Self-illuminating nanoparticles for in vivo ROS imaging and photodynamic cancer therapy

Photodynamic therapy (PDT) for cancer, has been limited by low reactive oxygen species (ROS) quantum yield of photosensitizer (PS) and lack of the proper imaging system. Herein, we report a self-illuminating nanoparticles (SI-NPs) as a new theranostic agent based on chemiluminescence (CL) resonance energy transfer (CRET). Firstly, the SI-NPs are able to produce CO<sub>2</sub> by responding to H<sub>2</sub>O<sub>2</sub>, which is abundant at tumor sites, occurring CRET. By heat energy from PS with pulsed laser, thermal expansion-induced vaporization of CO<sub>2</sub> is proceeded, followed by amplifying the PA signal. Notably, SI-NPs generated 9 times stronger in vivo PA signal than the CRET-free NPs. Second, for successful PDT, the low ROS quantum yields of PS should be significantly improved. Since enhanced ROS quantum yields by CL and electron transfer can effectively inhibited tumor growth with complete regression in 60 % of cases in vivo. Overall, the SI-NPs might hold great potential phototheranositics for cancer.

Objectives: To classify trajectories of fasting blood glucose (FBS) levels and examine each trajectory`s associations with risk of cardiovascular disease (CVD). Methods: The National Health Insurance Service-National Sample Cohort (NHIS-NSC) sampled in the 2002 NHIS database was followed until 2010, and 13,829 participants aged 20 years and above had conducted nationwide health examinations annually. We used Cox proportional hazards models to examine the association of trajectories to risk of CVD. Four distinct trajectory groups were identified for FBS: low-stable, moderate-stable, elevated-upward, and High-upward. Results: During 88947.9 person-years of follow-up (mean follow-up, 6.4 years), we documented 2,778 incident case of CVD. Age-standardized incidence rate were increased with FBS levels (5,296.2 in low-stable group, 6,292.6 in moderate-stable group, and 8047.9 in elevated-upward group), but not in High-upward group. In multivariate models adjusted for age and sex, FBS was a significant predictor of CVD in elevated-upward group (hazard ratio (HR)=1.6, 95% confidence interval (CI):1.4-1.8) and High-upward group (HR=1.6, 95% CI:1.3-2.1). However, further adjustment for clinical covariates, only elevated-upward group was significantly associated with CVD (HR=1.2, 95% CI:1.1-1.4). Conclusions: Using the trajectory approach, we found that elevated-upward and High-upward FBS trajectories were associated with greater risk of CVD. These findings indicate the importance of FBS management across the lifespan, prognostic assessments and the targeting of prevention strategies to high-risk individuals.
일반 궐련담배 흡연 및 전자담배 이중 사용자의 심근경색과 협심증 관련성: 일반인 대상 단면연구

This a cross-sectional study aimed to investigate associations with dual use of electronic and conventional cigarettes for myocardial infarction and angina among Korean general population. The data of this study is from Korean National Health and Nutrition Examination Survey (2016-2021) pertaining to 26,143 participants aged 19 years or older. Of 26,143 participants, 3,075 (11.8%) was current conventional cigarettes smoker and 1,707 (6.5%) ever tried both e-cigarettes and conventional cigarettes (dual user). Among those dual user, 73.5% was current conventional cigarettes smoker, and they had 2-fold higher association for myocardial infarction and angina compared with non-smoker based on multivariable logistic regression model adjusted for covariates (OR=2.02, 95% CI: 2.01-2.03). The significant interaction effect was observed between smoking status and metabolic syndrome (MetS) prevalence (p<0.0001). Most e-cigarette users are dual users, and dual users have a higher association with myocardial infarction and angina than conventional cigarette-only smokers and non-smokers, and interaction with metabolic syndrome has also been confirmed in this relationship, so more active measures should be taken to target dual users. Smoking cessation education and intensive lifestyle intervention should be prioritized.
A New Sesterterpene from the Korean Sarcotragus sp. Sponge

Sarcotragin C (1), a new sesterterpene metabolite was isolated from a Sarcotragus sp. sponge collected from Chuja Island, Korea. On the basis of the combined spectroscopic analyses, the structure of this compound was determined to be a linear norsesterterpene containing a leucine-derived g-lactam moiety. This compound exhibited moderate cytotoxicity against K562 and A549 cell-lines.
HPLC-DAD-MS/MS를 이용한 금은화 생리활성 물질의 동시분석

A high-performance liquid chromatography (HPLC) with diode array detector (DAD) and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of chlorogenic acid (1), sweroside (2), luteolin-7-O-glucoside (3), (E)-aldosecologanin (4) and 3,5-dicaffeoylquinic acid (5) from Lonicera joponica flower buds. The optimal chromatographic conditions were obtained on an ODS column (5 ${\mu}m$, 4.6${\times}$150 mm) with the column temperature $25^{\circ}C$. The mobile phase was composed of (A) water with 0.1% formic acid and (B) acetonitrile with 0.1% formic acid using a gradient elution, the flow rate was 0.3 ml/min. Detection wavelength was set at 250 nm. All calibration curves showed good linear regression ($r^2$>0.994) within test ranges. The developed method provided satisfactory precision and accuracy with overall intra-day and inter-day variations of 0.05${\sim}$1.95% and 0.15${\sim}$2.26%, respectively, and the overall recoveries of 97.71${\sim}$103.65% for the five compounds analyzed. The verified method was successfully applied to quantitative determination of the three types (phenolic compounds, iridoids and flavonoids) of bioactive compounds in 21 commercial L. japonica flower buds samples from different markets in Korea and China. The analytical results demonstrated that the contents of the five analytes vary significantly with sources.
