        Effect of ACADL on the differentiation of goat subcutaneous adipocyte

        Objective: The aim of this study was to clone the mRNA sequence of the Acyl-CoA dehydrogenase long chain (ACADL) gene of goats and explore the effect of ACADL on the differentiation of subcutaneous fat cells on this basis. Methods: We obtained the ACADL gene of goats by cloning and used quantitative realtime polymerase chain reaction (qPCR) to detect the ACADL expression patterns of different goat tissues and subcutaneous fat cells at different lipid induction stages. In addition, we transfect intramuscular and subcutaneous adipocytes separately by constructing overexpressed ACADL vectors and synthesizing Si-ACADL; finally, we observed the changes in oil red stained cell levels under the microscope, and qPCR detected changes in mRNA levels. Results: The results showed goat ACADL gene expressed in sebum fat. During adipocyte differentiation, ACADL gradually increased from 0 to 24 h of culture, and decreased. Overexpression of ACADL promoted differentiation of subcutaneous adipocytes in goat and inhibited their differentiation after interference. Conclusion: So, we infer ACADL may have an important role in positive regulating the differentiation process in goat subcutaneous adipocytes. This study will provide basic data for further study of the role of ACADL in goat subcutaneous adipocyte differentiation and lays the foundation for final elucidating of its molecular mechanisms in regulating subcutaneous fat deposition in goats.

        Right-sided acute diverticulitis in the West: experience at a university hospital in Argentina

        Purpose: In the West, diverticular disease is located mainly in the left colon. However, it can also present in the right colon, with an incidence of 1%–2% in Caucasians. The purpose of this study was to describe our experience in right-sided acute diverticulitis (RD). Methods: In this retrospective study, 410 patients with acute diverticulitis treated from 2013 to 2020 were included in a university hospital in Córdoba, Argentina. Colonic diverticulitis was stratified into 2 groups; RD and left-sided acute diverticulitis. Demographic and clinical variables, laboratory and imaging findings, type of treatment, follow-up, and recurrence were analyzed. Results: Sixteen patients (3.9%) with RD were identified; 62.5% were male and the mean age was 40.7±11.7 years. A total of 81.3% were Caucasian and 18.7% Native American. Significant differences were found between both groups of diverticulitis; patients with RD were younger (P=0.001), with lower BMI (P=0.01), comorbidity rate (P=0.01), Charlson comorbidity index (P=0.02), hospital stay (P=0.01), severity according to the Hinchey classification (P=0.001) and had a lower recurrence rate (P=0.001). There were no significant differences in sex (P=0.95), duration of pain until admission (P=0.05), laboratory findings (P=0.23) and treatment (P=0.34). Conclusion: Conservative treatment predominated in RD, with a lower rate of complications and recurrences, providing data that support conservative therapy as initial treatment in RD in our environment.

        Effects of Dietary Supplementation with Ferulic Acid or Vitamin E Individually or in Combination on Meat Quality and Antioxidant Capacity of Finishing Pigs

        This study aimed to evaluate the effects of vitamin E (VE), ferulic acid (FA) and their combination supplementation on meat quality and antioxidant capacities of finishing pigs. Sixty barrows were randomly allocated to four experimental diets using a $2{\times}2$ factorial arrangement with 2 VE supplemental levels (0 or 400 mg/kg) and 2 FA supplemental levels (0 or 100 mg/kg) in basal diets. After 28 days, six pigs per treatment were slaughtered. The results showed that VE supplementation increased loin eye area of pigs (p<0.05) and FA supplementation increased $pH_{45min}$ value (p<0.05). The interaction of $FA{\times}VE$ was observed in shear force of longissimus dorsi muscle (p<0.05). Moreover, supplementation with VE decreased hepatic and sarcous malondialdehyde (MDA) content, increased hepatic glutathione (GSH) content and sarcous glutathione peroxidase (GSH-Px) activity (p<0.05). Additionally, supplementation with FA increased hepatic GSH-Px activity and decreased sarcous MDA content (p<0.05). However, dietary treatment did not affect the expression of genes related to nuclear factor, erythroid 2-like 2 (NFE2L2) pathway. These results suggest that dietary FA and VE could partially improve meat quality and antioxidant capacity of finishing pigs, but not by activating NFE2L2 pathway under the normal conditions of farming.

      • Linoleic Acid in Association with Malate or Fumarate Increased CLA Production and Reduced Methane Generation by Rumen Microbes

        An in vitro study was conducted to investigate the effect of propionate precursor (malate or fumarate) on fermentation characteristics, production of conjugated linoleic acid (CLA) and methane (CH₄) by rumen microbes when incubated with linoleic acid (C18:2) as a CLA precursor. Total CH₄ production for 12h incubation, however, was greatly reduced (P<0.0001) by all the supplements compared to that of control, and its production from M-LA or F-LA treatment was smaller than that from LA treatment. Supplementation of M-LA or F-LA also increased concentrations of cis9, trans11-CLA (P<0.039 - P<0.001) for all incubation times and trans10,cis 12-CLA at 1h (P<0.013), 3h(P<0.036) and 12h (P<0.025) incubation times compared to LA supplementation.

        The Effect of Contact Frequency with Customers on Relationship Satisfaction

        Purpose: This study is to examine the moderating role of contact mode and customer attachment style on the effects of contact frequency on relationship satisfaction. Design/methodology/approach: To explain this phenomenon, literature reviews about contact frequency, contact mode, relationship satisfaction, the moderating effect of contact mode (personal vs.impersonal), and attachment style (anxious vs.avoidant).SEM (Structure Equation Model) was suggest to check these relationships using PLS. Findings: Bootstrapping analysis was performed using process macros to find the relationship between contact frequency and relationship satisfaction.As a results, Contact frequency does not significantly affect satisfaction with a company; however, this independent variable affects satisfaction with a company via satisfaction with the service provider.Anxious people are known to be dependent, while avoidant people are independent, so customers’ attachment styles influence their relationship satisfaction.Personal contact modes, such as by telephone, are found to be more positive than impersonal contact modes, such as group email, for relationship satisfaction.The results of this study show that customers prefer personal contact to impersonal contact.Moreover, for both anxious and avoidant customers, frequent contact has positive effects on customer satisfaction with service providers and organizations. Research limitations/implications: This study provide two contributions.First, it confirmed the mediation effect of the customer-service-provider relationship.Customers’ satisfaction with a company depends on their satisfaction with its service providers.Second, this study identified a moderating role played by the contact mode and the customer’s attachment style.In regard to future research, it would be nice to add cases for increasing customer-satisfaction level with service firms and to focus on other industries to expand current research findings. Originality/value: The findings provide customer segmentation criteria and guidelines for effective relationship marketing activities in service field.

        Application of Linkage Disequilibrium Mapping Methods to Detect QTL for Carcass Quality on Chromosome 6 Using a High Density SNP Map in Hanwoo

        The purpose of this study was to detect QTL for carcass quality on bovine chromosome (BTA) 6 using a high density SNP map in a Hanwoo population. The data set comprised 45 sires and their 427 Hanwoo steers that were born between spring of 2005 and fall of 2007. The steers that were used for progeny testing in the Hanwoo Improvement Center in Seosan, Korea, were genotyped with the 2,535SNPs on BTA6 that were embedded in the Illumina bovine SNP 50K chip. Four different linkage disequilibrium (LD) mapping models were applied to detect significant SNPs for carcass quality traits; the fixed model with a single marker, the random model with a single marker, the random model with haplotype effects using two adjacent markers, and the random model at hidden state. A total of twelve QTL were detected, for which four, one, three and four SNPs were detected on BTA6 under the respective models (p<0.001). Among the detected QTL, four, two, five and one QTL were associated with carcass weight, backfat thickness, longissimus dorsi muscle area, and marbling score, respectively (p<0.001). Our results suggest that the use of multiple LD mapping approaches may be beneficial in increasing power to detect QTL given a limited sample size and magnitude of QTL effect.

        Methodological Study on Measurement of Hydrogen Abundance in Hydrogen Isotopes System by Low Resolution Mass Spectrometry

        China's rapid economic growth has resulted in significant environmental side effects. Therefore, China has been interested in reducing her dependence on foreign oil and gas by developing technologies needed for hydrogen, in addition to her increasing energy mix of nuclear and renewable energy form, such as solar and wind power. There are three isotopes of hydrogen, i.e. protium (P or H), deuterium (D), and tritium (T). Both deuterium and tritium are important materials in nuclear fuel cycle industry. Tritium is one of the critical radioactive nuclides. Planning for and implementing contamination control as a part of normal operation and maintenance activities is an important function in any hydrogen facility, especially tritium facility. The development of hydrogen isotopes analysis is the key issues in this area. Mass spectrometry (MS) with medium (about 600) and high resolution (> 1,400) is commercially available; however, the routine analysis of hydrogen isotopes is done with low-resolution MS (< 200) in China. This paper summarizes the progress of MS measurement technology for hydrogen isotope abundance in China, focusing on our lab's research program and technical status. An analyzing method has been introduced for accurate measurement of tritium abundance in the H.D.T system by low resolution MAT-253 MS. The quotient of compression ratio coefficient is determined by building up equipment for laboratory-scale preparation of secondary standard gases and by considering the difference in sensitivity between hydrogen isotopes. The results show that the measured value is reproducible within the relative error range of 0.8% for gas samples of different tritium abundance.
