        Saponins and Other Constituents from the Leaves of <i>Aralia elata</i>

        <P>A new triterpenoid saponin, together with five known saponins, were isolated from the nonpolar <I>n</I>-hexane fraction of the leaves of <I>Aralia elata</I>. The structure of the new saponin, durupcoside C, was elucidated as hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-arabinopyranoside on the basis of spectroscopic analysis. The known saponins were characterized as 3-<I>O</I>-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranosyl hederagenin 28-<I>O</I>-β-<SMALL>D</SMALL>-xylopyranosyl(1→6)-β-<SMALL>D</SMALL>-glucopyranosyl ester, hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside, oleanolic acid 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside, hederagenin 3-<I>O</I>-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside (α-hederin), and hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-arabinopyranoside (collinsonidin). In addition, two known lipids, Arisaema glyceride 3 and ceramide mixtures were also isolated and characterized. Collinsonidin and two known lipids were isolated for the first time from this plant.</P>

        <P>Chronic inflammatory airway diseases including asthma are characterized by immune dysfunction to inhaled allergens. Our previous studies demonstrated that T cell priming to inhaled allergens requires LPS, which is ubiquitously present in household dust allergens. In this study, we evaluated the role of vascular endothelial growth factor (VEGF) in the development of T cell priming and its polarization to Th1 or Th17 cells when exposed to LPS-contaminated allergens. An asthma mouse model was induced by airway sensitization with LPS-contaminated allergens and then challenged with allergens alone. Therapeutic intervention was performed during allergen sensitization. The present study showed that lung inflammation induced by sensitization with LPS-contaminated allergens was decreased in mice with homozygous disruption of the IL-17 gene; in addition, allergen-specific Th17 immune response was abolished in IL-6 knockout mice. Meanwhile, in vivo production of VEGF was up-regulated by airway exposure of LPS. In addition, airway sensitization of allergen plus recombinant VEGF induced both type 1 and type 17 Th cell (Th1 and Th17) responses. Th1 and Th17 responses induced by airway sensitization with LPS-contaminated allergens were blocked by treatment with a pan-VEGF receptor (VEGFR; VEGFR-1 plus VEGFR-2) inhibitor during sensitization. These effects were accompanied by inhibition of the production of Th1 and Th17 polarizing cytokines, IL-12p70 and IL-6, respectively. These findings indicate that VEGF produced by LPS plays a key role in activation of naive T cells and subsequent polarization to Th1 and Th17 cells.</P>

      • Conflict cause analysis between stakeholders in a utility-scale PV plant and its policy improvement methods in Korea

        <P><B>Abstract</B></P> <P>Despite the technological advances and the Korean government's ongoing efforts to expand the PV market, disputes related to the quality and performance of the utility-scale PV systems are not uncommon. Therefore, actual dispute cases in Korea are analysed to elicit their root causes and suggest effective remedies in this paper. This analysis focuses on the following three types of disputes caused by: (i) PV system installation quality, (ii) PV system performance degradation and (iii) natural disaster damage. Based on interviews and examination of the PV plants in dispute, it was drawn that conflicts between the stakeholders stem from limitations to utility-scale PV plants’ evaluation and technical dispute management, lack of PV module reliability and lax contract terms for procurement. To mitigate the conflicts between domestic PV stakeholders, the following four policy improvements are suggested: improvements of PV plants’ deployment procedure and quality assurance system that can detect failures of equipment in early stages, introduction of a government-led dispute settlement system, enforcement of strict quality standards for PV modules and provision of information on PV procurement specifications. The lessons and suggested remedies will help policy makers understand the domestic PV market and redesign the policies for mitigation of potential disputes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Disputes pertaining to utility-scale PV system failures are not uncommon in Korea. </LI> <LI> Case studies show how disputes among stakeholders evolve in Korean PV market. </LI> <LI> Some PV dispute cases originate from insufficient national quality assurance system. </LI> <LI> This study suggests the dispute mitigation remedies for PV policy makers. </LI> </UL> </P>

        Sequence variation and the transcriptional activity of the upstream regulatory region in human papillomavirus 16 E7 variants in cervical cancer of Korean women.

        <P>In this study, we investigated the sequence variation and different transcriptional activities of the upstream regulatory region (URR) in HPV 16 E7 variants in cervical cancer tissue from Korean women. Using PCR-directed sequencing, the presence of sequence variations in URRs were analyzed and the sites of sequence variation were matched with the known transcriptional factor binding site (TFBS) in 26 HPV 16 E7 variants, 21 cases with A647G (KE7-1, high oncogenic potential) and 5 cases with T732C (KE7-2, low oncogenic potential). In addition, we determined the transcriptional activity of URR in a HPV 16 prototype, and in 4 cases of HPV 16 E7 variants, by using the functional chloramphenicol acetyl transferase (CAT) assay. The 26 HPV 16 E7 variants showed more than 11 sites of sequence variation in the URR. Ten sites of sequence variation were located in the known TFBS and the distribution of sequence variations in the URR showed clear differences between KE7-1 and KE7-2. The sequence variations T7781C and C7786T were matched with YY1 binding sites, G7193T and C7689A were matched with TEF1 binding sites, and C7394T and C7395T were matched with GRE binding sites. The other sequence variations, which were matched with the TFBS, were A7485C, G7489A, T7743G and G7842A. The URR activity of KE7-1 was significantly lower than that of the HPV 16 prototype, whilst KE7-2 was similar. Taken together with the results of the transcriptional activities of KE7-1 and KE7-2, our results suggest that the functional activity and sequence variations of HPV 16 URR may not be related to the oncogenic potential of HPV 16 E7 variants.</P>

      • Highly aligned and porous reduced graphene oxide structures and their application for stretchable conductors

        <P><B>Abstract</B></P> <P>Developing materials that can absorb large amounts of strain without fracture or significant degradation in their electronic properties is essential for the realization of stretchable electronics. Here, stretchable conductors made from reduced graphene oxide/polydimethylsiloxane (r-GO/PDMS) composite were fabricated to achieve stable resistance changes under uniaxial stretching. By controlling the weight fraction of GO in water, highly aligned and porous GO structure was obtained over 0.5wt% of GO solution. After thermal and chemical reduction of GO to r-GO film, PDMS was infiltrated into pores of the film to make free-standing and elastic composites. Electrical percolation of r-GO in the composite was achieved at the r-GO concentration over 0.3wt% along with improvement of electrical conductivity. However, highly aligned r-GO/PDMS composite only exhibited notable suppression of resistance change under the large deformation of 100% strain. The resistance change of 0.3wt% r-GO/PDMS composites at 100% strain was over 7 times higher than that of 0.7wt% r-GO/PDMS composites, exhibiting the dramatic effect of r-GO alignments. Even after repeated stretching/releasing cycles, the variation in resistance from 0 to 70% of strain was stable. The stretchable conductor based on the aligned r-GO/PDMS composite was successfully applied as a resistive strain sensor.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Ciglitizone inhibits cell proliferation in human uterine leiomyoma via activation of store-operated Ca2+ channels.

        <P>This study investigated the acute effects of a peroxisome proliferator-activated receptor (PPAR)-gamma ligand, ciglitizone, on cell proliferation and intracellular Ca2+ signaling in human normal myometrium and uterine leiomyoma. Changes in intracellular Ca2+ concentration ([Ca2+]i) were measured with fura-2 AM, and cellular viabilities were determined by viable cell count and 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide reduction assay. Ciglitizone (100 microM) induced greater inhibition of cell proliferation in uterine leiomyoma than in myometrium. Ciglitizone also dose-dependently increased [Ca2+]i in both myometrium and uterine leiomyoma; these [Ca2+]i increases were inhibited by PPAR-gamma antagonists and raloxifene. Ciglitizone-induced [Ca2+]i increase showed only an initial peak in normal myometrial cells, whereas in uterine leiomyoma there was a second sustained [Ca2+]i increase as well. The initial [Ca2+]i increase in both myometrium and uterine leiomyoma resulted from the release of Ca2+ by the sarcoplasmic reticulum via activation of ryanodine receptors. The second [Ca2+]i increase was observed only in uterine leiomyoma because of a Ca2+ influx via an activation of store-operated Ca2+ channels (SOCCs). Cell proliferation was inhibited and secondary [Ca2+]i increase in uterine leiomyoma was attenuated by cotreatment of ciglitizone with a SOCC blocker, lanthanum. The results suggest that ciglitizone inhibits cell proliferation and increases [Ca2+]i through the activation of SOCCs, especially in human uterine leiomyoma.</P>

      • Efficacy and Safety of FOLFIRI Regimen in Elderly Versus Nonelderly Patients with Metastatic Colorectal or Gastric Cancer

        <P>Background. Irinotecan-based chemotherapy is a standard backbone of therapy in patients with metastatic colorectal cancer (CRC) or gastric cancer (GC). However, there is still a paucity of information concerning the efficacy and safety of irinotecanbased regimens in elderly patients. Patients and Methods. Using the patient cohort (n=1,545) from the UGT1A1 genotype study, we compared the efficacy and safety between elderly and nonelderly patients with metastatic CRC (n=934) or GC (n=611) who received first-or second-line FOLFIRI (irinotecan, leucovorin, and 5-fluorouracil) chemotherapy. Results. Despite lower relative dose intensity in elderly patients, progression-free survival and overall survival were similar between elderly (age >= 70 years) and nonelderly (<70 years) patients in the CRC cohort (hazard ratio [HR], 1.117; 95% confidence interval [CI], 0.927-1.345; p=.244, and HR, 0.989; 95% CI, 0.774-1.264; p=.931, respectively) and the GC cohort (HR, 1.093; 95% CI, 0.854-1.400; p=.479, and HR, 1.188; 95% CI, 0.891-1.585; p=.241, respectively). In both cohorts, febrile neutropenia (22.1% vs. 14.6% in CRC cohort and 35.2% vs. 22.5% in GC cohort) and asthenia (grade 3: 8.4% vs. 1.7% in CRC cohort and 5.5% vs. 2.9% in GC cohort) were more frequent in elderly patients. In the CRC cohort, mucositis and anorexia were more frequent in elderly patients. In the GC cohort, nausea and vomiting were less frequent in elderly patients. Conclusion. The efficacy of the FOLFIRI regimen was similar between elderly and nonelderly patients in both the CRC and the GC cohorts. However, special attention should be paid to elderly patients because of increased risk for febrile neutropenia and asthenia.</P>

Neurochem.</I> (2012) <B>121</B>, 751–762.</P><P><B>Abstract</B></P><P>Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by a progressive loss of memory and cognition. One of the hallmarks of AD is the accumulation of beta‐amyloid (Aβ). Although endoplasmic reticulum stress, mitochondrial dysfunction, and oxidative stress have been implicated in Aβ toxicity, the molecular mechanism(s) of Aβ‐induced neurotoxicity are not fully understood. In this study, we present evidence that the glia‐derived stress protein metallothionein (MT) attenuates Aβ‐induced neurotoxicity by unique mechanisms. MT expression was increased in brain astrocytes of a NSE‐APPsw transgenic mouse model of AD. Astrocyte‐derived MT protected N2a neuroblastoma cells and primary cortical neurons against Aβ toxicity with concurrent reduction of reactive oxygen species levels. MT reversed Aβ‐induced down‐regulation of Bcl‐2 and survival signaling in neuroblastoma cells. Moreover, MT inhibited Aβ‐induced proinflammatory cytokine production from microglia. The neurotoxicity of Aβ‐stimulated microglia was significantly attenuated by MT‐I. The results indicate that MT released from reactive astrocytes may antagonize Aβ neurotoxicity by direct inhibition of Aβ neurotoxicity and indirect suppression of neurotoxic microglial activation. These findings broaden the understanding of neurotoxic mechanisms of Aβ and the crosstalk between Aβ and MT in AD.</P>

        Role of Interface Boundaries in the Deformation Behavior of TiAl Polysynthetically Twinned Crystal: In situ Transmission Electron Microscopy Deformation Study

        <P>To understand the role of boundaries in the deformation behavior of TiAl, in situ straining experiments in transmission electron microscopy have been performed on thin foils of polysynthetically twinned (PST) crystal of Ti-49.3 at.% Al. The deformation behavior of PST TiAl is anisotropic, depending on the angle between the lamellar boundaries and the straining axes. For L-orientation, deformation twins and ordinary dislocations transmit across the true-twin (TT) boundaries but are reflected at the pseudo-twin (PT) and rotational order-fault (RO) boundaries. For transverse (T) orientation, deformation twins are transmitted across all TT, PT, and RO boundaries. For I-orientation, shear deformation occurs parallel to the lamellar boundaries. There is a transmission of deformation across the interphase (IP) boundary in longitudinal orientation, but deformation is blocked and reflected at the IP boundary in T-orientation. The role of the various types of boundaries in localized deformation behavior was evaluated by considering Schmid factors and geometric compatibility factors.</P>
