        Estrogen receptor β promotes bladder cancer growth and invasion via alteration of miR-92a/DAB2IP signals

        Although early studies suggested that bladder cancer (BCa) is more prevalent in men than in women, muscle-invasive rates are higher in women than in men, suggesting that sex hormones might play important roles in different stages of BCa progression. In this work, we found that estrogen receptor beta (ERβ) could increase BCa cell proliferation and invasion via alteration of miR-92a-mediated DAB2IP (DOC-2⁄DAB2 interacting protein) signals and that blocking miR- 92a expression with an inhibitor could partially reverse ERβ-enhanced BCa cell growth and invasion. Further mechanism dissection found that ERβ could increase miR-92a expression at the transcriptional level via binding to the estrogen-response-element (ERE) on the 5′ promoter region of its host gene C13orf25. The ERβ up-regulated miR-92a could decrease DAB2IP tumor suppressor expression via binding to the miR-92a binding site located on the DAB2IP 3′ UTR. Preclinical studies using an in vivo mouse model also confirmed that targeting this newly identified ERβ/miR-92a/ DAB2IP signal pathway with small molecules could suppress BCa progression. Together, these results might aid in the development of new therapies via targeting of this ERβ-mediated signal pathway to better suppress BCa progression.

        Implementation of functional expansion tally method and order selection strategy in Monte Carlo code RMC

        The spatial distribution of neutron flux or reaction rate was calculated by cell or mesh tally in traditional Monte Carlo simulation. However, either cell or mesh tally leads to the increase of memory consumption and simulation time. In this paper, the function expansion tally (FET) method was developed in Reactor Monte Carlo code RMC to solve this problem. The FET method was applied to the tallies of neutron flux distributions of uranium block and PWR fuel rod models. Legendre polynomials were used in the axial direction, while Zernike polynomials were used in the radial direction. The results of flux, calculation time and memory consumption of different expansion orders were investigated, and compared with the mesh tally. Results showed that the continuous distribution of flux can be obtained by FET method. The flux distributions were consistent with that of mesh tally, while the memory consumption and simulation time can be effectively reduced. Finally, the convergence analysis of coefficients of polynomials were performed, and the selection strategy of FET order was proposed based on the statistics uncertainty of the coefficients. The proposed method can help to determine the order of FET, which was meaningful for the efficiency and accuracy of FET method.

        Proteomic Analysis of Goat Longissimus dorsi Muscles with Different Drip Loss Values Related to Meat Quality Traits

        Longissimus dorsi muscles from 3 goat species were assigned to high and low drip loss groups. Physio-chemical properties, sarcomere length, and proteome profiles were investigated. The high drip loss group had lower pH, higher brightness, and higher shear force values, and shorter sarcomere lengths than the low drip loss group. 22 differential proteins were identified between high and low loss groups. α-Enolase, NADH dehydrogenase, pyruvate dehydrogenase E1, HSP27, superoxide dismutase, peroxiredoxin-2, myosin, and the myosin light chain were among these proteins, which were metabolic enzymes, stress response factors, and structural proteins that affected glycolysis, oxidation, and muscle contraction. Drip loss was probably produced via proteins involved in glycolysis, oxidation, and muscle contraction.

        Transiting Exoplanet Monitoring Project (TEMP). II. Refined System Parameters and Transit Timing Analysis of HAT-P-33b

        <P>We present 10. R-band photometric observations of eight different transits of the hot Jupiter HAT-P-33b, which has been targeted by our Transiting Exoplanet Monitoring Project. The data were obtained by two telescopes at the Xinglong Station of National Astronomical Observatories of China (NAOC) from 2013 December through 2016 January, and exhibit photometric scatter of 1.6-3.0 mmag. After jointly analyzing the previously published photometric data, radial-velocity (RV) measurements, and our new light curves, we revisit the system parameters and orbital ephemeris for the HAT-P-33b system. Our results are consistent with the published values except for the planet to. star radius ratio (RP/R-*), the ingress/egress duration (tau) and the total duration (T-14), which together indicate a slightly shallower and shorter transit shape. Our results are based on more complete light curves, whereas the previously published work had only one complete transit light curve. No significant anomalies in Transit Timing Variations (TTVs) are found, and we place upper mass limits on potential perturbers, largely supplanting the loose constraints provided by the extant RV data. The TTV limits are stronger near mean-motion resonances, especially for the low-order commensurabilities. We can exclude the existence of a perturber with mass larger than 0.6, 0.3, 0.5, 0.5, and 0.3 M-circle plus near the 1: 3, 1: 2, 2: 3, 3: 2, and 2: 1 resonances, respectively.</P>

        Comprehensive transcriptome analysis discovers novel candidate genes related to leaf color in a Lagerstroemia indica yellow leaf mutant

        Lagerstroemia indica is a popular woody ornamental plant throughout the world. However, relatively little is known about the molecular processes regulating leaf color in L. indica compared with other ornamental plants. Although yellow leaf mutants from various organisms have been well characterized, L. indica yellow leaf mutant has not yet been reported. In this study, a L. indica yellow leaf mutant, named YL03, was characterized and its leaf transcriptome was sequenced. A total of 30,712,752 reads were generated and assembled de novo into 45,308 unigenes with an average length of 987.51 bp. Among these unigenes, 21,339 (47.10 %) were identified as putative homologs of annotated sequences in public databases. A total of 79 unigenes involved in chlorophyll biosynthesis and degradation, photosynthesis and chloroplast development were identified. The expression levels of those genes were detected using quantitative real-time PCR in this study. Among those genes, 11 unigenes showed highly significant difference in the mutant compared to wild type plants. Conclusively, the leaf color formation is greatly affected by the activity of chloroplast development and chlorophyll metabolism. And the possible formation pathway of yellow leaf mutant is deduced based on our results.

        Cloning and Functional Characterization of Putative Escherichia coli ABC Multidrug Efflux Transporter YddA

        A putative multidrug efflux gene, yddA, was cloned from the Escherichia coli K-12 strain. A drugsensitive strain of E. coli missing the main multidrug efflux pump AcrB was constructed as a host and the yddA gene was knocked out in wild-type (WT) and drug-sensitive E. coliΔacrB to study the yddA function. Sensitivity to different substrates of WT E.coli, E. coliΔyddA, E. coliΔacrB and E. coliΔacrBΔyddA strains was compared with minimal inhibitory concentration (MIC) assays and fluorescence tests. MIC assay and fluorescence test results showed that YddA protein was a multidrug efflux pump that exported multiple substrates. Three inhibitors, ortho-vanadate, carbonyl cyanide m-chlorophenylhydrazone (CCCP), and reserpine, were used in fluorescence tests. Ortho-vanadate and reserpine significantly inhibited the efflux and increased accumulation of ethidium bromide and norfloxacin, while CCCP had no significant effect on YddA-regulated efflux. The results indicated that YddA relies on energy released from ATP hydrolysis to transfer the substrates and YddA is an ABC-type multidrug exporter. Functional study of unknown ATP-binding cassette (ABC) superfamily transporters in the model organism E. coli is conducive to discovering new multidrug resistance-reversal targets and providing references for studying other ABC proteins of unknown function.

        Microbially induced calcium precipitation based anaerobic biosynthetic crystals for removal of F− and Ca2+ in groundwater: Performance optimization, kinetics, and reactor operation

        Anaerobic biosynthetic crystals (ANBC) were prepared based on microbially induced calcium precipitation (MICP) and their potential explored for groundwater defluoridation and decalcification. The preparation conditions of ANBC were optimized and the influence of key factors (initial fluoride ions (F−) concentration, pH, and initial calcium ions (Ca2+) concentration) on the crystals was investigated. During the operation of the reactor, at pH of 7.0, the hydraulic retention time (HRT) of 6 h, and Ca2+ concentration of 180 mg L−1, a maximum removal efficiency reached 93.31%, while 66.20% of Ca2+ could be removed. The adsorption dynamics study showed that the adsorption of ANBC was most in line with the pseudo-second-order model. The stability of ANBC operation was studied and failure reaction showed that the crystals maintained a stable removal ability after 35 times of repeated use. Further studies found that this was attributed to the continuous growth and synthesis of the crystals. The defluoridation and decalcification mechanism was further explored by scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and X-ray diffraction spectra (XRD). This study innovatively proposes a method for biosynthesis of crystals under anaerobic conditions based on MICP, which can efficiently and stably remove F− and Ca2+ in groundwater, providing a valuable strategy for groundwater contaminant remediation and energy saving.

        Preparation and Photocatalytic Activity of ABi2O6/BiOCl (A = Mg, Sr, Ca) Heterojunction

        MgBi2O6, CaBi2O6, and SrBi2O6 were successfully synthesized by a substitution reaction of NaBiO3 with MgCl2, CaCl2, and Sr(NO3)2, respectively. Then, ABi2O6/BiOCl (A = Mg, Ca, Sr) heterojunction compounds with different ABi2O6:BiOCl ratios were prepared by treating ABi2O6 with an appropriate amount of HCl. The color of the ABi2O6/BiOCl heterojunction was changed from dark to bright as the amount of BiOCl was increased. The absorbances of the ABi2O6/BiOCl heterojunctions in the visible range were decreased as the ratio of BiOCl became higher. The photocatalytic activities of the heterojunction samples were higher than that of sole ABi2O6 or BiOCl. The highest photocatalytic efficiency was achieved when the ABi2O6:BiOCl ratio was 15:85. The organic pollutant was decomposed completely within 1 h under visible light when the 15:85 catalyst was used. The reactive species in the photocatalytic reaction with the ABi2O6/BiOCl photocatalysts were ·O2- and h+ H+.

        Isogeometric analysis of functionally graded CNT-reinforced composite plates based on refined plate theory

        A simple and effective approach based on refined plate theory (RPT) is proposed to study the static and free vibration characteristics of functionally graded CNTreinforced composite (FG-CNTRC) plates. Compared to traditional higher order shear deformation theories (HSDTs), the proposed method shows more efficient for FG-CNTRC plates analysis. To solve the C1-continuity requirement of the RPT, we used isogeometric analysis (IGA) to approximate the displacement field, which shows more advantages than the FEA, since it can construct higher-order elements without additional variables. This is an advantage for plate structural analysis because more variables make the calculation cumbersome. Finally, four types of FG-CNTRC plates were investigated and the results show the accuracy and efficiency of the proposed method.

        Comparison of totally laparoscopic and laparoscopic-assisted approach in gastrectomy with D2 lymphadenectomy for advanced gastric cancer after neoadjuvant chemotherapy: a retrospective comparative study

        Purpose: Neoadjuvant chemotherapy is strongly recommended for advanced gastric cancer due to good local control and a high rate of R0 dissection with this strategy. Minimally invasive techniques such as laparoscopy-assisted or total laparoscopic approaches is becoming more and more acceptable in the treatment for gastric cancer. However, the safety and efficiency of total laparoscopic D2 gastrectomy (TLG) for advanced gastric cancer after neoadjuvant chemotherapy have not been well evaluated. Methods: A retrospective study in a single center from 2014 to 2016 was conducted. A total of 65 locally advanced gastric cancers were treated by laparoscopy-assisted gastrectomy (LAG) or TLG. Parameters which include operation time, blood loss, complications, hospital stay, 3-year overall survival, and 3-year disease-free survival were used for comparison. Results: The time of operation in the TLG group was shorter than in the LAG group (P = 0.013), blood loss was less (P = 0.002) and time to first flatus was shorter (P = 0.039) in the TLG group than that in the LLG group. Intraoperative and postoperative complications were comparable in both groups. No significant difference was found in 3-year overall and disease-free survival. Conclusion: For patients with locally advanced gastric cancer after neoadjuvant chemotherapy, laparoscopic D2 gastrectomy can be considered as a safe and efficient alternative. A further multicenter prospective randomized controlled study is needed to elucidate the applicability of this technique for advanced gastric cancer.
