Serotoninergic modulation of GABAergic synaptic transmission in developing rat CA3 pyramidal neurons

<P>Abstract</P><P>Serotoninergic modulation of GABAergic mIPSCs was investigated in immature (postnatal 12–16-days old) rat CA3 pyramidal neurons using a conventional whole-cell patch clamp technique. Serotonin or 5-hydroxytryptamine (5-HT) (10 &mgr;mol/L) transiently and explosively increased mIPSC frequency with a small increase in the current amplitude. However, 5-HT did not affect the GABA-induced postsynaptic currents, indicating that 5-HT acts presynaptically to facilitate the probability of spontaneous GABA release. The 5-HT action on GABAergic mIPSC frequency was completely blocked by 100 nmol/L MDL72222, a selective 5-HT<SUB>3</SUB> receptor antagonist, and mimicked by mCPBG, a selective 5-HT<SUB>3</SUB> receptor agonist. The 5-HT action on GABAergic mIPSC frequency was completely occluded either in the presence of 200 &mgr;mol/L Cd<SUP>2+</SUP> or in the Na<SUP>+</SUP>-free external solution, suggesting that the 5-HT<SUB>3</SUB> receptor-mediated facilitation of mIPSC frequency requires a Ca<SUP>2+</SUP>influx passing through voltage-dependent Ca<SUP>2+</SUP>channels from the extracellular space, and that presynaptic 5-HT<SUB>3</SUB> receptors are less permeable to Ca<SUP>2+</SUP>. The 5-HT action on mIPSC frequency in the absence or presence of extracellular Na<SUP>+</SUP> gradually increased with postnatal development. Such a developmental change in the 5-HT<SUB>3</SUB> receptor-mediated facilitation of GABAergic transmission would play important roles in the regulation of excitability as well as development in CA3 pyramidal neurons.</P>

<P><B>Abstract</B></P> <P>Despite expressing high levels of the epidermal growth factor receptor (EGFR), a majority of oral squamous cell carcinoma (OSCC) patients show limited response to cetuximab and ultimately develop drug resistance. However, mechanism underlying cetuximab resistance in OSCC is not clearly understood. Here, using a mouse orthotopic xenograft model of OSCC, we show that bone morphogenic protein-7-phosphorylated Smad-1, -5, -8 (BMP7-p-Smad1/5/8) signaling contributes to cetuximab resistance. Tumor cells isolated from the recurrent cetuximab-resistant xenograft models exhibited low EGFR expression but extremely high levels of p-Smad1/5/8. Treatment with the bone morphogenic protein receptor type 1 (BMPRI) inhibitor, DMH1 significantly reduced cetuximab-resistant OSCC tumor growth, and combined treatment of DMH1 and cetuximab remarkably reduced relapsed tumor growth <I>in vivo</I>. Importantly, p-Smad1/5/8 level was elevated in cetuximab-resistant patients and this correlated with poor prognosis. Collectively, our results indicate that the BMP7-p-Smad1/5/8 signaling is a key pathway to acquired cetuximab resistance, and demonstrate that combination therapy of cetuximab and a BMP signaling inhibitor as potentially a new therapeutic strategy for overcoming acquired resistance to cetuximab in OSCC.</P> <P><B>Highlights</B></P> <P> <UL> <LI> BMP7-p-Smad1/5/8 signaling contributes to cetuximab resistance in OSCC. </LI> <LI> DMHI, a BMP signaling inhibitor, reduced cetuximab-resistant OSCC tumor growth. </LI> <LI> Combined treatment of DMH1 and cetuximab significantly reduced relapsed tumor growth <I>in vivo.</I> </LI> <LI> p-Smad1/5/8 overexpression in OSCC patients was associated with poor disease free survival. </LI> </UL> </P>

A background-free, fast protein staining method in polyacrylamide gel electrophoresis using an acidic dye, zincon (ZC) and a basic dye, ethyl violet (EV) is described. It is based on the counterion dye staining technique that employs two oppositely charged dyes to form an ion-pair complex in staining solution. The selective binding of free dye molecules to proteins in acidic solution produces bluish violet-colored bands, It is a rapid and end-point staining procedure, involving only fixing and staining steps that are completed in 1-1.5 h. The detection limit of this method is 8-15 ng of protein that is comparable to the sensitivity of the colloidal Coomassie Brilliant Blue G (CBBG) stain. Due to its sensitivity and speed, this stain may be more practical than any other dye-based stains for routine laboratory purposes.
SHIFTED TABLEAU SWITCHINGS AND SHIFTED LITTLEWOOD-RICHARDSON COEFFICIENTS

We provide two shifted analogues of the tableau switching process due to Benkart, Sottile, and Stroomer; the shifted tableau switching process and the modified shifted tableau switching process. They are performed by applying a sequence of elementary transformations called switches and shares many nice properties with the tableau switching process. For instance, the maps induced from these algorithms are involutive and behave very nicely with respect to the lattice property. We also introduce shifted generalized evacuation which exactly agrees with the shifted J-operation due to Worley when applied to shifted Young tableaux of normal shape. Finally, as an application, we give combinatorial interpretations of Schur P- and Schur Q-function related identities.

This study was conducted to investigate suitable seeding rate under irrigation water-saving rice culture in ridge direct seeding on dry paddy during 1997∼1998. Dongjinbyeo was seeded from 60kg/ha to 120kg/ha on silty loam condition. The number of irrigation from 3-leaf stage to complete drainage were 7.5 times and total amount of irrigation was 258.75mm. The number of seedling stand rose with the increase of seeding rate, whereas the rate of seedling stand was decreased with the increase of it. Soil moisture tension was-2kPa at one day after stopped irrigation and lowered to-30kPa at five days after stopped irrigation. Leaf area index and shoot dry weight were increased with rising of seeding rate. Lodging index was increased with the rising of seeding rate, but lodging was not showed in field. Heading date, percent ripened grain and 1,000 grain weight had no difference with seeding rate, and number of spikelets per m^2 were highest at the rate of 100kg/ha. Rice yield was increased by 9% at the seeding rate of 100kg/ha and 4% at the rate of 80kg/ha and 120kg/ha compared with 60kg/ha. Therefore, it was considered that appropriate seeding rate was 100kg/ha under water-saving culture in ridge direct seeding on dry paddy.
Effects of Water Stress by PEG on Growth and Physiological Traits in Rice Seedlings

This study was conducted to evaluate the drought tolerance of Japonica and Indica rice cultivars during germinating and seedling stages by using the polyethylene glycol (PEG) solution. Each 5 cultivars of Japonica and Indica were cultured from 14 days after seeding(DAS) to 21 DAS using the PEG solution in a moderate water potential (-0.63 MPa). The lengths of radicle and plumule during the germinating stage were inhibitied by the PEG treatment to about 50% and 85%, respectively. The application of PEG to the seedling of two rice types caused to inhibit the plant height and leaf age about 23 % and 10%, respectively. Shoot and root dry weights by PEG treatment were inhibited more severely in Japonica than those in Indica. The difference on delaying of leaf area expansion between both rice types was not found with treatment of PEG, while the leaf color was increased in both Japonica and Indica by 19.9% and 9.2%, respectively. The average photosynthetic ability was inhibited more in Japonica to 36.0% than did Indica to 27.9%. The stomatal conductance was severely affected by PEG treatment, and the degree was varied in both rice types, ranged with 80-85% in Japonica and 29.3-81.6% in Indica. These results indicate there is little relationship between seed germination and seedling growth under the stress of low water potential.
