      • NDP kinase 2 interacts with two oxidative stress-activated MAPKs to regulate cellular redox state and enhances multiple stress tolerance in transgenic plants

        NDP kinases (NDPKs) are multifunctional proteins that regulate a variety of eukaryotic cellular activities, including cell proliferation, development, and differentiation. However, much less is known about the functional significance of NDPKs in plants. We show here that NDPK is associated with H_(2)O_(2)-mediated mitogen-activated protein kinase signaling in plants. H_(2)O_(2)stress strongly induces the expression of the NDPK2 gene in Arabidopsis thaliana (AtNDPK2). Proteins from transgenic plants overexpressing AtNDPK2 showed high levels of autophosphorylation and NDPK activity, and they have lower levels of reactive oxygen species (ROS) than wild-type plants. Mutants lacking AtNDPK2 had higher levels of ROS than wild type. H_(2)O_(2) treatment induced the phosphorylation of two endogenous proteins whose molecular weights suggested they are AtMPK3 and AtMPK6, two H_(2)O_(2)-activated A. thaliana mitogen-activated protein kinses. In the absence of H_(2)O_(2) treatment, phosphorylation of these proteins was slightly elevated in plants overexpressing AtNDPK2 but markedly decreased in the AtNDPK2 deletion mutant. Yeast two-hybrid and in vitro protein pull-down assays revealed that AtNDPK2 specifically interacts with AtMPK3 and AtMPK6. Furthermore, AtNDPK2 also enhances the myelin basic protein phosphorylation activity of AtMPK3 in vitro. Finally, constitutive overexpression of AtNDPK2 in Arabidopsis plants conferred an enhanced tolerance to multiple environmental stresses that elicit ROS accumulation in situ. Thus, AtNDPK2 appears to play a previously uncharacterized regulatory role in H_(2)O_(2)-mediated MAPK signaling in plants.

        Effects of High-Hydrostatic Pressure on Ginsenoside Concentrations in Korean Red Ginseng

        The effects of high-hydrostatic pressure (HHP) on the ginsenoside concentration in Korean red ginseng were investigated. HHP-pretreated Korean red ginseng samples were compared to samples produced by a conventional method. Six-year-old Korean fresh ginseng (Panax ginseng C.A. Meyer) samples were vacuum-packaged in polyethylene film and treated at room temperature for 1 min with HHP (200-600 MPa) and steamed at 98oC for 3 hr. Major ginsenosides of red ginseng were analyzed by HPLC. HHP-pretreated red ginseng showed a 45% higher level of total major ginsenosides than conventionally prepared red ginseng. The levels of 4 protopanaxadiol-type ginsenosides increased 34-43% and the levels of 5 protopanaxatriol-type ginsenosides increased 45-66%. Scanning electron microscopy and electrical conductivity spectrum analysis showed that HHP pretreatment damaged ginseng plant cells and increased extraction efficiencies of ginsenosides from red ginseng products.

        Experiencing the Art/Craft/Handmade/Studio Furniture Aesthetics in Postmodernism Theoretically (I) -Defining the Postmodern furniture aesthetics as Metaphor-

        This study explored the theoretical context of postmodern aesthetics in experiencing art/craft/handmade/studio furniture for 'wood culture experience class' or 'wood love experience class', which has been recently carried out by the Korea Forest Service and is apt to focus on making simple wood work just involved in the practical part. Qualitative conceptual analysis as the principal methodology was used to achieve the theoretical context in the experience programs. I developed the lesson for an aesthetics of art and everyday life with wood furniture which expresses the metaphorical aesthetics appeared as New Design furniture in postmodernism. As statement of objective in the lesson, I explored that students in elementary schools personally understand and define the postmodern aesthetics of New Design furniture as metaphor. As a result, the students will be able to define and identify New Design furniture as metaphor in their lives.

        This study explored interpretation of the symbolical and metaphorical aesthetics in experiencing art/craft/handmade/studio furniture theoretically for 'wood culture experience class' or 'wood love experience class' by the Korea Forest Service because the class tends to focus on making simple wood works involved just in the practical part. Qualitative conceptual analysis as the principal methodology was used to achieve the theoretical context in the experience programs. After the lesson one on defining the postmodern aesthetics in New Design furniture as metaphor in the previous paper, I developed lesson two for an aesthetics of art and everyday life with wood furniture which expresses the metaphorical and symbolical aesthetics appeared as New Design furniture in postmodernism. As statement of objective in the second lesson, I explored that students in elementary schools personally transform and interpret the postmodern aesthetics of New Design furniture metaphorically with the meanings of symbol. As a result, the students will be able to write about the aesthetics of New Design furniture as metaphor in interpretation transformed by his/her perspective of a piece of selected New Design furniture.

      • Toxicogenomics an method for assessing toxicological effect of chemicals: A Study on Statins

        Since there are increasing needs for newly developed drugs, the number of laboratory animals used for drug-safety research is increasing as well. This world-wide need for new drugs increases eventually the necessity to develop more elaborated assay system for drug efficacy-safety test which can replace old-time animal tests. To this purpose, genomic approaches based on microarray technology are now considered as a revolutionary tool for understanding the mechanism underlying drug-cell interaction and widely employed along the drug development procedures. Genomic biomarkers such as a set of genes will be served as surrogated biomarkers for assessing efficacy-safety of drugs. In this presentation, we will introduce an in vitro system based on microarray technology to assess statin-induced myopathy. Statins are competitive hydroxy-3-metyl glutaryl coenzyme A (HMG-CoA) reductase inhibitors that block the synthesis of cholesterol from mevalonic acid. Statins are the drugs most frequently used to reduce plasma cholesterol levels and decrease cardiovascular events. However, statins have been reported to cause myopathies, such as myalgias, myositis and even rhabdomyolysis in rareoccasions. Even the side effects associated with statins have been highlighted by the withdrawal of cerivastatin from the market in 2001, but little is currently known about the effect of statins on the RNA expression profile of skeletal muscle cells in myopathy. To address those issues, we set up an in vitro assay system using differentiated myocytes from rat L6 myoblasts. By using this in vitro system, we assayed the cytotoxicity and creatine kinase activity induced by two well-known statins, cerivastatin and pravastatin. As expected, only the cytotoxicity measured with MTT assay showed a linear correlation to the applied statin concentrations provingthat CK activity assay is not a proper method to detect muscle toxicity in vitro system. After the microarray analysis of gene expression, we discoveredgenes whose expression is changed in dose-dependent manner by cerivastatin. After statistical validation, fifteen of them were selected as potential candidates for statin-induced myopathy biomarkers, more precisely statin-induced cytotoxicity biomarker. Although further investigation on the genes whose expression is highly correlated with statin treatment, it seems that gene expression analysis by microarrays is a mean of choice to understand the mechanism of action and toxicity of a drug.

        Sensing Technologies for Grain Crop Yield Monitoring Systems: A Review

        Purpose: Yield monitoring systems are an essential component of precision agriculture. They indicate the spatial variability of crop yield in fields, and have become an important factor in modern harvesters. The objective of this paper was to review research trends related to yield monitoring sensors for grain crops. Methods: The literature was reviewed for research on the major sensing components of grain yield monitoring systems. These major components included grain flow sensors, moisture content sensors, and cutting width sensors. Sensors were classified by sensing principle and type, and their performance was also reviewed. Results: The main targeted harvesting grain crops were rice, wheat, corn, barley, and grain sorghum. Grain flow sensors were classified into mass flow and volume flow methods. Mass flow sensors were mounted primarily at the clean grain elevator head or under the grain tank, and volume flow sensors were mounted at the head or in the middle of the elevator. Mass flow methods used weighing, force impact, and radiometric approaches, some of which resulted in measurement error levels lower than 5% (R<sup>2</sup> = 0.99). Volume flow methods included paddle wheel type and optical type, and in the best cases produced error levels lower than 3%. Grain moisture content sensing was in many cases achieved using capacitive modules. In some cases, errors were lower than 1%. Cutting width was measured by ultrasonic distance sensors mounted at both sides of the header dividers, and the errors were in some cases lower than 5%. Conclusions: The design and fabrication of an integrated yield monitoring system for a target crop would be affected by the selection of a sensing approach, as well as the layout and mounting of the sensors. For accurate estimation of yield, signal processing and correction measures should be also implemented.

In this study, changes in the methylation of runt-related transcription factor-3 (RUNX3) were investigated in CCD18Co normal colon cells and DLD-1 colorectal adenocarcinoma cells.</P>

        Clinical Significance of Erosive or Ulcerative Lesions Isolated in Terminal Ileum

        Background/Aims: Although terminal ileal erosive or ulcerative lesions are frequently observed on colonoscopic examination, their clinical significance are unclear. We evaluated clinical course and significance of isolated terminal ileal erosive or ulcerative lesions. Methods: We retrospectively analyzed clinical features, number, size and histologic findings of 186 patients with isolated terminal ileal erosive or ulcerative lesions on colonoscopic examination from December 2003 to February 2012. Results: The indications for colonoscopy included screening for colorectal cancer or surveillance in 122 patients (65.6%), evaluations for symptoms in 64 patients (34.4%). Of the 186 patients, 170 underwent biopsy at the terminal ileal lesions. Histologic findings were mostly non-specific chronic inflammation except two cases of Crohn`s disease, one case of cytomegalovirus ileitis, and one case of intestinal tuberculosis. Forty six patients underwent follow-up colonoscopy and the mean duration was 17.8±14.2 months (range, 1-64 months). Of those who showed non-specific ileitis (44 patients), 35 (79.5%) showed resolution of lesions without specific treatment. In the remaining 9 (20.5%) patients, lesions were continued and two patients were diagnosed as Crohn`s disease and Behcet`s ileitis, respectively. There were no significant differences in the duration of follow-up, presence of symptoms, number and size of terminal ileal lesions between the patients who resolved and not resolved. Conclusions: Most isolated terminal ileal erosive or ulcerative lesions reveal non-specific histological findings and have a propensity to resolve without treatment. However, in small portions of patients, isolated terminal ileal lesions need careful attention because it is possible to be early inflammatory bowel diseases. (Intest Res 2012;10:350-356)
