      • IL-12 Regulates B7-H1 Expression in Ovarian Cancer-associated Macrophages by Effects on NF-κB Signalling

        Background and Aim: B7-H1, a co-inhibitory molecule of the B7 family, is found aberrantly expressed in ovarian cancer cells and infiltrating macrophage/dendritic-like cells, and plays a critical role in immune evasion by ovarian cancer. IL-12, an inducer of Th1 cell development, exerts immunomodulatory effects on ovarian cancer. However, whether IL-12 regulates B7-H1 expression in human ovarian cancer associated-macrophages has not been clarified. Therefore, we investigated the effects of IL-12 on the expression of B7-H1 in ovarian cancer-associated macrophages and possible mechanisms. Methods: PMA induced THP-1-derived macrophages or human monocyte-derived macrophages were treated with recombinant IL-12 (rIL-12) or infected with adenovirus carrying human IL-12 gene (Ad-IL-12-GFP) for 24 h, then cocultured with the SKOV3 ovarian cancer cell line for another 24 h. Macrophages were collected for real-time PCR and Western blot to detect the expression of B7-H1, and activation of the NF-${\kappa}B$ signaling pathway. Moreover, supernatants were collected to assay for IL-12, IFN-${\gamma}$ and IL-10 by ELISA. In addition, monocyte-derived macrophages treated with IFN-${\gamma}$ were cocultured with SKOV3 and determined for the expression of B7-H1. Furthermore, the expression of B7-H1 in monocyte-derived macrophages was also evaluated after blocking NF-${\kappa}B$ signaling. Results: The expression of B7-H1 was significantly upregulated in monocyte-derived macrophages treated with rIL-12 or Ad-IL-12-GFP compared with the control groups (p<0.05), accompanied by a remarkable upregulation of IFN-${\gamma}$ (p<0.05), a marked downregulation of IL-10 (p<0.05) and activation of NF-${\kappa}B$ signaling. However, the upregulation of B7-H1 was inhibited by blocking the NF-${\kappa}B$ signaling pathway (p<0.05). Expression of B7-H1 was also increased (p<0.05) in monocyte-derived macrophages treated with IFN-${\gamma}$ and cocultured with SKOV3. By contrast, the expression of B7-H1 in THP-1-derived macrophages was significantly decreased when treated in the same way as monocyte-derived macrophages (p<0.05), and IL-10 was also significantly decreased but IFN-${\gamma}$ was almost absent. Conclusions: IL-12 upregulates the expression of B7-H1 in monocyte-derived macrophages, which is possible though inducing the secretion of IFN-${\gamma}$ and further activating the NF-${\kappa}B$ signal pathway. However, IL-12 downregulates the expression of B7-H1 in THP-1-derived macrophages, associated with a lack of IFN-${\gamma}$ and inhibition of expression of IL-10.

      • Visual Landmark Design, Detection and Identification for Vision-guided CAD Model-based Mobile Robot Localisation

          A new pattern landmark has been designed and its detection and identification algorithms are proposed for an efficient CAD model-based localisation system for a vision-aided mobile robot. The known horizontal projection model of the vertical wall closed to the landmark, together with the sensed positions based on a CAD-model relative to the robot, help to determine the robot"s current position without any reference to its previous positions. The landmark is used to guide the searching process while the local CAD model is used for localisation. The accurate co-ordinates of the landmark are not important. A self-similar landmark with bar-colour-coding pattern after defining a "barlet" function is designed such that an upward-aiming camera can reliably detect it even if it is partially blocked by unforeseeable obstructions. The landmark can be effectively detected and recognised by using wavelet-based MRA for landmark searching and matching the colour pattern one by one with the pre-tested colour bands of HSV space for landmark recognition. Experimental results show the feasibility of the proposed landmark and its identification algorithm for localisation.

        Microstructure and Mechanical Characteristics of Dissimilar TIG Welded 9% Cr Heat-Resistant Steels Joints

        Dissimilar 9% Cr heat-resistant steels (G115 and CB2) with good creep properties for ultra-supercritical steam turbines were butt-joined by tungsten inert gas welding. The microstructure of welded metal (WM) was quenched martensite without carbide precipitates and lath packets existed inside prior austenite grains (PAGs), which leaded to higher hardness of WM. Partially melted zone at G115 side was composed of untempered martensite within equiaxed PAGs. The lowest hardness occurred in both G115 and CB2 steels which was attributed to tempered martensite with many M 23 C 6 precipitates. The heat-affected zone consisted of three sub-grains and their microstructure was detailly analyzed in current work. As current increased from 130 to 150 A, both the tensile strength at room temperature and 650°C increased while strength had no obvious change with further increasing current. The values of 673 MPa and 309 MPa corresponded to the tensile stress with 150 A at room temperature and 650°C, respectively. The fracture mode of joints at room temperature was cleavage and ductile failure at 130 and 150 A, respectively. The high-temperature fracture surface at 150 A was composed of deep and fi ne dimples.

        Single-cell RNA sequencing reveals B cell–related molecular biomarkers for Alzheimer’s disease

        In recent years, biomarkers have been integrated into the diagnostic process and have become increasingly indispensable for obtaining knowledge of the neurodegenerative processes in Alzheimer’s disease (AD). Peripheral blood mononuclear cells (PBMCs) in human blood have been reported to participate in a variety of neurodegenerative activities. Here, a single-cell RNA sequencing analysis of PBMCs from 4 AD patients (2 in the early stage, 2 in the late stage) and 2 normal controls was performed to explore the differential cell subpopulations in PBMCs of AD patients. A significant decrease in B cells was detected in the blood of AD patients. Furthermore, we further examined PBMCs from 43 AD patients and 41 normal subjects by fluorescence activated cell sorting (FACS), and combined with correlation analysis, we found that the reduction in B cells was closely correlated with the patients’ Clinical Dementia Rating (CDR) scores. To confirm the role of B cells in AD progression, functional experiments were performed in early-stage AD mice in which fibrous plaques were beginning to appear; the results demonstrated that B cell depletion in the early stage of AD markedly accelerated and aggravated cognitive dysfunction and augmented the Aβ burden in AD mice. Importantly, the experiments revealed 18 genes that were specifically upregulated and 7 genes that were specifically downregulated in B cells as the disease progressed, and several of these genes exhibited close correlation with AD. These findings identified possible B cell-based AD severity, which are anticipated to be conducive to the clinical identification of AD progression.

        The Antifungal Mechanism of Electrolyzed Oxidizing Water against Aspergillus flavus

        Fungicidal electrolyzed oxidizing waters (EOW)include neutralized electrolyzed oxidizing water (NEW)and acidic electrolyzed oxidizing water (AcEW). Theseforms were evaluated against Aspergillus flavus conidia(spores) and mycelia. EOW possessed strong antifungalactivities against A. flavus conidia and mycelia due todestruction of the cellular structures of the A. flavusconidium and mycelium. K+ and Mg2+ leakage results fromdamage to the normal cellular functions of A. flavus conidiaand mycelia. Among the physicochemical parameters ofEOW, the available chlorine concentration (ACC) isprimarily responsible for the EOW antifungal activity. A. flavus can be eliminated by use of AcEW and NEW as anew, alternative method of fungal control that is superior tosome current physical methods and to synthetic chemicalfungicides.

        Network isolation system (NIS) can help electrical vehicle charging station (EVCS) to exchange messages with different systems (such as central monitor system, battery package and electrical vehicle (EV)) in a secure and efficient way. This paper focuses on the structure of EVCS, and then analyzes the boundaries of the trusted and the untrusted areas. On the top boundary, EVCS can communicate with the central monitor system. On bottom boundary, EVCS can communicate with EVs or battery packages. Based on this demarcation, this paper proposes the NIS architecture for EVCS. The new architecture can limit the top/bottom boundary of EVCS to enhance the securities

        Inhibition of cancer cell growth and migration by dihydroxynaphthyl aryl ketones

        Dihydroxynaphthyl aryl ketones 1-5 exhibit activity as tubulin polymerization inhibitors by targeting the colchicine binding site of microtubules making them potential anticancer drugs. Therefore, analogues 1-5 have been evaluated for their cytotoxic activity against the cancer cell lines DU-145 (prostate), T24 (bladder) and MCF-7 (breast). notable differences in biological activity were observed for compounds 1-5, most likely related to the nature of the aryl substituent bonded to the carbonyl group. among the tested compounds, only compound 5 showed selectivity for cancer cells over healthy, non-transformed cells. T24 cancer cells treated with compound 5 presented a concentration-dependent decrease in cell proliferation and a loss of migration ability. The cytotoxicity of compounds 1-5 on the selected cell-based assays is discussed in terms of it lipophilicity and polarizability parameters.

        Experimental Study of the Shear Capacity of Steel Beam-to-L-CFST Column Connections

        This paper aims to investigate the shear performance of a new and innovative type of vertical stiff ener connection between steel beams and L-shaped columns composed of concrete-fi lled steel tubes connected by transverse and vertical steel plates (L-CFST columns) in high-rise residential buildings. Quasi-static cyclic loading tests were performed on fi ve full-scale specimens to investigate the panel zone behaviours. Based on the experimental results, the hysteretic responses, skeleton curves of the shear force–deformation, ductility, stiff ness degradation, energy dissipation and strain are discussed. The variables studied in these experiments include the joint type, axial compression ratio, cross-sectional area and width-to-thickness ratio of the vertical stiff ener, and presence of concrete. The results indicate that the cross-sectional area of the vertical stiff eners plays a critical role in the performance of the panel zone. Two types of failure modes were observed outside the panel zone: fracturing of the vertical stiff eners and fracturing of the beam fl ange connecting plate. Shear deformation of the panel zone was obvious in the exterior joint specimens, and the corresponding maximum shear deformation reached 0.05 rad. Furthermore, the scopes of the panel zone in the corner and exterior joint specimens were determined by the strain distribution.

        Optimization of medium components for D-ribose production by transketolase-deficient Bacillus subtilis NJT-1507

        Statistical experimental designs were used to optimize the composition of culture media for the production of D-ribose by Bacillus subtilis. A fractional factorial design 2(5-2) was used to determine medium components that significantly affected D-ribose production. The concentrations of glucose and (NH4)2SO4 were the significant factors. Central composite design and response surface methodology were then used to estimate the quadratic response surface and determine the factor levels for maximum production of D-ribose. Finally, the optimal medium composition was obtained (g/L): glucose, 172.75; (NH4) 2SO4, 13.2; yeast powder, 4; corn steep liquor, 8 and MnSO4, 0.5. This optimization strategy increased D-ribose production from 73.21 g/L to 88.57 g/L, an increase of 22% compared with the original conditions. The D-ribose production yield to glucose concentration was also enhanced from 0.37 g/g to 0.52 g/g. Confirmatory experiments were also performed to demonstrate the accuracy of the model. Under the optimal medium using ammonia to control pH in a 5 L fermenter, the D-ribose yield was increased to 95.28 g/L after 3 days of cultivation at 37 oC.

      • Prognostic Significance of Preoperative Serum Alpha-fetoprotein in Hepatocellular Carcinoma and Correlation with Clinicopathological Factors: a Single-center Experience from China

        Objectives: To investigate the prognosis significance of preoperative serum alpha-fetoprotein (AFP) and the correlation with clinicopathological factors of hepatocellular carcinoma (HCC) patients who underwent hepatectomy. Materials and Methods: Clinicopathological data of retrospective analysis were collected for 251 HCC patients undergoing hepatectomy in this study. According to preoperative AFP level, patients were categorized into AFP-negative (0-20ng/mL) and AFP-positive (>20 ng/mL) groups for Kaplan-Meier analysis and Cox proportional hazard regression modeling. Results: The results demonstrated that increased AFP was associated with longer prothrombin time (PTs), liver capsule invasion, low grade differentiation, and late Barcelona Clinic Liver Center (BCLC) stage. Moreover, the female patients had a greater prevalence of increased preoperative AFP than male patients [284.8 (3.975-3167.5) vs (3.653-140.65); Z-2.895, p=0.004]. The 1-, 3-, and 5-year recurrence-free survival (RFS) rates were 78.1, 57.5, and 40.6 % in the AFP-negative group and 61.8, 37.7, and 31.4 %, respectively, in the AFP-positive group (log-rank test 8.312, p=0.004). The 1-, 3-, and 5-year overall survival (OS) rates were 94.4, 83.8, and 62.3% in the AFP-negative group and 87.2, 60.0, and 36.7%, respectively, in the AFP-positive group. The difference was statistically significant (log-rank test, 16.884, p=0.000). Cox proportional-hazards model identified preoperative AFP to be an independent prognostic predictor of overall survival. Conclusions: Preoperative serum AFP is an independent predictor of prognosis among HCC patients following surgical resection. Female patients have a higher preoperative AFP than their male counterparts.
