose is to clarify that Hyangsayukgunja-tang increases appetite and affects gut hormones .
Gut hormones act peripherally to modulate digestion and absorption of nutrients. However they also act as neurotransmitters within the central nervous system to control food intake. Peptide YY(PYY) supress appetite, oppositely ghrelin increases appetite through afferent vagal fibers to the brain stem or directly to the hypothalamus.
Methods :
We administered high and low dose of Hyangsayukgunja-tang(High dose; 1g/kg Low dose; 200mg/kg) for 2 weeks. During 2 weeks, body weight and food intake were measured daily. 2weeks later we weighed epididymal and retroperitoneal white adipose tissues and liver. We examined the changes in plasma peptide level(ghrelin, amylin, PYY, leptin) and mRNA expression levels of ghrelin and Ghrelin o-acyltransferase(GOAT).
Result :
1. The weight & rate of body weight gain showed significant different patterns between administration of Hyangsayukgunja-tang and control by times(P<0.005, Generalized Estimating Equation). The rate of body weight gain in rats was significantly increased at 8, 12, 14 days after administration of a high dose of Hyangsayukgunja-tang(P<0.025, Mann-Whitney U test).
2. Feed efficiency showed significant different patterns between administration of Hyangsayukgunja-tang and control by times(P<0.005, Generalized Estimating Equation).
3. The plasma ghrelin level in rats was significantly increased for 14days after administration of a low dose of Hyangsayukgunja-tang(P<0.005, Mann-Whitney U test).
4. The plasma PYY level in rats was significantly decreased for 14days after administration of a high dose of Hyangsayukgunja-tang(P<0.005, Mann-Whitney U test).
5. The mRNA expression levels of ghrelin and GOAT was up-regulated.
Conclusion :
In rats, Hyangsayukgunja-tang increased the mRNA expression levels of ghrelin and GOAT, thus Hyangsayukgunja-tang increases appetite by enhance the plasma ghrelin level, oppositely inhibit the plasma PYY level.
