      • 樹木園 造成地域內 地존作業을 위한 아까시나무 除去試驗

        This experiment was conducted at the Seoul National University Arboretum at Seodun-dong, Suweon, Kyeonggi-do, Korea (1) to find out methods killing Robinia pseudoacacia L. effectively and (2) to minimize ecological and aesthetic problems. Glycine and 2, 4-D were treated in several concentration following the saw-scratching and cutting of stems on July 22 and 29, 1985. The results were measured on October 17, 1885. The results obtained were as follows: 1. Ecological problems were decreased by these methods. 2. In case of herbicides treatment on section area, aesthetic problems were minimized, and amounts of herbicide consumption were five times as little as in case of spraying plants with herbicides. 3. The death rate amounted to 100% when the ratio of Glycine solution to water was 1:3, so that the concentration, 1:3 was most economical to kill the tree throughly. 4. The death rate amounted to 100% when the ratio of Glycine solution mixed with 2, 4-D to water was 1:4, therefore Glycine solution mixed with 2, 4-D was more effective than pure Glycine solution. Since 2, 4-D solution was less expensive than Glycine solution and only small amounts of 2, 4-D solution was added, it was most economical and effective method that Glycine solution mixed with 2, 4-D was diluted to the concentration, 1:4 and treated on the stem.

      • 水原樹木園地域內의 自然森林植生 調査結果

        樹木園 活成豫定地를 對象으로 土壤 水分含量과 植生을 調査하여 앞으로의 樹木園 造成에 필요한 情報를 얻음과 동시에 樹木園 造成에 따른 植生破壞後 原來植生에 關한 情報가 必要할 때 參考하기 爲하여 1985年 6月 12日에 本 調査를 實施하였는데 그 結果는 다음과 같다. 1. 全體的으로 겨이삭(Agrostis clavata var. nukabo)의 優占度가 매우 높았으며 씀바귀(Ixeris dentata)와 흰씀박귀(Ixeris dentata var. albiflora)는 극히 乾燥한 곳을 제외하고는 대부분 優占度가 높았다. 濕한 곳에는 왕비늘사초(Carex maximowiczii), 기장대풀(Isachne globosa) 等이, 극히 乾燥한 곳에서는 잔디(Zoysia japonica)가, 보통 乾燥한 곳에서는 그늘사초(Carex lanceolata), 김의털(Festuca ovina) 等이 優占度가 높았다. 2. 特히 稀貴植物인 선제비꽃(Viola raddeana)群落과 진퍼리용담(Grntiana scabra var. buergeri for stenophylla)이 Block 3(습지)에 있으므로 앞으로 이 區域은 注意깊게 保存되어야 할 것이다. 3. 土壤 水分含量이 最低 14.7%에서 最高 89.9%까지 分布하여 乾濕의 差異가 심하게 나타났다. 特히 調査 時期가 乾濕期인데도 土壤 水分含量이 70%가 넘는 곳이 3개소나 된다. 이 곳은 地下水位가 높아서 濕地이므로 濕地植物園을 造成해야 될 것이다. 植生의 造成狀態가 區域에 따라 變異가 심하게 나타났기 때문에 이 林地가 매우 不安定한 狀態에 있다고 볼 수 있을 것이다. Soil moisture contents and vegetation of the forest land planed to establish the Suweon Arboretum had been investigated to obtain some informations for establishing arboretum, and also for keeping the original vegetation composition data after the arboretum was established. The results obtained in this investigation could be summarized as follows. 1. In general, the coverage of Agrostis clavata var. nukabo was very high. The coverage of lxeris dentata and ixeris dentata var. albiflora were high except for very arid blocks. Coverages of Carex maximowiczii and Isachne globosa were high in wet blocks. The coverage of Zoysia japonica was high in very arid blocks and that of Carex lanceolata and Festuca ovina in arid blocks were high. 2. Soil moisture content was distributed from 14.7% to 88.9%. Especially the fact of the high moisture content (74.7, 77.5, 88.9%) in dry season implied that the ground water level of this area was high. The vegetation composition of this forest was varied, too. Therefore, it is considered that the stability of this forest was fragile.

      • Factors on Somatic Embryogenesis and Organogenesis of Soybean (Glycine max)

        To enhance in vitro plantlet regeneration efficiency of soybean through embryogenesis, the culture conditions such as meterial part and size of immature seed, 2,4-D pH and solidifying agents for somatic embryogenesis were investigated. Somatic embryogenesis was induced from the immature embryo, immature cotyledon, and embryonic axis explants of the immature seed on MS medium supplemented with 2.0 mg/L 2,4-D. The highest rate (up tp 23%) of somatic embryogenesis was obtained from the immature cotyledon, followed by embryonic axis and immature embryo. The rate varied with the developmental stages of seed. The maximum rate (25%) of embryogenesis was obtained from 3-4 mm length of the seed after 25 days of flowering. The optimum concentration of 2,4-D for embryogenesis was 10mg/l. The optimum pH was at 5.8 and solidifying agent for medium was better with 0.4% gelrite than with agar. For rapid multiplication of shoot tip from germinating somatic embryos, they were cultured on MS medium containing 2 mg/L IBA and 1 mg/L BAP, After then somatic embryos with one and three cotyledons were transferred to the plant growth regulator free medium. The medium exhibited the higher rate (50%) of development than the multiplication medium. To select the section with shoot formation ability, the calli and shoot formation from three sections of 5-days-seedings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section and the cotyledon section. The shoot formation rates and shoot number of the four excised sections of the first leaf were aldo investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and number (3-4/ explants) than those in other two explants. Effects of cytokinins on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0mg/L) exhibited the highest in shoot formation rate (94%) and combination (1: 1, v/v) of zeatin (1.0mg/L) and IAA (1.0 mg/L) eshibited the highest in shoot formation rate (96%) and numbers (16/explant), twice more than zeatin alone (1.0 mg/L). The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at variois concentrations. The highest root formation (8/shoot) was achieved on the medium supplementes with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.

        Influences of feeding frequency of extruded pellet and moist pellet on growth and body composition of juvenile Japanese flounder <i>Paralichthys olivaceus</i> in suboptimal water temperatures

        <P>ABSTRACT: </P><P>This study was carried out to investigate the influences of feeding frequency of extruded pellet and moist pellet on growth and body composition of juvenile flounder (initial mean weight 6.3 g) in suboptimal water temperatures. A 2 (diets: extruded pellet and moist pellet) × 2 (feeding frequencies: two and three times daily) × 2 (water temperatures: 12 and 17°C) factorial design with three replications was used. After 60 days of feeding, the feeding frequency did not significantly affect growth performance of fish. Weight gain, daily feed intake, feed efficiency and protein efficiency ratio were significantly (<I>P</I> < 0.05) higher for fish reared at the higher water temperature. At the same water temperature, weight gain, feed efficiency, and protein efficiency ratio were significantly (<I>P</I> < 0.05) higher for fish fed the extruded pellet than moist pellet. The results of this study indicate that feeding frequency of two times daily is sufficient for optimal growth of juvenile flounder when reared in suboptimal water temperatures, and an increase in water temperature from 12 to 17°C improved growth and feed efficiency.</P>

      • Inhibition of matrix metalloproteinases expression in human dental pulp cells by all-trans retinoic acid

        <P>All-trans retinoic acid (ATRA) inhibits matrix metalloproteinase (MMP)-2 and MMP-9 in synovial fibroblasts, skin fibroblasts, bronchoalveolar lavage cells and cancer cells, but activates MMP-9 in neuroblast and leukemia cells. Very little is known regarding whether ATRA can activate or inhibit MMPs in human dental pulp cells (HDPCs). The purpose of this study was to determine the effects of ATRA on the production and secretion of MMP-2 and -9 in HDPCs. The productions and messenger RNA (mRNA) expressions of MMP-2 and -9 were accessed by gelatin zymography and real-time polymerase chain reaction (PCR), respectively. ATRA was found to decrease MMP-2 level in a dose-dependent manner. Significant reduction in MMP-2 mRNA expression was also observed in HDPCs treated with 25 µmol⋅L<SUP>−1</SUP> ATRA. However, HDPCs treated with ATRA had no effect on the pattern of MMP-9 produced or secreted in either cell extracts or conditioned medium fractions. Taken together, ATRA had an inhibitory effect on MMP-2 expression in HDPCs, which suggests that ATRA could be a candidate as a medicament which could control the inflammation of pulp tissue in vital pulp therapy and regenerative endodontics.</P>

        Preparation of LiFe PO<sub>4</sub> Using Chitosan and its Cathodic Properties for Rechargeable Li-ion Batteries

        The LiFeP$O_4$ powder was synthesized by using the solid state reaction method with Fe($C_2O_4){\cdot}2H_2O,\;(NH_4)_2HPO_4,\;Li_2CO_3$, and chitosan as a carbon precursor material for a cathode of a lithium-ion battery. The chitosan added LiFePO4 powder was calcined at 350 ${^{\circ}C}$ for 5 hours and then 800 ${^{\circ}C}$ for 12 hours for the calcination. Then we calcined again at 800 ${^{\circ}C}$ for 12 hours. We characterized the synthesized compounds via the crystallinity, the valence states of iron ions, and their shapes using TGA, XRD, SEM, TEM, and XPS. We found that the synthesized powders were carbon-coated using TEM images and the iron ion is substituted from 3+ to 2+ through XPS measurements. We observed voltage characteristics and initial charge-discharge characteristics according to the C rate in LiFeP$O_4$ batteries. The obtained initial specific capacity of the chitosan added LiFeP$O_4$ powder is 110 mAh/g, which is much larger than that of LiFeP$O_4$ only powder.
