<P>RNA-guided genome surgery using CRISPR-Cas9 nucleases has shown promise for the treatment of diverse genetic diseases. Yet, the potential of such nucleases for therapeutic applications in nongenetic diseases is largely unexplored. Here, we focus on age-related macular degeneration (AMD), a leading cause of blindness in adults, which is associated with retinal overexpression of, rather than mutations in, the VEGFA gene. Subretinal injection of preassembled, Vegfa gene-specific Cas9 ribonucleoproteins (RNPs) into the adult mouse eye gave rise to mutagenesis at the target site in the retinal pigment epithelium. Furthermore, Cas9 RNPs effectively reduced the area of laser-induced choroidal neovascularization (CNV) in a mouse model of AMD. Genome-wide profiling of Cas9 off-target effects via Digenome-seq showed that off-target mutations were rarely induced in the human genome. Because Cas9 RNPs can function immediately after in vivo delivery and are rapidly degraded by endogenous proteases, their activities are unlikely to be hampered by antibody-and cell-mediated adaptive immune systems. Our results demonstrate that in vivo genome editing with Cas9 RNPs has the potential for the local treatment for nongenetic degenerative diseases, expanding the scope of RNA-guided genome surgery to a new dimension.</P>
Enhanced optical response of hybridized VO2/graphene films.

<P>Application of graphene as transparent electrodes is an active research area due to its excellent electrical and optical properties. Vanadium dioxide (VO2) is an attractive material since it is a thermochromic material that undergoes a structural phase transition when heat is applied. The phase transition results in the change of electrical and optical characteristics. We report optical characteristics of hybrid materials of graphene and VO2. We observed a 12% improvement in infrared transmittance with VO2 films deposited on graphene sapphire substrates compared to that of bare sapphire substrates. We also found that the phase transition temperature decreases as the number of graphene layers on the substrates increases. In the case of VO2 films on the substrate that was coated with four layers of graphene, the mean phase transition temperature was lowered to 56 C.</P>

<P>We report that poly(3,4-ethylenedioxythiophene) derived from poly(ionic liquid) (PEDOT:PIL) constitutes a unique polymeric hole-injecting material capable of improving device lifetime in organic light-emitting diodes (OLEDs). Imidazolium-based poly(ionic liquid)s were engineered to impart non-acidic and non-aqueous properties to PEDOT without compromising any other properties of PEDOT. A fluorescent OLED was fabricated using PEDOT:PIL as a hole-injection layer and subjected to a performance evaluation test. In comparison with a control device using a conventional PEDOT-based material, the device with PEDOT:PIL was found to achieve a significant improvement in terms of device lifetime. <P>A new composite membrane that consists of sulfonated poly(arylene ether sulfone) (SPES) and a highly functionalized inorganic filler of sulfated zirconia (s-ZrO<SUB>2</SUB>) was synthesized and evaluated for enhancement of water retention as well as proton conductivity at high temperatures above 120 °C in the absence of water. Since the SPES has less hydrophobic/hydrophilic difference between the polymer backbone and –SO<SUB>3</SUB>H ionic groups, the proton channels are relatively small and not easily distributed, which results in no significant change at highly saturated conditions. Therefore, the effective interconnection between the –SO<SUB>3</SUB>H ionic groups and bound water adsorbed on the surface of the s-ZrO<SUB>2</SUB> is the key in achieving a higher proton transfer for PEM fuel cells at high temperatures above 120 °C. Analysis by small angle X-ray scattering (SAXS) of recast membranes showed that SPES random copolymers with various s-ZrO<SUB>2</SUB> contents have a similar ionic cluster size and less pronounced phase separation regardless of the dry/swollen condition of the membrane. However, the phase images obtained by atomic force microscopy (AFM) showed that the connectivity of ionic groups varies, depending on the content of s-ZrO<SUB>2</SUB> in the polymer matrix. Larger and more interconnected hydrophilic regions were found with the SPES membrane when there was no significant particle aggregation in the hydrophilic moieties of the polymer matrix. OBJECTIVES: As few mpox cases have been reported in Korea, we aimed to identify the characteristics of mpox infection by describing our epidemiologic investigation of a woman patient (index patient, the third case in Korea) and a physician who was infected by a needlestick injury (the fourth case). METHODS: We conducted contact tracing and exposure risk evaluation through interviews with these 2 patients and their physicians and contacts, as well as field investigations at each facility visited by the patients during their symptomatic periods. We then classified contacts into 3 levels according to their exposure risk and managed them to minimize further transmission by recommending quarantine and vaccination for post-exposure prophylaxis and monitoring their symptoms. RESULTS: The index patient had sexual contact with a man foreigner during a trip to Dubai, which was considered the probable route of transmission. In total, 27 healthcare-associated contacts across 7 healthcare facilities and 9 community contacts were identified. These contacts were classified into high (7 contacts), medium (9 contacts), and low (20 contacts) exposure risk groups. One high-risk contact was identified as a secondary patient: a physician who was injured while collecting specimens from the index patient. CONCLUSIONS: The index patient visited several medical facilities due to progressive symptoms prior to isolation. Although the 2022 mpox epidemic mainly affected young men, especially men who have sex with men, physicians should also consider mpox transmission in the general population for the timely detection of mpox-infected patients.

<P>In the electronic components and device packaging process, the Cu conductor surface is generally coated with Au and Ni. The Au coating is applied to prevent the oxidation of the Cu surface and enhance the solderability, while the Ni coating is applied as a diffusion barrier between the solder alloy and the Cu substrate, in order to restrict the formation and growth of intermetallic compounds. The dynamic reactive wetting characteristics of Sn-Ag-Cu alloys are related to the properties of the coating materials. In the present study, the dynamic reactive wetting behavior of Sn-Ag-Cu alloys on Cu substrates coated with Ni and Au was firstly investigated on millisecond scale. On the bare Cu surface, the metal droplet rebounded several times due to the poor wettability and started to spread from 1 s and became equilibrated by 20 s, whereas on the Ni/Au coated surface, the metal droplet settled down without rebounding because of the good wettability, which caused it to react instantaneously, and became equilibrated by 1 s.</P>

<P>Herein, we present a fluorescent-peptide drug delivery system composed of biotin–naphthalimide–HJ inhibitor peptide2, prodrug 1. <P>The aldimine-directed C-H amidation of various arenes with N-acyl azides as amidation surrogates under cationic iridium(III) catalysis is described. This transformation efficiently provides a range of 2-aminobenzaldehyde derivatives with excellent site selectivity and functional group compatibility. The resulting 2-aminobenzaldehyde framework provides facile access to a range of biologically interesting heterocycles. In addition, all synthetic compounds were screened for anti-inflammatory activity against interleukin-1 beta (IL-1 beta) and tumor necrosis factor alpha (TNF-alpha) with lipopolysaccharide (LPS)-induced RAW264.7 cells. Generally, a range of ortho-amidated benzaldehydes displayed promising inhibitory activity against IL-1 beta and TNF-alpha compared to dexamethasone as a positive control. Notably, compounds (3ae and 4ac) were found to exhibit potent anti-inflammatory activity stronger than that of dexamethasone.</P>
