To investigate the volatile flavor components of perilla leaves, analyses of kinds of cultivar by GC, GC/MS, UV, IR and NMR were carried out. The results obtained were summarized as follows : 1. The essential oils were extracted with diethyl ether after steam disillation. 2. The identified volatiles include 10 hydrocarbones, 6 alcohols, 3 aldehydes, 2 ketones and 1 miscellaneous components. 3. The major volatile component perilla ketone was identified by UV, IR and NMR spectrum. 4. The perilla ketone were identified in Suwon 8, 19, 21, 23, 24, 25, 26 and purpurea while perillaldehyde was identified in purpurea only.
Expression of Streptokinase Gene in Bacillus Subtillis

Streptokinase gene (Skc) containing the promoter and signal sequence was cloned in B. subtilis using the pUB140 plasmid vector. The streptokinase gene was expressed and streptokinase was secreted into culture broth. The coding sequence of the streptokinase gene was also expressed as fused gene using promoter and signal sequence of α-amylase gene from B. subtilis NA64 strain. In this α-amylase secretion vector system, streptokinase activity in culture broth was two to three-fold higher than that with its own promoter and signal sequence.

For the ultimate elucidation of complete nucleotide sequennce of the defective interfering particle(DI particle) of vesicular stomatitis virus (VSV), the complementary double strand DNA (ds cDNA) of single strand RNA (ss RNA) of DI-2 was first synthesized in vitro using a sequential enzymatic reactions (RNA ligase, 5'-kinase, RNA ligase, reverse transcriptase, OH- hydrolysis of RNA-DNA hybrid and reverse transcriptase). The resultant ds cDNA of the DI-2 RNA was confirmed temporarily by size ~tsucrose gradient sedimentation analysis) and hybridization experiment.

Lipase of Staphylococcus sp. was purified from the culture supernatant, and its molecular mass estimated to be 44 kDa by SDS-PAGE. Its optimum temperature and pH for the hydrolysis of p-nitrophenyl substrates was 28℃ and pH 8.5, respectively The gene encoding the lipase was cloned in Escherichia coli in the NH₂-teminally truncated form by using the shotgun method, and sequenced. The mature enzyme had a 49-93 % amino acid sequence homology with other staphylococcal lipases. This lipase was used for the hydrolysis of the 3-O-acetate of cephalosporin-C to give an intermediate, deacetylated cephalosporin-C that is useful for further chemical elaborations.
Chemical Modification of Sheep Hemoglobin with Methoxy - Polyethylene Glycol

Sheep hemoglobin (SHb) was modified with methoxy-polyethylene glycol (mPEG) to develop a potential blood substitute. mPEG has been used to decrease antigenicity and immunogenicity of foreign proteins. When the mPEG was attached to SHb, the modified hemoglobins showed decreased electrophoretic mobility on SDS-PAGE and decreased free amino groups. When the remaining free amino groups of mPEG modified SHb were determined by TNBS free amino group titration methods, about 34% of total free amino groups were modified with mPEG. This mPEG-SHb conjugate of 34% amino groups modified showed no precipitation by double immunodiffusion with polyclonal antibodies against SHb. This modified hemoglobin still has oxygen transport activity. So this antigenicity decreased hemoglobin may be used in humans as a potential blood substitute.
Nuclear Polyhedrosis Virus of Fall Webworm , Hyphantria cunea ( Drury )

Since the serious attention has given to the use of insect pathogenic viruses for the biological control of insect pests, we tried to isolate the infectious virus from the diseased larvae of fall webworm, Hyphanlria cunea, which is a pest of roadside trees and diciduous trees. The isolated virus was able to succeed in replication of H. cunea virus in an established insect cell line derived from the ovary of adults of cabbage looper, Trichoplusia ni. Based on pathological and morphological properties (by EM pictures), it was found that this virus is included in nuclear polyhedrosis virus (Baculovirus subgroup a). The polyhedral inclusion bodies ranged from 0.6 to 2.5 ㎛ in diameter and rodshaped nucleocapsids were enveloped in multiples of up to 22. Polyhedral protein preparations from H. cunea NPV and A. California NPV (reference virus) were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). In SDS-PAGE, the major protein component ranged from 28,000 to 29,000 dalton among the isolates. It was also found that the genome of H. cunea was DNA.
쥐 우유중의 Myo - Inositol 생성에 관한 연구

A high concentration of myo-Inositol in rat's milk was observed (61-91㎎. of myo-Inositol per 100g of milk) by gas-liquid chromatographic method, using a 3% SE-52 column. Feeding experiments showed that approxirnately 85% of myo-Inositol in mills was from dietary origins the rest was considered to be synthesized by 1L-myo-Inositol-l-phosphate lyase. Results suggested that the biosynthesis was not sufficiently high to permit the maintenance of its myo-Inositol level in milk. However, study using(^(14)C)-glucose injection into lactating female rats confirmed biosynthesis myo-Inositol from glucose in mammary gland. This biosynthesis reached a maximum within an hour after (^(14)C)-glucose injection intraperitoneally as lactose biosynthesis did. Study using (³H)-myo-Inositol confirmed that most of the myo-Inositol in milk was transported from blood plasma myo-Inositol against a concentration gradient. About four hours after the beginning of the injection of (^(14)C)-glucose, the specific radioactivity myo-Irositol in milk was 8% of that of glucose in the blood. When (³H)-myo-Inositol was injected, the specific radioactivity of myo-Inositol in milk was about 26% of that of blood six hours after injection.
