추출방법에 따른 감귤 과피 유래 Flavonoid의 추출효율 및 항산화 효과에 대한 비교

본 연구에서는 감귤 과피 가공부산물로부터 기능성 polyphenol 및 flavonoid가 새로운 추출방법인 아임계 추출법을 통해 추출되었고, 열수(80oC), 에탄올, 설탕용액을 이용한 기존 추출법과의 추출효율이 비교 분석되었다. 건조된 감귤 과피로부터 총 polyphenol(27.25±1.33mg QE/g DCP) 및 flavonoid(7.31±0.41mg QE/g DCP)에 대한 최대수율이 아임계 추출법(190°C, 1300 psi, 10 min)을 통해 획득되었으며, 이것은 기존 추출법 가운데 가장 높은 수율을 보인 에탄올 추출법을 통한 총 polyphenol(3.79±0.73mg QE/g DCP) 및 flavonoid(0.86±0.27mg QE/g DCP) 수율 대비 7.2배와 8.5배 이상 더 높은 것이었다. 추출방법에 따른 감귤 과피 추출물의 항산화 활성은 큰 차이를 나타내지 않았으나, 이러한 결과는 건조 감귤 과피 1g 당 아임계 추출법(190oC, 1300 psi, 10 min)에 의한 항산화 성분의 상대 수율(relative yield, %)이 다른 추출방법을 통해 획득된 것보다 대략 9.5배 이상 더 높다는 것을 설명하는 결과로, 아임계 추출법이 기존의 전통적 추출법에 비하여 감귤 과피의 기능성 polyphenol 및 flavonoid를 추출하는데 매우 적합한 방법임을 제시하고 있다. The extraction of polyphenol and flavonoid from citrus peel was performed by the ethanol, sugar, hot water (80oC), and subcritical water extraction methods. The maximum yields of total polyphenolic compounds (27.25±1.33mg QE/g DCP, QE and DCP indicate quercetin equivalent and dried citrus peel, respectively) and flavonoids (7.31±0.41mg QE/g DCP) were obtained by subcritical water extraction (SWE) with operating conditions of 190oC, 1300 psi, and 10 min. The yields by SWE were over 7.2-, and 8.5-fold higher than those of total polyphenols (3.79±0.73mg QE/g DCP) and flavonoids (0.86±0.27mg QE/g DCP) obtained using the ethanol extraction, which showed the highest extraction efficiency among tested conventional methods, respectively. Antioxidant activities of extracts obtained by different methods showed no significant differences. However, the relative antioxidant yield per 1 g dried citrus peel by SWE (190oC, 10 min) was over 9.5-fold higher than that by the ethanol extraction.

To analyze and construct multi-echelon inventory system for the Supply Chain, we apply the information technology such as IDEF and UML rather than the optimization theory. These information approach method are appropriate for the Supply Chain.

This paper presents vibration control performances of a commercial magnetorheological(MR) suspension via new control strategy considering hysteresis of the field-dependent damping force of MR damper. A commercial MR damper which is applicable to high class passenger vehicle is adopted and its field-dependent damping force is experimentally evaluated. Preisach hysteresis model for the MR damper is identified using experimental first order descending(FOD) curves. Then, a feed-forward compensation strategy for the MR damper is formulated and integrated with a linear quadratic regulation(LQR) feedback controller for the suspension system. Control performances of the proposed control strategy for the MR suspension is experimentally evaluated with quarter vehicle test facility.
Detected Range of the Deicer around the Highway

The composition of the deicer sprayed on the highway is spreading over the highway by the scattering or snow removal activity, or car movement and consequently affecting the vegetation environment around the highway. These are the cause of the damage of fruit trees and crops, and also the cause of corrosion of highway structures. The goal of this study is to estimate the detected range of deicer components from a highway. The concentration of the deicer components in gauze and soil were investigated according to the crossing distance from the highway. The data collected were then used to estimate the concentration range of deicer components in a rage of distance up to 100 m from the highway where the deicer was spread. The sample time and weight of gauze were measured before and after installation, and the soil was collected at more than three points in parallel with the highway at the gauze installation point. The components in gauze and soil were investigated in addition to the deicer components (Ca2+, Na+, Cl-) as well as Mg2+ and K+. As Ca2+ and Cl- components of deicer were affected by agricultural use, Na+ component was selected as a tracer and further SAR (Sodium Adsorption Ratio) of soil was analyzed to examine the degree of influence on vegetation indirectly. The gauze concentration was evaluated by the concentration of the deicer ingredient at the background concentration of the blank gauze. The total amount of the deicer sprayed in the study road for 4 months (winter season) was about 93 ton/km. In the gauze test, the spread of the deicer was detected at a distance of 100 m in study area, but the concentration of the deicer in the gauze by distance decreased rapidly within 10 m from the highway. And concentration of the deicer components in gauze and soil came down after rainy season (August ~ Sep.). The results showed that the components of the deicer could be spread widely by the wind. The effective range of the deicer on vegetation based on SAR in soil was estimated to be less than 20 m from the highway. This study examines the concentration changes of the deicer components in gauze and soils and shows that deicer components sprayed on the highway are accumulated and moved over time by wind, snow removal, terrain, water system and land use around the highway.

<P>Inflammation is a basic pathological mechanism leading to a variety of vascular diseases. The inflammatory reaction involves complex interactions between both circulating and resident leukocytes and the vascular endothelium. In this study, we report evidence for a novel action of TNF-related activation-induced cytokine (TRANCE) as an inflammatory mediator and its underlying signaling mechanism in the vascular wall. TRANCE significantly increased endothelial-leukocyte cell interactions, and this effect was associated with increased expression of the cell adhesion molecules, ICAM-1 and VCAM-1, on the endothelial cells. RT-PCR analysis and promoter assays revealed that expression of these cell adhesion molecules was transcriptionally regulated mainly by activation of the inflammatory transcription factor, NF-kappaB. TRANCE induced IkappaB-alpha phosphorylation and NF-kappaB activation via a cascade of reactions involving the TNFR-associated factors, phospholipase C, PI3K, and protein kinase C (PKC-alpha and PKC-zeta). It also led to the production of reactive oxygen species via PKC- and PI3K-dependent activation of NADPH oxidase in the endothelial cells, and antioxidants suppressed the responses to TRANCE. These results demonstrate that TRANCE has an inflammatory action and may play a role in the pathogenesis of inflammation-related diseases.</P>

<P>Vascular endothelial growth factor (VEGF) and hepatocyte growth factor (HGF) are potent angiogenic factors that have been used clinically to induce angiogenesis. However, concerns have been raised about VEGF because of its proinflammatory actions, which include enhancing the adhesion of leukocytes to endothelial cells. We have examined the possible antiinflammatory effects of HGF on the vasculature. HGF, unlike VEGF, did not alter leukocyte adhesion to endothelial cells. Instead it inhibited VEGF-induced leukocyte-endothelial cell interactions and the endothelial expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1). In a skin inflammation model, VEGF-treated mice showed a significant increase of leukocytes infiltrated or adherent to the luminal surface of blood vessels, as compared with vehicle- or HGF-treated mice. The VEGF effect was markedly suppressed by coadministration of HGF. RT-PCR and promoter analysis revealed that HGF downregulated VEGF-mediated expression of ICAM-1 and VCAM-1 at the transcriptional level. Furthermore, these inhibitory effects coincided with suppression of IkappaB kinase activity, and this in turn prevented the activation of the inflammatory transcription factor NF-kappaB. Taken together, our results demonstrate that HGF suppresses VEGF-induced inflammation presumably by inhibiting the endothelial NF-kappaB pathway. This suggests that combined treatment with HGF and VEGF could be superior to treatment with either factor alone for enhancing therapeutic angiogenesis while avoiding inflammation.</P>
