      • 인슐린의 세포사멸억제 작용에서 신호전달경로의 특성

        Insulin has a variety of activities like as glucose uptake, inhibition of lipolysis, glycogen biosynthesis in a number of cells and tissues. Recently insulin has known to exert an antiapoptotic activity in different cell systems. However, it is not clear by which insulin can protect apoptosis cells from apoptosis. Thus, CHO cells expressing the human insulin receptors (CHO-IR) were used to evaluate the importances of different signaling steps activated by insulin in this process. The Ras/Raf-1/MEK/ERK cascade and the serine/threonine protein kinase Akt are two survival pathways that may be activated in response to insulin. We tested the hypothesis that inhibition of farnesylated Ras was casually related to manumycin-induced apoptosis and showed that the response to manumycin was found to be independent of K-Ras function. Moreover, blocking Ras/Raf-1/MEK/ERK cascade by the expression of a transdominant inhibitory mSOSI mutant in CHO-IR cells kept cells to sensitive to the antiapoptotic action of insulin. Insulin-dependent activation of Akt was blocked by 4th treatment with manumycin. This study suggest that the deplection of short-lived farnesylated proteins by manumycin suppresses the antiapoptotic action of insulin at least in part by disrupting Akt­activation but not of the K-Ras/Raf-1/MEK/ERK-dependent cascade.

      • 금강 수서생태계의 구조와 기능에 관한 연구 : 수질 및 미생물

        소백산맥의 주봉인 덕유산에서 발원하여 군산지역에서 황해로 유입되는 금강의 본류는 총 유로연장이 401.4km에 이르는 우리나라 4대강 중의 하나이며, 대전광역시, 충청남북도 및 전라북도 일부지역의 생활용수 및 산업용수의 공급원으로서 대단히 중요한 역할을 담당하고 있다. 1960년대 이후 계속적으로 추진된 경제개발과 공업화 정책에 의해 인구의 도시집중화, 생활수준의 향상등이 이루어지고, 이에 따라 양질의 생활용수 및 산업용수에 대한 수요는 계속적으로 크게 증가하고 있는 실정이다. 그러나 금강의 경우 도시의 생활하수, 공업단지로부터의 공업폐수, 유역의 축산폐수 등에 의해 수질을 비롯한 수환경의 악화가 점차 심화되고 있으며, 이에 따른 수서생태계의 변화도 매우 큰 것으로 보고되고 있다. 특히 최근에 조성된 바 있는 금강 하구둑이 94년 8월부터 가동되고 있고, 또 1995년 이후 금강종합개발이 중류와 하류지역을 중심으로 대대적으로 시행될 예정이어서 금강 본류의 수환경은 장차 많은 변화가 예상된다. 수환경의 변화는 생육하는 생물들의 종조성은 물론 군집의 구조와 기능에 큰 영향을 미치기 때문에 금강 본류의 수서생태계에도 많은 변화가 수반될 것으로 예측되고 있다. 이와 같이 금강본류를 대상으로 하는 사업이 시행된 후 야기되는 수환경의 변화는 장기간에 걸친 조사를 요구하며, 이로부터 얻은 결과를 사업 시행 이전의 조사 결과와 비교함으로써만이 효과적으로 그 경향을 파악할 수 있다. 또한 이러한 비교 분석은 수질관리를 위한 대책 수립과 효율적인 사후 환경관리를 모색하는데 대단히 중요하다. 이에 반하여 지금까지 금강을 대상으로 한 수환경 및 수서생태계의 조사 연구는 단편적인 분야에 대하여 극히 단기간 수행된 것이 대부분이고 정량분석이 아닌 정성분석에만 치중되었기 때문에 금강의 수질 및 생태게보존을 위한 자료로서의 기능을 발휘하지 못하고 있다. 이러한 관점에서 본 환경문제연구소에서는 장기(향후 5년 이상)적인 계획하에 금강본류를 대상으로 수서생태계의 구조와 기능을 조사ㆍ분석하고, 이를 통하여 금강의 수질 및 생태계보존과 수환경관리를 위한 기초자료를 수립하고자 한다. 이와 같이 본 환경문제연구소가 장기적으로 추진하려 하는 금강 수서생태계학술조사는 금강 수서생태계의 현황은 물론 금강을 대상으로 하는 여러가지 형태의 사업에 의한 변화 양상을 정확하게 파악하고, 이에 대한 적절한 대처 방안을 강구하게 할 수 있는 기초자료를 제공하여 줄 것으로 기대된다. 지금까지 금강에 대한 종합적인 생태계 조사가 이루어지지 않고 있음을 고려할 때, 본 학술조사는 장차 금강 수서생태계에 대한 연구의 지침이 될 것으로 기대되며, 이는 본교가 금강 생태계의 보존과 관리를 통하여 지역사회에 기여하는 중추적 학술기관으로서의 위치를 확고히 마련하는 계기가 될 것이다. 본 보고서는 이러한 연구계획의 1차년도 연구결과로서 수질 및 미생물, 수생식물, 어류, 동식물 플랭크톤, 수서곤충을 중심으로 금강 중ㆍ하류 유역 수서생태계를 구성하고 있는 주요 생물군의 구조와 기능을 정량적으로 조사ㆍ분석하는데 그 목적을 두었다. Heterotrophic microbial populations and physicochemical parameters of water samples collected from six stations on the Keum River have been examined from November 1994 to July 1995. The ranges for BOD and COD were 1.5-11.3㎎/L and 2.1-13.7㎎/L, respectively. At the station closest to the estuary, the values of BOD and COD were higher than at other stations, but those of PO₄-P were lower. Metal ions including Cu, Mn, Pb, Cd, and Hg were not detected at all. The population densities of heterotrophic bacteria amounted to 13.7-22.0% of the total bacteria. The densities of coliforms, Salmonella-Shigella, and yeasts were in the range of 49-538 cfu/ml, 16-271 cfu/100ml, and 30-259 cfu/㎖, respectively.

      • 飼料資源開發을 위한 雜灌木樹葉의 飼料價値와 韓牛의 放牧利用에 關한 硏究

        As a part of forestry utilization for livestock feed resources the feeding value ague of shrub leaf by growth stage and grazing behabiour of the native cattle at shrubby grassland in function of seasons were investigated and the following results were obtained. 1. The crude protein content of oak leaf declined as the growth stage increased whereas the contents of crude fiber and crude fat showed the opposite tendency. 2. The intake of oak leaf paralleled with the amount given while the intake of the mixed grass leas remarkable increased and the body weight gain was also remarkably increased. 3. The average number of oak leaf intake was 70.5 times for spring, 148.7 for summer, and 395.7 for autumn. 4. The grazing time by the native cattle was decreased in the order of spring, autumn, and summer. As for the grazing behaviour, the intake activity was most active during morning, mid-day and late afternoon or three times a day. 5. The duration of rumination was found to be in the decreasing order or summer, autumn and spring, cattles ruminated more when the grasses were more abundant. 6. As for the inactive period of animal, it was short during spring when the animals spent more time for eating but it was about 5 times longer during both summer and autumn.

      • Proteomic analysis of differentially expressed proteins induced by rice blast fungus and elicitor in suspension-cultured rice cells

        We used two-dimensional electrophoresis (2-DE) and other proteomic approaches to identify proteins expressed in suspension-cultured rice cells in response to the rice blast fungus, Magnaporthe grisea. Proteins were extracted from suspension-cultured cells at 24 and 48 h after rice blast fungus inoculation or treatment with elicitor or other signal molecules such as jasmonic acid (JA), salicylic acid, and H_(2)O_(2). The proteins were then polyethylene glycol fractionated before separation by 2-DE. Fourteen protein spots were induced or increased by the treatments, which we analyzed by N-terminal or internal amino acid sequencing. Twelve proteins from six different genes were identified. Rice pathogen-related protein class 10 (OsPR-10), isoflavone reductase like protein, β-glucosidase, and putative receptor-like protein kinase were among those induced by rice blast fungus; these have not previously been reported in suspension-cultured rice cells. Six isoforms of probenazole-inducible protein (PBZ1) and two isoforms of salt-induced protein (SalT) that responded to blast fungus, elicitor, and JA were also resolved on a 2-DE gel and identified by proteome analysis. The expression level of these induced proteins both in suspension-cultured cells and in leaves of whole plants was analyzed by Western blot. PBZ1, OsPR-10, and SalT proteins from incompatible reactions were induced earlier and to a greater extent than those in compatible reactions. Proteome analysis can thus distinguish differences in the timing and amount of protein expression induced by pathogens and other signal molecules in incompatible and compatible interactions.

        Properties of ZnO:Ga thin films deposited by RF magnetron sputtering under various RF power

        ZnO:Ga thin films were deposited by RF magnetron sputtering technique from ZnO (3 wt.% Ga2O3) target onto glass substrates under various RF power. The influence of RF power on the structural, electrical, and optical properties of ZnO:Ga thin films was investigated by X-ray diffraction, atomic force microscopy, Hall method and optical transmission spectroscopy. As the RF power increases from 50 to 110W, the crystallinity is deteriorated, the root main square surface roughness is decreased and the sheet resistance is increased. The increase of sheet resistance is caused by decreasing carrier concentration due to interstitial Ga ion. All films are transparent up to 80% in the visible wavelength range and the adsorption edge is a red-shift with increasing RF power.

      • Cofilin Phosphorylation Decreased by Serum-free Starvation with Low Glucose in the L6 Myoblasts

        <P>[Purpose] Many studies have been using cell culture models of muscle cells with exogenous cytokines or glucocorticoids to mimic atrophy in in vivo and in vitro tests. However, the changes in the phosphorylation of atrophy-related cofilin are still poorly understood in starved skeletal muscle cells. In this study, we first examined whether or not phosphorylation of cofilin is altered in L6 myoblasts after 3, 6, 12, 24, 48, and 72 hours of serum-free starvation with low glucose. [Methods] We used Western blotting to exam protein expression and phosphorylation in atrophied L6 myoblasts. [Results] L6 cell sizes and numbers were diminished as a result of serum-free starvation in a time-dependent manner. Serum-free starvation for 3, 6, 12, 24, 48, and 72 hours significantly decreased the phosphorylation of cofilin, respectively. [Conclusion] These results suggest that starvation-induced atrophy may be in part related to changes in the phosphorylation of cofilin in L6 myoblasts.</P>

      • Both α2,3- and α2,6-Linked Sialic Acids on O-Linked Glycoproteins Act as Functional Receptors for Porcine Sapovirus

        <▼1><P>Sapovirus, a member of the <I>Caliciviridae</I> family, is an important cause of acute gastroenteritis in humans and pigs. Currently, the porcine sapovirus (PSaV) Cowden strain remains the only cultivable member of the <I>Sapovirus</I> genus. While some caliciviruses are known to utilize carbohydrate receptors for entry and infection, a functional receptor for sapovirus is unknown. To characterize the functional receptor of the Cowden strain of PSaV, we undertook a comprehensive series of protein-ligand biochemical assays in mock and PSaV-infected cell culture and/or piglet intestinal tissue sections. PSaV revealed neither hemagglutination activity with red blood cells from any species nor binding activity to synthetic histo-blood group antigens, indicating that PSaV does not use histo-blood group antigens as receptors. Attachment and infection of PSaV were markedly blocked by sialic acid and <I>Vibrio cholerae</I> neuraminidase (NA), suggesting a role for α2,3-linked, α2,6-linked or α2,8-linked sialic acid in virus attachment. However, viral attachment and infection were only partially inhibited by treatment of cells with sialidase S (SS) or <I>Maackia amurensis</I> lectin (MAL), both specific for α2,3-linked sialic acid, or <I>Sambucus nigra</I> lectin (SNL), specific for α2,6-linked sialic acid. These results indicated that PSaV recognizes both α2,3- and α2,6-linked sialic acids for viral attachment and infection. Treatment of cells with proteases or with benzyl 4-O-β-D-galactopyranosyl-β-D-glucopyranoside (benzylGalNAc), which inhibits <I>O</I>-linked glycosylation, also reduced virus binding and infection, whereas inhibition of glycolipd synthesis or <I>N</I>-linked glycosylation had no such effect on virus binding or infection. These data suggest PSaV binds to cellular receptors that consist of α2,3- and α2,6-linked sialic acids on glycoproteins attached via <I>O</I>-linked glycosylation.</P></▼1><▼2><P><B>Author Summary</B></P><P>Although enteropathogenic sapoviruses and noroviruses are leading causes of acute gastroenteritis in both humans and animals, the study of viral pathogenesis and immunity of these ubiquitous pathogens has been hampered due to the lack of a fully permissive cell culture system. Porcine sapovirus Cowden strain provides a suitable system that can be used to identify the molecular mechanisms of viral pathogenesis. Previous studies have shown that carbohydrates and glycolipids play important roles in the attachment of members of the <I>Caliciviridae</I>; histo-blood group antigens (HBGAs) are used by <I>Norovirus</I> genogroups I to IV, as well as members of the <I>Lagovirus</I>, and <I>Recovirus</I> genera, whereas terminal sialic acid is recognized as a receptor for feline calicivirus and murine norovirus. To date, however, the role of carbohydrates in the life cycle of sapoviruses has remained largely unknown. We found that porcine sapovirus binds to susceptible host cells through both α2,3- and α2,6-linked terminal sialic acids which are attached to <I>O</I>-linked glycoproteins. These efforts, findings and insights will significantly contribute to a better understanding of the sapovirus life cycle.</P></▼2>

        Dislocation Reduction of GaN Layer with a SiN Mask by Using Metalorganic Chemical Vapor Deposition

        GaN layers with an in-situ SiN mask were grown by using metalorganic chemical vapor deposition (MOCVD) and the physical properties of the GaN layer were studied. Also, the influence of a SiN mask on the optical property of GaN layer was investigated. By inserting a SiN mask, the (102) full width at half maximum (FWHM) decreased from 480 arcsec to 409 arcsec, and the threading dislocation (TD) density decreased from 3.21 $\times$ 10$^{9}$ cm$^{-2}$ to 9.7 $\times$ 10$^{8}$ cm$^{-2}$. The photoluminescence (PL) peak intensity of sample with a SiN mask increased by 2 times compared to that of sample without a SiN mask. We have shown that a SiN mask significantly improved the physical and the optical properties of the GaN layer.

      • Decrease of PKB/Akt Phosphorylation is Partially Mediated by SAPK/JNK Activation in Serum-free L6 Myoblasts Starved with Low Glucose

        <P>[Purpose] Studies have been using cell cultures of muscle cells to mimic atrophy in <I>in vivo</I> and <I>in vitro</I> tests. However, changes in the activation of atrophy-related PKB/Akt is not fully understood in serum-free starved skeletal muscle cells. The purpose of the present study was to determine the change of PKB/Akt phosphorylation in L6 myoblasts under serum-free starvation conditions. [Methods] We used western blotting to examine PKB/Akt expression and phosphorylation in atrophied L6 myoblasts. [Results] The phosphorylation of PKB/Akt was significantly lower in L6 myoblasts under serum-free starvation than that of the control group. Serum-free starvation for 6, 12, 24, 36, 48, 72, 96, and 120 hours significantly decreased the phosphorylation of PKB/Akt. Furthermore, the decrease of PKB/Akt phosphorylation under serum-free starvation was partially restored by SP600125, an inhibitor of SAPK/JNK. [Conclusion] These results suggest that decrease of PKB/Akt phosphorylation due to serum-free starvation with low glucose is partially related to the activity of SAPK/JNK in L6 myoblasts.</P>
