시스테인계 단백질 가수분해효소의 한 종류인 caspase는 세포사멸과정에 중요한 역할을 수행하고 있다. 그 중, 실행자 caspase인 caspase-7와 caspase-3는 서열이나 구조가 매우 유사하며, 기질이 가지는 DEVD 모티브 서열을 인지하는 기질에 대한 특이성도 유사하다. 비록 caspase-7과 caspase-3가 많은 유사성을 가지고 있으나, 세포사멸 과정 동안에 세포 내에서 서로 다른 위치에 존재한다고 알려져 있다. 이러한 사실은 세포사멸 동안에 caspase-3와 caspase-7이 각각 다른 특정한 기질을 분해하여 기능을 수행할 가능성을 내포한다. 이러한 점으로 미루어, 세포 사멸 동안에 caspase-7이 caspase-3와는 독립적인 기능을 할 가능성을 유추 할 수 있다. 그러나 현재까지 caspase-7의 연구는 caspase-3 연구와 비교했을 때 크게 활성화 되어 있지 못한 상태이다.
본 논문에서는, caspase-7의 새로운 기질을 동정하기 위하여, caspase-3가 결핍되어 있는 인간 유방암 세포주인 MCF-7의 세포 추출물에 재조합 caspase-7를 처리하여 사라지거나 줄어든 단백질을 이차원적 전기영동과 질량 분석기 방법을 통하여 동정하였다. 총 18개의 단백질이 동정되었는데 caspase-3와 구별된 caspase-7만의 특이적 기질을 확인 할 수 있었다. 특히 프로테아좀 서브유닛인 α2, α6, Rpt1이 세포사멸과정 중에 caspase-7에 의해 in vitro와 in vivo 에서 분해됨을 확인 하였다. 이러한 caspase-7에 의한 프로테아좀의 분해는 프로테아좀의 활성을 감소 시키고 세포내의 유비퀴틴화된 단백질들의 양을 크게 증가시켰다. 결과적으로, caspase-7에 의한 프로테아좀의 분해는 프로테아좀에 의한 단백질의 회전율을 줄어 들게 하여 세포사멸 과정의 진행을 촉진 시키게 된다.
본 연구에선 이차원적 전기영동법과 질량분석기를 이용하여 18개의 caspase-7의 새로운 기질을 동정하였고, 특히 caspase-7에 의한 프로테아좀 서브유닛의 분해는 세포사멸 과정의 진행을 촉진시킴을 확인하였다. 이러한 결과는 세포사멸을 통한 세포의 죽음에 관한 연구에 크게 도움을 줄 것이라 기대된다. 뿐만 아니라 프로테아좀은 암 등의 질병 치료를 위한 새로운 신약개발의 표적이 될 수 있을 것이다.

A family of cysteine proteases called caspases plays a critical role in the execution of apoptosis. Among caspases, caspase-3 and caspase-7 are closely related in sequences as well as in substrate specificity. Both caspases show overlapping substrate specificities with special preference for the DEVD motif. However, these highly related caspases are differentially localized in cells during apoptosis, implying the presence of caspase-3 or caspase-7 specific substrates. In this study, to identify new human caspase-7 substrates, I digested cell extracts of a caspase-3-deficient MCF-7 cell line with recombinant caspase-7 and analyzed eliminated or decreased spots by 2-dimensional electrophoresis and MALDI-TOF mass. The results show that several subunits of the proteasome including α2, α6, and Rpt1 undergo caspase-7 dependent proteolysis in vitro and in vivo during apoptosis. This caspase-7 dependent cleavage of proteasome subunits results in the reduction of the proteasome activity and thereby increases the amount of ubiquitinated proteins in the response to apoptotic stimuli. These data strongly suggest that similar to caspase-3, caspase-7 dependent proteolysis also reduces the protein turnover rate by the proteasome and facilitates the execution of the apoptotic program. These results show that our degradomic studies are very useful for mining the identification of unique and novel caspase-7 substrates. Furthermore, the list of candidate caspase-7 specific substrates provides valuable clues for studying apoptosis and may be used as a target for the discovery of novel drug that are associated with various diseases including cancer.
