Protein‐stabilizing and cell‐penetrating properties of α‐helix domain of 30Kc19 protein
Ryu, Jina,Kim, Hyoju,Park, Hee Ho,Lee, Hong Jai,Park, Ju Hyun,Rhee, Won Jong,Park, Tai Hyun WILEY‐VCH Verlag 2016 Biotechnology journal Vol.11 No.11

Abstract

The protein‐stabilizing and cell‐penetrating activities of Bombyx mori 30Kc19 α‐helix domain (30Kc19α) are investigated. Recently, 30Kc19 protein has been studied extensively as it has both protein‐stabilizing and cell‐penetrating properties. However, it is unknown which part of 30Kc19 is responsible for those properties. 30Kc19 protein is composed of two distinct domains, an α‐helix N‐terminal domain (30Kc19α) and a β‐trefoil C‐terminal domain (30Kc19β). The authors construct and produce truncated forms of 30Kc19 to demonstrate their biological functions. Interestingly, 30Kc19α was shown to be responsible for both the protein‐stabilizing and cell‐penetrating properties of 30Kc19 protein. 30Kc19α shows even higher protein delivery activity than did whole 30Kc19 protein and has low cytotoxicity when added to cell culture medium. Therefore, based on its multifunctional properties, 30Kc19α can be developed as a novel candidate for a therapeutic protein carrier into various cells and tissues.

The significant increase in life expectancy is closely related to the growing interest in the impact of aging on the function and appearance of the skin. Skin aging is influenced by several factors, and solar ultraviolet (UV) irradiation is considered one of the most important causes of skin photoaging. The aim of this study was to examine the anti-photoaging role of porphyra-334 from Porphyra (P.) yezoensis, a mycosporine-like amino acid (MAA), using high-performance liquid chromatography (HPLC), and electrospray ionization-mass spectrometry (ESI-MS). In the present study, extracted UV-absorbing compounds from P. yezoensis included palythine, asterina-330 and porphyra-334. Porphyra-334 was the most abundant MAA in P. yezoensis, and it was therefore used for conducting antiphotoaging experiments. The effect of porphyra-334 on the prevention of photoaging was investigated by measuring reactive oxygen species (ROS) production and matrix metalloproteinase (MMP) levels, as well as extracellular matrix (ECM) components and protein expression in UVA-irradiated human skin fibroblasts. Porphyra-334 suppressed ROS production and the expression of MMPs following UVA irradiation, while increasing levels of ECM components, such as procollagen, type I collagen, elastin. These results suggest that porphyra-334 has various applications in cosmetics and toiletries because of its anti-photoaging activities and may serve as a novel anti-aging agent.

Distributed Control System(DCS) have been one of the main research focuses in industry for many decades. With these growing research trends, it is important to instruct the open control system which features expandability, portablility and interoper ability. This paper reports a survey of DCS based on EPICS for developing open control system in industrial applications.

Patients with colorectal cancer may experience symptoms such as diarrhea, nausea, and anorexia, during surgery and chemotherapy, which can increase the risk of malnutrition. In addition, dietary habits play a key role in the onset of colorectal cancer; therefore, it is necessary to improve dietary habits to prevent recurrence during treatment after diagnosis. In this study, a clinical nutritionist conducted 4 interviews for patients diagnosed with colorectal cancer and scheduled for colectomy: before surgery, after surgery, 1st chemotherapy, and 2nd chemotherapy, and provided nutrition care for each treatment course to determine its effects on nutrition status and disease prognosis. Significant weight loss but no decrease in muscle mass was observed during treatment. Body fat mass, although not statistically significant, showed a decreasing tendency. The percentage of people who responded ‘yes’ to the below items increased after compared to before receiving nutrition education: ‘I eat meat or eggs more than 5 times a week,’ ‘I eat seafood at least three times a week,’ ‘I eat vegetables at every meal,’ ‘I eat fruits every day,’ and ‘I eat milk or dairy products every day.’ These results indicate that the patients changed their dietary habit from a monotonous eating pattern to a pattern of consuming various food groups after receiving nutrition education. These results suggest that continuous nutrition care by clinical dietitians, according to the patient’s treatment process, can help improve the patient's nutritional status and establish healthy eating habits.

Objective: Recently the FIGO staging system of cervical cancer was revised. New system adopted a surgical staging concept and imaging tests such as PET, MRI, and CT. The lymph node metastasis was included as a stage-affecting factor for the first time. Therefore we were curious about the performance of predicting prognosis. Methods: We collected cervical cancer patients’ medical records from 2002 to 2015 in our institute. Totally 1,018 patients received primary treatment in our hospital. Patients who had been treated in other hospitals were excluded. Combined malignancy, major medical illness, follow up loss were exclusion criteria. We analyzed the data with SPSS v 20 and STATA v 13. Results: We analyzed totally 1,018 patients with medical record search. Mean age at diagnosis was 52.7 year. The number of patients with stage 2B was 190 using old system while 128 using new system. The number of patients with stage 3C1 was 208 and 54 for 3C2. The five year PFS of 2B patients 74.9%, while 3C1 patients show 67.9% (p=0.077). Additionally, 3C2 patients show 52.7% of five year PFS duration. The five year PFS of 1B2 patients was 94.0%, while 1B3 patients showed 84.8% (p=0.154). Conclusion: This is data from single institute. However it showed new FIGO staging system seems to be appropriate to predict the prognosis of cervical cancer after introduction of radiological and surgical staging component. We suggest multi-institute study to verify the issue.
