Development and Evaluation of QTL-NIL for Grain Weight from an Interspecific Cross in Rice

        Yeo Tae Yun,Dong Min Kim,In Kyu Park,Chong Tae Chung,Yeaul Kyu Seong,Sang Nag Ahn 한국육종학회 2010 한국육종학회지 Vol.42 No.4

In a previous study, we mapped 12 QTLs for 1,000 grain weight (TGW) in the 172 BC2F2 lines derived from a cross between Oryza sativa ssp. Japonica cv. Hwaseongbyeo and O. rufipogon. These QTLs explained 5.4 - 11.4% of the phenotypic variance for TGW. Marker-aided selection combined with backcrosses was employed to develop QTL-NILs for each QTL. BC2F2 lines with each target QTL were backcrossed to Hwaseongbyeo twice and then allowed to self to produce BC4F5 populations. SSR markers linked to TGW were employed to select QTL-NILs with the respective target QTL. Six QTL-NILs with the recurrent parent, Hwaseongbyeo were evaluated for nine traits for three years from 2007 and 2009. Differences were observed between each of the 6 QTL-NILs and Hwaseongbyeo in TGW. In addition to TGW, these QTL-NILs displayed differences in other agronomic traits possibly indicating a tight linkage of genes controlling these traits. The direction of the QTL for TGW in 6 QTL-NILs was consistent as in the BC2F2 lines from the same cross. Difference in TGW between each of the QTL-NILs and Hwaseongbyeo was associated with the difference in one or two grain shape traits; grain length, grain width, and grain thickness. SSR markers linked to the QTL for TGW will facilitate selection of the grain shape character in a breeding program to diversify grain shape and provide the foundation for map-based gene isolation. Also, the QTL-NILs developed in this report and the progenies from crosses between the QTL-NILs will be useful in clarifying epistatic interactions among QTLs for TGW.

Effects of Amino Acid-enriched Ruminally Protected Fatty Acids on Plasma Metabolites, Growth Performance and Carcass Characteristics of Hanwoo Steers

        Park, Byung-Ki,Choi, Nag-Jin,Kim, Hyeong-Cheol,Kim, Tae-Il,Cho, Young-Moo,Oh, Young-Kyoon,Im, Seok-Ki,Kim, Young-Jun,Chang, Jong-Soo,Hwang, In-Ho,Jang, Hyun-Yong,Kim, Jong-Bok,Kwon, Eung-Gi Asian Australasian Association of Animal Productio 2010 Animal Bioscience Vol.23 No.8

This study was conducted to determine the effects of amino acid-enriched ruminally protected fatty acid (AARPFA) on plasma fatty acids and amino acids, growth performance and carcass characteristics of Korean native steers (Hanwoo) by simultaneous supply of fatty acids and limiting amino acids (methionine and lysine). Eighteen finishing Hanwoo steers, 18 months of age and weighing an average of $459.0{\pm}38.9\;kg$, were used for studies of the metabolism of plasma fatty acids and amino acids during supplementation of AARPFA. Also, 45 finishing Hanwoo steers, 16 months of age and weighing an average of $408.6{\pm}26.5\;kg$, were used for growth performance and carcass characteristics during supplemention of AARPFA. There were three treatments which comprised a basal diet supplemented with AARPFA at 0 g (T1), 50 g (T2) or 100 g (T3), respectively. Concentrations of saturated, unsaturated and total fatty acids in plasma were increased in T3 compared with other treatments (p<0.05). Concentrations of methionine and lysine in plasma were linearly increased with increasing levels of AARPFA (p<0.01). Average daily gain, dry matter intake and feed conversion ratio were not different among the treatments. Marbling score measured by ultra-sound scanning was higher in T3 than in T1 at 24 months of age (p<0.05). Rib eye area, back fat thickness, yield index and yield grade score were similar across the treatments. Marbling score and quality grade score were higher in T3 compared with other treatments (p<0.01). Thus, plasma fatty acids, methionine and lysine metabolism were affected by supplementing with 100 g of AARPFA which also had positive effects on marbling score and meat quality grade of finishing Hanwoo steers.

A medium-maturing, disease resistant and good-quality rice variety Geumanbyeo

        Hung-goo Hwang,Yeon-gyu Kim,Ha-cheol Hong,Yong-hwan Choi,Man-kee Baek,Im-soo Choi,Young-chan Cho,Chang-in Yang,Hong-yeol Kim,Jong-doo Yea,Young-seop Shin,Kyung-ho Kang,Sang-nag Ahn 한국육종학회 2007 한국육종학회지 Vol.39 No.4

“Geumanbyeo” is a new japonica rice variety developed by the rice breeding team of National Institute of Crop Science, RDA in 2002, which was derived from a cross between SR11878-14-4-1 of good eating-quality and high yield potential, and Suweon345 of lodging tolerance and resistance to diseases. Geumanbyeo is a medium-maturing variety with heading date of August 12 on ordinary planting in the middle plain area. It has a semi-erect plant type with culm length of 82cm, erect pubescent leaf blade and slightly tough culm with good canopy architecture. Geumanbyeo has a little bit fewer tillers per hill, and more spikelets per panicle than Hwaseongbyeo. It is slightly less tolerant to the cold stress in terms of both heading delay and spikelet fertility than Hwaseongbyeo. This variety shows delayed leaf senescence and considerable tolerance to viviparous germination at ripening stage. It shows moderately resistant to leaf blast and K1 race of bacterial blight, but susceptible to virus diseases and insect pests. The milled rice of Geumanbyeo exhibits translucent, relatively clear non-glutinous endosperm and medium-short grain. It showed lower amylose content of 18.6% and similar gelatinization temperature and better palatability of cooked rice compared to Hwaseongbyeo. The milled rice yield of this variety is about 5.32 MT/ha at ordinary culture in local adaptability test for three years. Geumanbyeo would be highly adaptable to the central lowland and mid-southern mountainous areas in Korea. N/A Background : Clinical manifestations and complications of measles are more severe in adults than children. But, there has been few clinical and epidemiologic data about adult measles in Korea. Objective : We investigated clinical features, laboratory findings, disease courses and complications and discussed the reason why measles incidence has been increasing recently in young adults to find out the solutions to prevent measles. Method : From April 1999 to February 2001, we examined prodromal symptoms, skin rash, complications, routine CBC, and blood chemistry of 42 adults who were admitted in KangBuk SamSumg Hospital for measles and performed qualitative serum measles specific IgM and IgG antibody test using ELISA method. Results : 1. Age distribution of most adult measle patients was between 15 and 25 years old(78.6%). 2. Fever was found in all patients and cough, coryza, headache, myalgia, conjunctivitis, sore throat, nausea, diarrhea, and vomit were found in decreasing order of frequency. 3. Skin rashes were started from the face and spread to trunk, arms and then legs in 37 patients out of 42. Five patients had skin rash in the face and trunk at the same time, and one had hemorrhagic rash. Koplik`s spots were found in 26 patients (62%). Among them, 2 had Koplik`s spots befort the rash and 24 had Koplik`s spots with the rash simultaneously or thereafter. Koplik`s spots were disappeared as skin rash disappeared. Exfoliative dermatitis was found following the skin rash in one patient. 4. among 26 patients who performed qualitiative serum measles specific IgM and IgG antibody test using ELISA, the seropositivity of measles specific IgM was 70%, of measles specific IgG was 27%, and of both measles specific IgM and IgG was 12%. 5. Complications were like this; hepatitis in 23, cervical lympadenitis in 20, laryngitis and bronchitis in 7, otitis media in 4, pneumonia on chest X-ray in 3, hepatosplenomegaly on upper abdomen ultrasonography in 5. Two patients were in pregnancy of 12 and 16 weeks, and the former had spontaneous abortion. Pnemonia and otitis media were recovered within average 5 days after adminstrating antibiotics and antipyretics.(Korean J Dermatol 2003;4(10) : 1304~1310)

In this paper, the procedure and methodology of the engine room cooling performance analysis for a forklift truck are presented using a CFD(Computation Fluid Dynamics) technique. The mesh generation of the CFD modeling of the engine room was a time consumed and difficult work for a CFD engineer to obtain the reasonable analysis results. Special exclusive codes were investigated to reduce the simulation time and obtain accurate results for the engine room heat analysis. In addition, the well-defined boundary condition in the CFD model and the accurate experimental data of a fan and radiator were also important factors to obtain a good simulation result closed to measure value. To verify the proposed procedure and methodology, the radiator inlet coolant temperature was compared with the simulation and experiment results. It shows that the proposed simulation method and modeling techniques are effective one for the cooler system design of a forklift engine room.

      • QTL Analysis for Cold Tolerance Using Introgression Lines Derived from a Cross between Tongil-Type Rice and Weedy Rice

        In-Kyu Park,Chang-Sik Oh,Dong-Min Kim,Sang-Nag Ahn 한국작물학회 2010 한국작물학회 학술발표대회 논문집 Vol.2010 No.04

QTL analysis for cold tolerance-related traits was conducted using 75 introgression lines (IL) developed from a cross between a japonica weedy rice and Tongil-type rice. A molecular linkage map consisting of 136 SSR markers was constructed to identify QTLs associated with cold tolerance. 75 ILs and the parents were evaluated for three traits associated with cold tolerance: seedling height and SAPD values at the seedling stage. The plants were grown for 15 days in the low temperature condition (13/20℃ day/night) and the control condition (25/20℃ day/night) in the growth chamber. A total of six QTL were identified for two traits and phenotypic variance explained by each QTL ranged from 4.3% to 35.7%. Among two QTL for seedling height, one QTL, sh1 for seedling height was detected at both conditions. The other QTL on chromosome 6 was detected in the low temperature condition. Four QTL were identified for SPAD value and two were detected on chromosomes 2 and 5. At these loci, Milyang 23 alleles increased the SPAD value. The other two QTL on chromosomes 1 and 4 were detected at the low temperature plot. At these loci, Hapcheonaengmi 3 alleles increased the SPAD values. These results indicate that Hapcheonaengmi 3 alleles might increase tolerance to low temperature in the Milyang 23 background because SPAD value is positively correlated with chlorophyll content and N content in rice. The markers linked to low temperature tolerance at the seedling stage would be useful in selecting for lines with enhanced cold tolerance in a breeding program.
