        <P>The HUPO Brain Proteome Project (HUPO BPP) held its 12th workshop in Toronto on 26 September 2009 prior to the HUPO VIII World Congress. The principal aim of this project is to obtain a better understanding of neurodiseases and ageing, with the ultimate objective of discovering prognostic and diagnostic biomarkers, in addition to the development of novel diagnostic techniques and new medications. The attendees came together to discuss progress in the human clinical neuroproteomics and to define the needs and guidelines required for more advanced proteomic approaches.</P>

      • Schizophrenia: From the brain to peripheral markers. A consensus paper of the WFSBP task force on biological markers

        <P>Objective. The phenotypic complexity, together with the multifarious nature of the so-called 'schizophrenic psychoses', limits our ability to form a simple and logical biologically based hypothesis for the disease group. Biological markers are defined as biochemical, physiological or anatomical traits that are specific to particular conditions. An important aim of biomarker discovery is the detection of disease correlates that can be used as diagnostic tools. Method. A selective review of the WFSBP Task Force on Biological Markers in schizophrenia is provided from the central nervous system to phenotypes, functional brain systems, chromosomal loci with potential genetic markers to the peripheral systems. Results. A number of biological measures have been proposed to be correlated with schizophrenia. At present, not a single biological trait in schizophrenia is available which achieves sufficient specificity, selectivity and is based on causal pathology and predictive validity to be recommended as diagnostic marker. Conclusions. With the emergence of new technologies and rigorous phenotypic subclassification the identification of genetic bases and assessment of dynamic disease related alterations will hopefully come to a new stage in the complex field of psychiatric research.</P>

      • Inter-Lab Proteomics: Data Mining in Collaborative Projects on the Basis of the HUPO Brain Proteome Project's Pilot Studies.

        <P>Several projects were initiated by the Human Proteome Organisation (HUPO) focusing on the proteome analysis of distinct human organs. The initiative dedicated to the brain, its development and correlated diseases is the HUPO Brain Proteome Project (HUPO BPP). An objective data submission, storage, and reprocessing strategy have been established with the help of the results gained in a pilot study phase and within subsequent studies. The bioinformatic relevance of the data is drawn from the inter-laboratory comparisons as well as from the recalculation of all data sets submitted by the different groups. In the following, results of the single groups as well as the centralised reprocessing effort are summarised, demonstrating the added-value of this concerted work.</P>

      • A-GPS (Assisted GPS), is this the system we need for indoor location & navigation?

        For a long time the GSM and GPS world has been living side by side. There have been some half hearted attempts to invoke the GPS unit into GSM units but performance and usability has suffered. The A-GPS approach is interesting and we wanted to verify general performance for fleet application and alarm/emergency situations. The result is now promising but still more to enhancements are anticipated. We achieved near acceptable availability (~70%) under indoor conditions. Accuracy was as anticipated (50-100 meter) as we are using reflected signals. Time to first fix is in general good in outdoor conditions but too long for indoor conditions (45-60 seconds). We would like manufactures to put some more effort to get better performance in the future as test with Nordnav high sensitivity measurement system proves that conditions indoor are at a level where better performance should be possible.

      • Type IIP supernova light curves affected by the acceleration of red supergiant winds

        <P>We introduce the first synthetic light-curve model set of Type IIP supernovae exploded within circumstellar media in which the acceleration of the red supergiant winds is taken into account. Because wind acceleration makes the wind velocities near the progenitors low, the density of the immediate vicinity of the red supergiant supernova progenitors can be higher than that extrapolated by using a constant terminal wind velocity. Therefore, even if the mass-loss rate of the progenitor is relatively low, it can have a dense circumstellar medium at the immediate stellar vicinity and the early light curves of Type IIP supernovae are significantly affected by it. We adopt a simple beta velocity law to formulate the wind acceleration. We provide bolometric and multicolour light curves of Type IIP supernovae exploding within such accelerated winds from the combinations of three progenitors, 12-16 M-circle dot; five beta, 1-5; seven mass-loss rates, 10(-5)-10(-2)M(circle dot) yr(-1); and four explosion energies, (0.5-2) x 10(51) erg. All the light-curve models are available at https://goo.gl/o5phYb. When the circumstellar density is sufficiently high, our models do not show a classical shock breakout as a consequence of the interaction with the dense and optically thick circumstellar media. Instead, they show a delayed 'wind breakout', substantially affecting early light curves of Type IIP supernovae. We find that the mass-loss rates of the progenitors need to be 10(-3)-10(-2)M(circle dot) yr(-1) to explain typical rise times of 5-10 d in Type IIP supernovae assuming a dense circumstellar radius of 10(15) cm.</P>

      • EMCCD-based spectrally resolved fluorescence correlation spectroscopy

        <P>We present an implementation of fluorescence correlation spectroscopy with spectrally resolved detection based on a combined commercial confocal laser scanning/fluorescence correlation spectroscopy microscope. We have replaced the conventional detection scheme by a prism-based spectrometer and an electron-multiplying charge-coupled device camera used to record the photons. This allows us to read out more than 80,000 full spectra per second with a signal-to-noise ratio and a quantum efficiency high enough to allow single photon counting. We can identify up to four spectrally different quantum dots in vitro and demonstrate that spectrally resolved detection can be used to characterize photophysical properties of fluorophores by measuring the spectral dependence of quantum dot fluorescence emission intermittence. Moreover, we can confirm intracellular cross-correlation results as acquired with a conventional setup and show that spectral flexibility can help to optimize the choice of the detection windows.</P>

      • Role of radiotherapy fractionation in head and neck cancers (MARCH): an updated meta-analysis

        <P><B>Summary</B></P> <P><B>Background</B></P> <P>The Meta-Analysis of Radiotherapy in squamous cell Carcinomas of Head and neck (MARCH) showed that altered fractionation radiotherapy is associated with improved overall and progression-free survival compared with conventional radiotherapy, with hyperfractionated radiotherapy showing the greatest benefit. This update aims to confirm and explain the superiority of hyperfractionated radiotherapy over other altered fractionation radiotherapy regimens and to assess the benefit of altered fractionation within the context of concomitant chemotherapy with the inclusion of new trials.</P> <P><B>Methods</B></P> <P>For this updated meta-analysis, we searched bibliography databases, trials registries, and meeting proceedings for published or unpublished randomised trials done between Jan 1, 2009, and July 15, 2015, comparing primary or postoperative conventional fractionation radiotherapy versus altered fractionation radiotherapy (comparison 1) or conventional fractionation radiotherapy plus concomitant chemotherapy versus altered fractionation radiotherapy alone (comparison 2). Eligible trials had to start randomisation on or after Jan 1, 1970, and completed accrual before Dec 31, 2010; had to have been randomised in a way that precluded prior knowledge of treatment assignment; and had to include patients with non-metastatic squamous cell carcinoma of the oral cavity, oropharynx, hypopharynx, or larynx undergoing first-line curative treatment. Trials including a non-conventional radiotherapy control group, investigating hypofractionated radiotherapy, or including mostly nasopharyngeal carcinomas were excluded. Trials were grouped in three types of altered fractionation: hyperfractionated, moderately accelerated, and very accelerated. Individual patient data were collected and combined with a fixed-effects model based on the intention-to-treat principle. The primary endpoint was overall survival.</P> <P><B>Findings</B></P> <P>Comparison 1 (conventional fractionation radiotherapy <I>vs</I> altered fractionation radiotherapy) included 33 trials and 11 423 patients. Altered fractionation radiotherapy was associated with a significant benefit on overall survival (hazard ratio [HR] 0·94, 95% CI 0·90–0·98; p=0·0033), with an absolute difference at 5 years of 3·1% (95% CI 1·3–4·9) and at 10 years of 1·2% (−0·8 to 3·2). We found a significant interaction (p=0·051) between type of fractionation and treatment effect, the overall survival benefit being restricted to the hyperfractionated group (HR 0·83, 0·74–0·92), with absolute differences at 5 years of 8·1% (3·4 to 12·8) and at 10 years of 3·9% (−0·6 to 8·4). Comparison 2 (conventional fractionation radiotherapy plus concomitant chemotherapy versus altered fractionation radiotherapy alone) included five trials and 986 patients. Overall survival was significantly worse with altered fractionation radiotherapy compared with concomitant chemoradiotherapy (HR 1·22, 1·05–1·42; p=0·0098), with absolute differences at 5 years of −5·8% (−11·9 to 0·3) and at 10 years of −5·1% (−13·0 to 2·8).</P> <P><B>Interpretation</B></P> <P>This update confirms, with more patients and a longer follow-up than the first version of MARCH, that hyperfractionated radiotherapy is, along with concomitant chemoradiotherapy, a standard of care for the treatment of locally advanced head and neck squamous cell cancers. The comparison between hyperfractionated radiotherapy and concomitant chemoradiotherapy remains to be specifically tested.</P> <P><B>Funding</B></P> <P>Institut National du Cancer; and Ligue Nationale Contre le Cancer.</P>

      • In vivo visualization of Alzheimer's amyloid plaques by magnetic resonance imaging in transgenic mice without a contrast agent

        <P>One of the cardinal pathologic features of Alzheimer's disease (AD) is the formation of senile, or amyloid, plaques. Transgenic mice have been developed that express one or more of the genes responsible for familial AD in humans. Doubly transgenic mice develop “human-like” plaques, providing a mechanism to study amyloid plaque biology in a controlled manner. Imaging of labeled plaques has been accomplished with other modalities, but only MRI has sufficient spatial and contrast resolution to visualize individual plaques noninvasively. Methods to optimize visualization of plaques in vivo in transgenic mice at 9.4 T using a spin echo sequence based on adiabatic pulses are described. Preliminary results indicate that a spin echo acquisition more accurately reflects plaque size, while a T<SUB>2</SUB>* weighted gradient echo sequence reflects plaque iron content, not plaque size. In vivo MRI–ex vivo MRI–in vitro histologic correlations are provided. Histologically verified plaques as small as 50 μm in diameter were visualized in living animals. To our knowledge this work represents the first demonstration of noninvasive in vivo visualization of individual AD plaques without the use of a contrast agent.
