<P><B>Abstract</B></P> <P>Pyrogallol is naturally found in aquatic plant and has been proposed as a substrate of tyrosinase. In this study, we evaluated the dual effect of pyrogallol on tyrosinase as an inhibitor in the presence of L‑DOPA simultaneously via integrating methods of enzyme kinetics and computational molecular dynamics (MD) simulations. Pyrogallol was found to be a reversible inhibitor of tyrosinase in the presence of L‑DOPA and its induced mechanism was the parabolic non-competitive inhibition type (<I>IC</I> <SUB>50</SUB> = 0.772 ± 0.003 mM and <I>K</I> <SUB>i</SUB> = 0.529 ± 0.022 mM). Kinetic measurements by real-time interval assay showed that pyrogallol induced rapid inactivation process composing with slight activations at the low dose. Spectrofluorimetry studies showed that pyrogallol mainly induced regional changes in the active site of tyrosinase accompanying with hydrophobic disruption at high dose. The computational MD simulations further revealed that pyrogallol could interact with several residues near the tyrosinase active site pocket such as HIS61, HIS85, HIS259, ASN260, HIS263, VAL283, and ALA296. Our study provides insight into the mechanism by which hydroxyl group composing pyrogallol inhibit tyrosinase and pyrogallol is a potential natural anti-pigmentation agent.</P>

Boron defect engineered boron nitride (BN) nanosheets have been prepared with the barbituric acid as co-reactant sources. By virtue of B defects, the electronic structure of BN undergo significantly variation, in which the conduction band of BN declined from −1.35 to −0.54 eV and decreased chemical acid hardness. After B defect engineering, BN nanosheets display greatly improved adsorptive desulfurization performance towards dibenzothiophene (DBT), with optimized adsorption capacity of 43.6 mgS g−1 for model oil. The increased adsorptive desulfurization performance was derived from decreased chemical acid hardness by B defect engineering, thus can build preferable interplay between the soft base DBT.
Innovation Agglomeration and Chinese Spatial Economic Disparities

Optimizing distribution of innovative capitals and promoting collaborative innovation are important ways to achieve regional coordinated development in China. Using panel data of 30 China’s provinces during the period of 1990~2010,this paper studies effects of innovation agglomeration on the industrial agglomeration and economic growth. The study results show that: Innovative capital accumulation is an important feature in China, but the accumulation degrees of innovative capital between provinces are very different. Meanwhile, knowledge spillovers show strong geographical limitation. Innovation agglomeration and local knowledge spillovers play significant roles on promoting industrial agglomeration and the region's economic growth. But the uneven distribution of innovation plus the diminishing knowledge spillovers effect may lead to a further degree of regional income disparities.

A new pattern landmark has been designed and its detection and identification algorithms are proposed for an efficient CAD model-based localisation system for a vision-aided mobile robot. The known horizontal projection model of the vertical wall closed to the landmark, together with the sensed positions based on a CAD-model relative to the robot, help to determine the robot"s current position without any reference to its previous positions. The landmark is used to guide the searching process while the local CAD model is used for localisation. The accurate co-ordinates of the landmark are not important. A self-similar landmark with bar-colour-coding pattern after defining a "barlet" function is designed such that an upward-aiming camera can reliably detect it even if it is partially blocked by unforeseeable obstructions. The landmark can be effectively detected and recognised by using wavelet-based MRA for landmark searching and matching the colour pattern one by one with the pre-tested colour bands of HSV space for landmark recognition. Experimental results show the feasibility of the proposed landmark and its identification algorithm for localisation.
Experimental study on performance of steam-water injector with central water nozzle arrangement

The steam-water injector (SI) is a simple mechanical device that has been widely used in industry. Wedid an experimental study to find the influence of physical and geometrical parameters on performance of the SI. Thephysical parameters studied were steam inlet pressure, water inlet pressure and water inlet temperature. Whereas, thegeometrical parameters studied were steam nozzle area ratio, area ratio of steam nozzle to water nozzle and the mixingsection converging angle. Pump head was introduced to evaluate the lifting-pressure performance of the SI under dif-ferent operating and geometrical conditions. Optimal values of steam nozzle area ratio and mixing section convergingangle were 1.3 and 11.6orespectively, for the present work, and optimal value of area ratio of steam nozzle to waternozzle increased with increasing water inlet pressure. Two head-capacity curves were introduced to highlight the effectof various physical and geometrical parameters on the performance of SI.
miR-186 Regulates Glycolysis through Glut1 During the Formation of Cancer-associated Fibroblasts

Emerging evidence has suggested that glycolysis is enhanced in cancer-associated fibroblasts (CAF), and miR-186 is downregulated during the CAF formation. However, it is not clear whether miR-186 is involved in the regulation of glycolysis and what the role of miR-186 plays during the CAF formation. In this study, quantitative PCR analysises show miR-186 is downregulated during the CAF formation. Moreover, miR-186 targets the 3' UTR of Glut1, and its overexpression results in the degradation of Glut1 mRNA, which eventually reduces the level of Glut1 protein. On the other hand, knockdown of miR-186 increased the expression of Glut1. Both time course and dose response experiments also demonstrated that the protein and mRNA levels of Glut1 increase during CAF formation, according to Western blot and quantitative PCR analyses, respectively. Most importantly, besides the regulation on cell cycle progression, miR-186 regulates glucose uptake and lactate production which is mediated by Glut1. These observations suggest that miR-186 plays important roles in glycolysis regulation as well as cell cycle checkpoint activation.

To isolate novel and useful microbial enzymes from uncultured gastrointestinal microorganisms, a fecal microbial metagenomic library of the pygmy loris was constructed. The library was screened for amylolytic activity, and 8 of 50,000 recombinant clones showed amylolytic activity. Subcloning and sequence analysis of a positive clone led to the identification a novel gene (amyPL) coding for α-amylase. AmyPL was expressed in Escherichia coli BL21 (DE3) and the purified AmyPL was enzymatically characterized. This study is the first to report the molecular and biochemical characterization of a novel α-amylase from a gastrointestinal metagenomic library.

We determined a minimum effective dose of gallic acid (3,4,5-trihydroxy benzoic acid; 50 mg/ kg, i.p.) and piperine (10 mg/kg, p.o.) through their therapeutic potential and further evaluated them individually and in combination against beryllium-induced biochemical alterations and oxidative stress consequences in female albino rats. The administration of beryllium altered blood biochemical variables by significantly depleting hemoglobin, albumin and urea, whereas it enhanced bilirubin and creatinine. The release of serum transaminase, lactate dehydrogenase and γ-glutamyl transpeptidase was significantly greater, and was concomitant with a decrease in serum alkaline phosphatase. A significant increase in lipid peroxidation and a decrease in glutathione, superoxide dismutase and catalase in the liver and kidney was an indication of oxidative stress due to beryllium exposure. Individual administration of gallic acid and piperine moderately reversed the altered biochemical variables, whereas the combination of these was found to completely reverse the beryllium-induced biochemical alterations and oxidative stress consequences. We concluded that gallic acid exerts a synergistic effect when administered with piperine and provides a more pronounced therapeutic potential in reducing beryllium-induced hepatorenal dysfunction and oxidative stress consequences.

Hypothyroidism alone can lead to myocardial fibrosis and result in heart failure, but traditional hormone replacement therapy does not improve the fibrotic situation. Hydrogen sulfide (H2S), a new gas signaling molecule, possesses antiinflammatory, antioxidant, and anti-fibrotic capabilities. Whether H2S could improve hypothyroidism-induced myocardial fibrosis are not yet studied. In our study, H2S could decrease collagen deposition in the myocardial tissue of rats caused by hypothyroidism. Furthermore, in hypothyroidism-induced rats, we found that H2S could enhance cystathionine-gamma-lyase (CSE), not cystathionine β-synthase (CBS), protein expressions. Finally, we noticed that H2S could elevate autophagy levels and inhibit the transforming growth factor-β1 (TGF-β1) signal transduction pathway. In conclusion, our experiments not only suggest that H2S could alleviate hypothyroidism- induced myocardial fibrosis by activating autophagy and suppressing TGF-β1/ SMAD family member 2 (Smad 2) signal transduction pathway, but also show that it can be used as a complementary treatment to conventional hormone therapy.
