급성 골수성 백혈병 환자에서 관해 유도 항암 화학요법 후 발생한 베르니케 뇌증

Wernicke's encephalopathy is characterized by the triad of ocular symptoms, ataxia, and mental confusion and neuropsychiatric condition generally caused by acute thiamine deficiency. Although it is common in the severe alcoholics, several other causes also have been identified, such as total parenteral nutrition (TPN) use, hyperemesis gravidarum, anorexia nervosa, hemodialysis, uremia, and acquired immunodeficiency syndrome(AIDS). Chemotherapy in patient with malignanacy and long term total parenteral nutrition following bone marrow transplantation are related with Wernicke's encephalopathy. A 48-year-old woman had been treated with Daunorubicin and Cytarabine for remission induction chemotherapy of acute myeloblastic leukemia and intravenous hyperalimentation due to persistent hematochezia and ileus caused by infiltration of leukemic cell in terminal ileum. She suddenly complained of diplopia, tremor, and mental confusion at 32th day after chemotherapy and generalized tonic-clonic seizure at 34th day after chemotherapy. The diagnosis of Wernicke's encephalopathy was made with clinical presentations and brain MRI. Brain MR T2-weighted & Flair image revealed a high signal intensity lesions bilaterally at the medial thalamus and peri-acueductal grey matter of the midbrain. The patient was successfully treated with vitamin B1. We presented a case of acute Wernicke's encephalopathy developed after remission induction chemotherapy followed by intravenous hyperalirnentation in a patient with acute myeloblastic leukemia.

SiC-fiber의 전구체로 쓰이는 polymethylsilane(PMS), polydimethylsilane(PDMS),polyemthylphenylsilane(PMPS)은 Na촉매 하에 dichloro-organosilanes의 탈 염소화 반응으로 합성된다. 유기용매에 잘 녹는 PMPS를 이용하여 온도, 시간, 첨가제의 농도에 따른 수율과 평균분자량을 조사하였으며,고온에서의 잔류량을 확인하였다. PMPS의 합성시간이 증가할수록 수율과 분산도가 증가하였고 평균분자량이 감소하였다. 활성탄을 첨가하여 합성 시 PMPS은 높은 평균 분자량을 가졌다. Na/C=0.05 첨가 시까지 평균 분자량이 높아졌고, 고분자 분산도는 균일했다. Na/C=0.1 첨가 시 분자량과 분산도가 크게 증가했다. PMPS의 고온 잔류량은 활성탄의 첨가량을 많이 하고, 첨가 후 반응 시간을 늘려줄수록 증가하였다. 반응 시 첨가되는 활성탄의 양과 합성시간을 조절함으로 PMPS의 평균 분자량을 조절할 수 있었다. Polymethylsilane(PMS), polydimethylsilane(PDMS),polyemthylphenylsilane(PMPS), which were used as a precursor of SiC-fiber, were synthesized by dehalocoupling were dichloro-organosilanes using Na as a reductant. By using the PMPS which is easily melted at organic solvent, investigated how yield, average molecular weight and remaining quantity in high temperature be influenced by temperature, time and concentration of additive. When reaction time of PMPS became longer, yield and polydispersity were increased and average molecular weight was decreased. synthesis of PMPS with active carborn as an additive showed high average molecular weight was high while polydispersity was regular. In case of Na/C=0.1, average molecular weight and polydispersity was remarkably increased. Residual quantity in high temperature of PMPS was increased in lots of active carborn and long reaction time. Average molecular weight of PMPS can be controlled by reaction time and amount of active carborn added in polymerization.

<P>Umbilical cord blood (UCB)-derived mesenchymal stem cells (MSCs) are multipotent adult stem cells, which can differentiation into cells of connective tissue and neural lineages. This study investigated the potential for neuronal differentiation of red fluorescent protein (RFP)-transgenic cat UCB-derived MSCs. The cells were cultured in pre-induction medium for 24 hr and in neuronal-induction medium for 72 hr. Immunofluorescent staining showed that 6.85% of the total cells were β III-tubulin-positive, 3.37% were neurofilament light (NF-L)-positive and 7.04% were neurofilament medium (NF-M)-positive. A β III-tubulin band was detected by western blot analysis. Our results demonstrate that RFP-transgenic UCB-derived MSCs can be differentiated into neuronal cells <I>in vitro</I>. Thus, RFP-transgenic MSCs could provide alternative tracing material for stem cell transplantation.</P>

Electrorheological Suspensions of Two Polarizable Particles

Bi-disperse Electrorheological (ER) suspensions of two polarizable particles of the same size are investigated to understand the ER behavior of poly-disperse suspensions composed of various polarizable particles. The electrostatic polarization model is employed to describe ER suspensions, and solutions to the equation of motion are obtained by dynamic simulation. Even with the applied electric field, metastable structures and sheared configurations at a shear rate of 0.01 and 10 s^(-1) show no inhomogeneous higher polarizable particle distributions (no higher polarizable particle cluster formation) regardless of the ratio of the two types of particles. The shear stress increases with the increase of the higher polarizable particle concentration both in the electrostatic force and hydrodynamic force dominant regions.

Bt crystal proteins, encoded by cry genes, are a group of insecticidal proteins unique in the Gram-positive and spore-forming bacterium, Bacillus thuringiensis. These cry genes are widely applied as one of the most successful candidates for constructing transgenic plants resistant to pest insects. In our previous report, we found Cry1-5 had high insecticidal activity against Spodoptera larvae although its amino acid sequences showed high similarity (97.9%) to those of Cry1Ab which had low activity. In comparison with Cry1Ac, Cry1-5 had 12 different residues in domain Ⅰ and domain Ⅱ, and we focused on domain Ⅰand domain Ⅱ regions and designed 10 mutagenic primers to change 12 residues. Through multi site-directed mutagenesis, we mutated the modified cry1Ac gene by plant codon usage in pOBⅠ-Mod-cry1Ac based on cry1-5 and constructed 63 various mutant cry genes. In the further study, we will express those mutant proteins as a fusion form with polyhedrin using baculovirus expression system and subsequently do bioassay to Spodoptera larvae.

To develop an improved baculovirus insecticide with additional advantages, a novel recombinant baculovirus, AcB5B-AaIT was constructed. B. thuringiensis crystal protein gene (cry1-5) and insect-specific neurotoxin gene (AaIT) were introduced into Autographa californica nucleopolyhedrovirus genome by fusion of polyhedrin-cry1-5-polyhedrin under the control of polyhedrin (polh) gene promoter, and AaIT under the control of early promoter of ORF3004 from Cotesia plutellae bracovirus, respectively. About 150 kDa of Polyhedrin-Cry1-5-Polyhedrin fusion protein expressed by AcB5B-AaIT was occluded into the polyhedra produced by the recombinant virus, and activated as about 65 kDa of crystal protein when treated with gut-juice of Bombyx mori. The AcB5B-AaIT showed about 50% reduced LT50 value compared to that of the recombinant virus, Ap1Ac, expressing Cry1Ac against Plutella xylostella larvae. In addition, Spodoptera exigua larvae fed the recombinant polyhedra of AcB5B-AaIT showed about 4 fold higher refusing diet effect compared S. exigua larvae fed the recombinant polyhedra of the recombinant virus, Ap1C, expressing Cry1C. AcB5B-AaIT could be transferred to wild-type baculovirus along with serial passage by the homologous recombination between two polyhedrin genes contained in polh-cry1-5-polh fusion protein gene. These results suggested that the novel recombinant baculovirus, AcB5B-AaIT, could be applied as advanced viral insecticide.

N/A Chitosan has been degraded by chemical and enzymatic processes, and the efficiency and oligosaccharide distribution of the products were compared. When chemically hydrolyzed with 12 N HCI at 80 ℃, the reaction product contained the monomer to octamer, in which the yield of high degree of polymerization (dp) chitooligosaccharides was low and glucosamine was identified as major product. When enzymatically hydrolyzed with Bacillus sp. P16 chitosanase, the reaction products contained dimer to nanomer, in which the yield of high dp chitooligosaccharides was relatively high, and Pentamer and hexamer were identified as major products. Thus, enzymatic processes have advantages over chemical hydrolysis in terms of the efficiency and distribution of high dp chitooligosaccharides.bacillus sp. P16 chitosanase was evaluated as a potential source for the production of high dp chitooligosaccharides from chitosan.
Advanced recombinant baculovirus expressing insect neurotoxin and Bacillus thuringiensis Cry toxin

To develop an advanced baculovirus insecticide with additional advantages, such as higher toxicity and recovering to wild-type baculovirus, a novel recombinant baculovirus, NeuroBactrus was constructed. Bacillus thuringiensis crystal protein gene (cry1-5) and an insect-specific neurotoxin gene (AaIT) were introduced into Autographa californica nucleopolyhedrovirus genome by fusion of polyhedrin-cry1-5-polyhedrin under the control of poyhedrin gene promoter, and by fusion of orf603 partial genes and AaIT under the control of early promoter of ORF3006 from Cotesia plutellae bracovirus. About 150 kDa of Polyhedrin-Cry1-5-Polyhedrin fusion protein expressed by NeuroBactrus was occluded into the polyhedra, and activated as about 65 kDa of crystal protein when treated with trypsin. RT-PCR analysis indicated that transcription of AaIT gene occurs by 2 h postinfection (p.i.) and increased at 16 h p.i.. NeuroBactrus showed high toxicity against Plutella xylostella larvae and significant reduction in median lethal time (LT50) against Spodoptera exigua larvae compared to those of wild-type AcNPV. Re-recombinants derived from NeuroBactrus, NBt-Del5 (deleted cry1-5), NBt-DelA (deleted AaIT) and NBt-Del5A (deleted cry1-5 and AaIT; wild-type baculovirus) were generated in serial passages in vitro. This result showed that the NeuroBactrus could be transferred to wild-type baculovirus along with serial passages by the homologous recombination between two polyhedrin genes and two partial orf603 genes.
