        For the unconfined vapor cloud explosion(UVCE) accident by the continuous release of gas-liquid flow of various saturated liquids in a vessel at ground level, overpressures were estimated by TNT equivalency model with two estimation methods, such as UVCE Ⅰ model based on a constant release time and UVCEⅡ model based on a real travel time of vapor by dispersion and analyzed with various release conditions. As a simulation result, the simple, easy, and correct method of evaluation of consequences of the UVCE accident was proposed by using consequences of UVCEⅠ model and correlation equations for differences of overpressures between UVCE models, so that this evaluation method could be used easily in the small and medium size enterprises without using the dispersion model.

        효과적인 SCM 구축을 위한 Positioning & Strategy에 관한 사례연구

        Recently, an enterprise's survival strategy is a difficult task, because the competition of a business is more complex ever. The research aims to demonstrate the dynamic structural change in a conceptual point of SCM positioning & strategy. But it has many problems since it is difficult and wide to define. Therefore, the Manager don't aware a total executable implementation even though the enterprise knows necessity of SCM. This paper focuses on a proposal of alternatives with reasonable positioning like most of enterprises willing to overcome such as carelessness and unready strategy of investment, additionally, the thesis should effort to find an element through an analysis of cases, a statistical method of effective SCM, and actual survey to propose an alternative between Positioning & Strategy in Enterprise SCM.

      • 여드름양 발진에서의 말라쎄지아 포자 검출 빈도에 관한 연구

        Steroid acne and pityrosporum folliculitis have similar clinical manifestations. So it was not casy to do differential diagnosis of these two diseases. We have tried to find out the different points between these diseases in clinical characteristics, positive rate of the malassezia spores, and their response to antifungal agents. We studied 21 patients with acneiform eruptions which were distinct from acne vulgaris. Patients were divided into two groups by steroid treatment history. Among 21 patients, 6 were administered steroid just before eruption and 15 were free from steroid within 6 months. We have examined malassezia spores with 10% KOH/Parker blue black ink mount from papules, pustules and comedones according to the patients skin lesions. The malassezia spores detected in 5 paticnts(83.3%) in the former and 9 patients(60%) in the latter. In malassezia folliculitis, the frequency of which showed above the (3+) score of malassezia spore under microscopy was higher than in steroid acne. Mean duration of the disease was 70.4 weeks in latter group, which was longer than 5.2 weeks of the former group. And there were some differences between two groups in the distribution of the lesions. Mean treatment duration was same in two groups (4 wks, 3.5 wks). Steroid acne patients would be treated better with antifungal agents than with acne regimens.

      • 재래식 시장 지하 상인의 호흡기증상 및 폐기능 조사 연구

        The prevalence of respiratory symptoms and pulmonary function were studied in a group of 55 whole-sale market merchants Working in underground. As a control group, 55 whitecollar workers was also included in this study. Following results were obtained. A higher complaint for respiratory symptoms(cough, sputum, chest tightness, shortness of breath) was found among merchants than control workers. The measurement value for FVC, FEV1.0, FEV 1.0% in the merchants were reveal to lower figure in comparison with control groups. The measurement Value for MVV, PEF in the merchants were lower than control groups, but was not significant statistically. The pattern of pulmonary ventilation disturbance in the whole-sale market merchant were the same as followings : restrictive disturbance(6 person), abstrictive disturbance(1 person), and combined disturbance(1 person).

        Transcranial magnetic stimulation can diagnose electrical burn-induced myelopathy

        <P><B>Abstract</B></P><P><B>Introduction</B></P><P>We conducted motor- and sensory-evoked potential analyses and investigated their diagnostic value in differentiating between electrical burns without evidence of neurologic injury and those with clinical evidence of myelopathy.</P><P><B>Methods</B></P><P>We studied high-voltage electrical burn injury patients with lower extremity weakness and evidence of myelopathy and those without any evidence of neurological complications. Motor-evoked potentials (MEPs), somatosensory-evoked potentials (SEPs), and MRI studies of the spinal cord, as well as transcranial magnetic stimulations, were performed. The central motor conduction time (CMCT) was also calculated.</P><P><B>Results</B></P><P>The upper limb MEP and upper and lower limb SEP variables did not statistically differ between the 2 groups. The CMCT and total motor conduction time recorded in the lower limb were delayed in the burn myelopathy group. MRI revealed no abnormal signal changes in myelopathy patients.</P><P><B>Conclusion</B></P><P>Our study demonstrates that MEP is useful in identifying myelopathy in patients who have sustained high-voltage electrical burns.</P>

      • Proteinase Activity of Naegleria spp. with Special Reference to their Pathogenicity

        자유생활아메바의 병원성과 단백분해효소 활성도의 상관성을 밝히고자 병원성인 Naegleria fowleri와 비병원성인 N. gruberi 영양형의 용해물과 배양액으로부터 단백분해효소 활성도를 관찰하였다. 또한 Chinese hamster ovary(CHO) 세포를 사용하여 51Cr 방출 검사법과 주사 전자현미경법으로 세포독성 물질을 평가(assay)하였고 단백분해효소의 생화학적 성질을 규명한 결과 다음과 같은 결과를 얻었다. Naegleria 영양형을 마우스에 접종시킨 후 사망율을 관찰한 결과 N. fowleri를 접종시킨 마우스에서 사망율이 75% 이었으나, N. bruberi는 사망율이 0%로 병원성이 없음을 확인하였다. 여러가지 단백분해효소 억제제들과 기질을 사용하여 단백분해효소의 활성을 측정한 결과 gelatin을 기질로한 전기영동과 azocasein을 기질로한 활성부위 잔기 억제실험에서 N. fowleri와 N. gruberi의 단백분해효소는 서로 상이하였다. N. fowleri의 용해물과 배양액을 이용하여 CHO 세포에 대한 세포독성을 관찰한 결과 그 용해물과 배양액 모두 단백질 농도의 증가에 따라 세포독성이 증가하였으나 N. gruberi의 용해물과 배양액에서 세포독성이 거의 없었다. N. fowleri의 용해물과 배양액의 단백분해효소는 Rf값을 서로 달리하는 효소분획으로 되어있음이 gelatin을 기질로한 SDS-PAGE에서 관찰되었으며, Rf 0.87의 단백분해효소 분획을 용액물과 배양액에서 공통적으로 갖고 있었다. 이 N. fowleri 배양액의 정제는 초원심분리, ammonium sulfate 분별침전, Spectra/Gel AcA 44 column chromatography를 이용하여 배양액내 단백분해효소가 23.6배로 정제되었고, 회수율은 19.9%였다. Gelatin을 기질로한 SDS-PAGE를 한 결과 Rf값이 0.87이었으며, 분자량을 SDS-PAGe로 측정한 결과 17,500 dalton이었다. N.fowleri의 배양액에서 부분정제한 분획은 여러 단백분해효소 활성부위 잔기 억제제들과 기질을 사용하여 단백분해효소 활성이 측정되었고, 단백분해효소의 활성을 억제하였던 leupeptin, antipain, pepstatin에 의해 세포독성도 억제되었다. 이로써 cysteine과 aspartic acid의 잔기가 활성에 관여함을 알 수 있었다. 부분정제한 배양액내 효소분획을 CHO 세포에 처리한 후 주사 전자현미경으로 관찰한 결과 처리 30분후 표적세포의 융모막(microvilli)이 점차 감소하였고 세포막에 수포와 구멍이 형성되었으며, 3시간 후에는 세포막이 일부가 파괴되었다. 실험성적을 종합하면 N. fowleri 배양액에서 단백분해효소의 활성이 측정되었고, 세포독성을 나타내는 물질을 부분정제 할 수 있었다. It is suspected that the proteinases of parasites may be closely related to their pathogenicity and cytotoxicity with regard to the host-parasite relationship. The aim of the present study was to elucidate whether proteinases from the lysates or secretory materials of pathogenic Naegleria fowleri and nonpathogenic N. gruberi are related to their cytotoxic activities. Mice inoculated with 1×105 N.fowleri trophozoites showed a cumulative mortality rate of 75% during observation, while no mice died in a group inoculated with the same amount of trophozoites of nonpathogenic N. gruberi. Utilizing a Chinese hamster ovary (CHO) cell line incubated with radioactive 51Cr (51Cr-labeled CHO cells), cytotoxicities of free-living amoebae were assayed. Scanning electron microscopic observations of target CHO cells affected with a purified cytotoxic substance of N. fowleri were also carried out. To determine the types and electrophoretic banding patterns of proteinases, the effect of various proteinase inhibitors on cell activity was examined, and it was found that enzymes from N. fowleri showed different patterns than N. gruberi. Cytotoxic activities, as determined by the release of 51Cr from CHO cells, were significantly detected in the lysate and cultured media of N. fowleri, but very little cytotoxic activity was observed in the lysate and cultured media of N. gruberi. The rate of flow(Rf) values for the electrophoretic banding patterns varied between the proteinase of the lysate and the cultured media of N. fowleri, but a Rf value of 0.87 was common in both bands. Purification of proteinase in the cultured media of N. fowleri was performed by centrifugation, ammonium sulfate fractionation, and spectra/Gel AcA 44 column chromatography. Purification was achieved up to a 23.6-fold increase in comparison with the raw materials of the intact N. fowleri cultured media and yielded about 19.9% of the total amount. The partially purified cytotoxic substance determined by SDS-PAGE had a molecular weight of 17,500 daltons and an Rf value of 0.87 was determined by the gelatin containing SDS-PAGE. Enzymatic activities in the N. fowleri cultured media were examined using various proteinase inhibitors and substrates, and it was confirmed that cysteine and aspartic acid residues were responsible for the cytotoxic activity. The scanning electron microscopic findings of the CHO cells treated with a partially purified cytotoxic substance of N. fowleri and observed for 30 minutes up to 180 minutes, showed deterioration of the microvilli over the entire surface of the cell, plasma membrane blebbing and holes in the plasma membrane. From the above findings of the biochemical and physical properties of the cytotoxic substance from N. fowleri cultured media, it is suggested that the cysteine proteinase secreted from N. fowleri may damage the cell membrane of target cells.

        Contemplation of Risk Management for Internet Banking System

        Next Generation internet banking service for important role of banking business competitive power separates service function of individual banking and should equip various service that coincides in each special qualities these are customer management, portfolio management, global corporation support etc. The past few years have been characterized by rapid changes in technology and the introduction of corporate and retail banking services through the Internet. the integration of e-banking platforms with legacy systems and the increasing dependence of banks on third party information service providers, all dramatically amplify the magnitude of risks to which banks are exposed. Risk management disciplines have not evolved at the same speed and many institutions, especially the smaller ones, have not been able to incorporate Internet banking risk controls within their existing risk management structures. This article provides an overview of the various risks which are heightened with Internet banking, and a holistic approach to managing these risks. In addition, the presentation will explain the different risk areas and the controls to be adopted to mitigate these risks.

      • Biochemical Studies on Proteinase in Acanthamoeba culbertsoni

        병원성 자유생활아메바 A. culbertsoni의 세포내 단백분해효소와 세포외 단백분해효소의 생화학적 특서을 규명하고자 하였다. A. culbertsoni를 초음파를 이용하여 파쇄한 후 원심분리하여 상청액을 세포내 단백분해 효소원으로 사용하였으며, log phase까지 배양한 배지를 원심분리하고 상청액을 취하여 filter로 거른 후 세포외 단백분해 효소원으로 사용하였다. 단백분해효소의 적정 pH를 알아보고자 azocasein을 기질로 사용하여 pH 4.0에서 pH 8.5까지 변화시켜주면서 활성을 측정하여 본 결과 세포내 단백분해효소와 세포외 단백분해효소의 적정 pH는 각각 pH 5.5와 pH 6.0이었다. 활성에 필수적인 아미노산 잔기를 확인하고자 DTNB, NEM, PMSF, NAI, PLP, TNBS, EDTA, DEP, phenlyglyoxal를 처리한 결과, 세포와 단백분해효소는 serine이 활성에 관여할 것으로 생각되며 세포내 단백분해효소는 serine과 arginine이 활성에 관여할 것으로 사료되었다. 또한, gelation를 기질로 하는 1% SDS가 포함된 PAGE를 이용해서 세포내 단백분해효소와 세포외 단백분해효소가 각각 Rf값을 달리하는 5개와 2개의 효소 분획으로 되어있음을 확인하였다. 한편, PMSF를 처리하여 준 결과 분획된 효소의 활성이 감소함을 전기영동을 이용하여 관찰할 수 있었다. Biochemical properties of the proteinase in Acanthamoeba culbertsoni were examined. Intracelluar and extracellular proteinase activity in Acanthamoeba culbertsoni were maximal in the condition of pH 5.5, respectively. To investigate their essential amino acids for catalysis, chemical modification was carried out by using various inhibitors. Phenylmethylsufonylfluoride (PMSF), hydroxyl group directed reagent, inhibited the intracellular and extracellular proteinase, and phenyloglyoxal, arginine directed reagent, also inhibited the intracellular proteinase. The rate of inactivation by PMSF and phenylglyozal were proportional to the concentration of the modification reagents and incubation time, suggesting that the essential amino acids of intracellular proteinase activity were serine and arginine, and the essential amino acid of extracellular proteinase activity was serine. A highly sensitive electrophoretic method involving gelation-containing polyacrylamide gels was used to analyze the proteinase. Multiple proteinase forms in the extracts of A. culbertsoni trophozoites and culture medium were found by this method. Intracellular and extracellular proteinase had at least five and two bands of activity of electrophoresis, respectively. All of the enzymes had characteristics of serine proteinase.
