McCoy 세포에서 Chlamydia trachomatis의 병원성 인자에 관한 연구

본 연구는 병원성 세균인 Chlamydia trachomatis(C. trachomatis)가 숙주세포에침습시 C. trachomatis의 병원성 인자와 C. trachomatis와 상호작용하는 숙주 세포막의 인자를 규명함으로써 숙주세포에 대한 감염율을 향상시켜 보다 향상된 진단 방법을 개발하기 위하여 수행하였다. C. trachomatis로 감염된 세포에서 기본소체의 막 단백질을 분리한 결과 각 혈청형의 39 kDa과 42 kDa에서 major outer membranes(MOMPs)이 관찰되었으며, 혈청형 E와 G의 MOMPs는 42.5 kDa으로 동일하였지만 혈청형 F, H, I, 그리고 K의 MOMPs는 각각 다르게 관찰되었다. C. trachomatis가 숙주세포로 침습시 관련된 병원성 인자에 대하여 조사하고자 단층세포로 배양된 배양용기에 C. trachomatis를 24, 48 그리고 73시간대별로 접종시켜 관찰한 결과 72시간의 MOMPs의 양이 가장 많이 증가하였으며 이 결과로 MOMPS이 C. trachomatis 병원성에 중요한 작용을 함을 확인할 수 있었다. C. trachomatis가 침습시 숙주세포의 막 단백질 변화양상을 관찰하기 위해 정상세포와 C. trachomatis로 1, 2, 3 그리고 4시간대별로 접종 시간을 달리한 숙주세포의 막 단백질을 분리한 결과 42 kDa의 단백질 양이 Chlamydia에 감염된 세포 내에서 증가됨을 관찰할 수 있었다. 이로 42 kDa의 막 단백질이 침습시 숙주세포 표면에 대한 수용체로서 작용함을 알 수 있었다. The present study was performed to analyze the pathogenic factors involved in the invasion process of Chlamydia trachomatis(C. trachomatis) into McCoy cells. In this study, elementary bodies(EBs) of C. trachomatis serotypes(E, F, G, H, I, K and LGV) could be isolated by the method of Percoll density gradient centrifugation and the pathogens of serotypes were also compared from whole-cell lysates by using sodium dodecyl sulfate-polyacrylamide gel elelectrophoresis. All chlamydial serotypes showed a single predominant protein ranging from 39 to 42 kDa. The major outer membrane proteins(MOMPs) of the E and G serotypes had an identical molecular weight of 42.5 kDa. In contrast, the MOMPs of the F, H, I and K serotypes were showed different molecular weights of MOMPs. By immunoblotting with anti-MOMP antibody it was revealed that the quantify of MOMPs was time-dependently increased when McCoy cells were infected with Chlamydia for 24, 48 and 72 hr. These results indicated that MOMPs are closely related to chlamydial infection into the host cell.

<P>PURPOSE: To evaluate the effect of neoadjuvant combined chemotherapy and radiation therapy (CCRT) on preoperative accuracy of multidetector computed tomography (CT) for resectability and tumor staging in patients with pancreatic head cancer. MATERIALS AND METHODS: This retrospective study received institutional review board approval and was exempted from informed consent requirements. From May 2002 to March 2007, 38 patients with pancreatic head adenocarcinoma underwent multidetector CT before surgery. Of these, 12 patients received neoadjuvant CCRT. Imaging findings were evaluated for tumor resectability and tumor staging. Surgical and pathologic results were used as the reference standard. The accuracy of resectability and individual components of each T category were compared between the patients with neoadjuvant CCRT and without it by using the chi(2) test or Fisher exact test. A P of less than .05 was considered as significant. RESULTS: The accuracy in determining resectability was 83% (10 of 12) in patients who had received neoadjuvant CCRT and 81% (21 of 26) in patients who had not, without significant difference (P > .05). Of 32 patients who underwent pancreaticoduodenectomy, histopathologic tumor staging was reported for T1 (n = 2), T2 (n = 1), and T3 (n = 9) lesions in patents with neoadjuvant CCRT (n = 12), and for T3 in all patients without neoadjuvant CCRT (n = 20). T-staging accuracy was 67% (eight of 12) with neoadjuvant CCRT and 95% (19 of 20) without it, with a significant difference (P = .0185). CONCLUSION: Neoadjuvant CCRT reduces the accuracy of tumor restaging after treatment of pancreatic head cancer, but this effect is not so great as to affect the determination of resectability.</P>

<P>Technical strategies for improving the device characteristics of the In-Ga-Zn-O (IGZO) vertical channel thin-film transistors (VTFTs) were presented and investigated. The vertical sidewall was constructed by dry-etch process and subsequently covered with IGZO, Al2O3, and AZO as active, gate insulator, and gate electrode layers by means of conformal atomic-layer-deposition. An abrupt profile and flat back-channel were achieved by employing the spin-coated polyimide (PI) spacer. The Off-current was additionally alleviated simply by cutting the area of an active layer. The fabricated IGZO VTFT using PI spacer with an 'active-cut' structure exhibited an On/off ratio of 10(3), a linear mobility of 7.1 cm(2)/Vs, and a subthreshold swing of 1.2 V/decade.</P>

Hybrid cell line that produced MAbs against Chlamydia trachomatis surface antigens were prepared by the fusion of mouse myeloma cells with BALB/c mouse. Those Mabs were used to examine the antigenic specificity and biological function of several Chlamydia serotypes. Using those MABs, chlamydial inclusions were detected by using chemiluminence and it turns out that the chemiluminents method was very useful to detect the chlamydial inclusions or infection. It was confirmed that this method is easy to detect and to reduce the time consuming compared with examination of chlamydial inclusions order fluorescence microscopy.

<P><B>Abstract</B></P><P>Bisphosphonates have been commonly used for the treatment of osteoporosis. However, there have been recent case reports of atypical fractures citing their long-term use, which inhibits the turnover of bone components.</P><P>A 64-year-old woman visited the outpatient clinic with pain in her right thigh and ambulation difficulty. We found fractures at both pedicles of L4 vertebra. subtrochanteric region of right femur, and left femoral shaft upon a radiologic examination. She had taken intravenous ibandronic sodium for osteoporosis over 3 years.</P><P>We changed the bishophonates to a parathyroid hormone because it was suspected that the multiple fractures were caused by the medication. Further, rehabilitation, including progressive weight bearing, was started. After 3 months of the conservative treatment, she was able to walk independently.</P><P>In conclusion, it is necessary to evaluate the possibility of atypical fractures in osteoporotic patients when they complain of lower extremity pain and to consider alternative treatments instead of bisphosphonates.</P>
생활의 질적 향상을 위한 부엌의 작업대 및 수납장에 관한 연구

The purpose of this study was to examine present conditions of kitchen work centers and cabinets and to determine responses of residents in relation to them. The sample comprised of 42 households in Seoul. A questionnaire was developed, which consisted of three parts; sociodemographic information of the households, present conditions of kitchen work centers and cabinets, and their responses in relation to them. Data was analyzed by hand, using frequency and percentage. Findings from this study were as follows: 1. L shape and one wall kitchen shape were dominated, 55.0% and 32.0% respectively. The kitchen work centers were arranged most frequently in sequence of refrigerator center, sink center, mixing center, cooking center, and serving center. 2. Widths/heights of kitchen work centers and cabinets were close to the recommended measurements, however the vertical distance between the top line of kitchen work centers and the bottom line of wall cabinets showed higher than the recommended measurement. 3. Residents were satisfied to some degree with the sequence of arrangement, heights, widths, design, and color of kitchen work centers and cabinets, however they need more storage space. 4. The functional aspect and design were the major factors influenced residents' decision making for the buying behavior of the kitchen work centers and cabinets.

국내산 수수(Sorghum bicolor) 품종에 따른 전자공여능, 환원력, 항균활성 및 α-glucosidase 저해활성의 비교

수수의 기능성식품으로써 이용 가능성을 조사하기 위해 수수의 70% ethanol 추출물의 전자공여능, 환원력, 총 페놀함량, 총 플라보노이드함량, 항미생물, α-glucosidase 억제활성을 측정하였다. 전자공여능 측정 결과 10 μg/mL 농도에서 메수수, 붉은장목 수수가 94%로 BHT(13%), BHA(74%)보다 월등히 높은 활성을 보였다. 환원력을 측정한 결과 농도가 높아짐에 따라 활성이 증가하는 경향을 보였으며, 수수 22개 품종 중에서 메수수가 0.71의 OD값으로 가장 높은 항산화 활성을 나타내었다. 추출물의 총 페놀 함량은 빗자루수수가 22.9 mg GAE/g로 높은 함량을 나타내었으며, 흰수수가 16.4 mg GAE/g로 가장 낮은 함량을 보였다. 총 플라보노이드 함량은 장목수수가 3.5 mg QE/g로 높은 함량을 보였다. 항미생물 실험 결과 E. coli에서 모든 수수가 활성을 나타내었으며, 특히 시경수수(MIC=8 μg/mL)와 찰(금산)수수(MIC=8 μg/mL)가 뛰어난 활성을 나타내었다. α-Glucosidase 억제활성 측정 결과 붉은장목수수, 목탁수수, 긴장목수수가 98%로 높은 억제율을 보여주었다. Electron donating ability (EDA), reducing power, total phenolic contents, total flavonoid contents, antimicrobial activity, and α-glucosidase inhibitory effects of twenty-two sorghum varieties were examined using ethanol extracts (70%, v/v). The electron donationg abilities (EDA) of Mesusu and Bulgeunjangmoksusu (94% at 10 μg/mL) indicated higher antioxidant activity compared to those of other varieties and standard antioxidants such as BHT (13%) and BHA (74%). The reducing power of Mesusu (OD<SUB>700</SUB>=0.71) suggested higher antioxidant activity, which was dependent on sample concentration. Bitjarususu showed the highest content of total phenolics (22.9 mg GAE/g); however, extracts from Heuinsusu exhibited the lowest content of total phenolics (16.4 mg GAE/g). Jangmoksusu showed the highest total flavonolic contents (3.5 mg QE/g), and Sigyeongsusu and Chal (GS) susu displayed the most antibacterial activity (MIC=8 μg/mL) against Escherichia coli. Extracts of Bulgeunjangmoksusu, Moktaksusu, and Ginjangmoksusu showed the highest α-glucosidase inhibitory effect (98%) at the concentration of 5 μg/mL.
