        Inhibitory Activities of Ononin on Particulate Matter-induced Oxidative Stress

        Exposure to high levels of atmospheric particulate matter (PM) with an aerodynamic diameter of less than 2.5 μm (PM2.5) is associated with increased risk of respiratory injury, hospitalization, and respiratory death. The mechanism underlying the correlation between PM2.5 exposure and adverse health effects has not been fully elucidated, but oxidative stress due to PM2.5 appears to be an important molecular mechanism in PM2.5-mediated toxicity. The aim of this study was to determine the effects of ononin, a major isoflavone, on PM2.5-induced oxidative stress, and the mechanisms underlying these effects. Human pulmonary artery endothelial cells (HPAECs) were treated with PM2.5 and then ononin. Exposure to PM2.5 decreased cell viability in HPAECs in a time- and dose-dependent manner, possibly due to increased release of extracellular lactate dehydrogenase and generation of intracellular reactive oxygen species. PM2.5-induced oxidative damage was observed to occur via alterations to superoxide dismutase and catalase activities. Serum- and glucocorticoid-inducible kinase 1 (SGK1), a crucial cell survival factor, was downregulated by PM2.5. However, ononin treatment increased cell viability, decreased oxidative stress, and recovered the expression of SGK1 cell survival genes. Additionally, ononin increased the phosphorylation of mammalian target of rapamycin (mTOR) and dramatically suppressed the PM2.5-induced expression of toll-like receptor 4, MyD88, and the autophagy-related proteins LC3 II and Beclin 1. These findings indicate that ononin exhibits pulmonary protective effects against PM2.5- induced lung injury via inhibition of oxidative stress and mTOR-dependent autophagy pathways and is thus a potential therapeutic agent for PM2.5-induced lung injury.

        Antiplatelet, anticoagulant, and profibrinolytic activities of baicalin

        Baicalin is a flavonoid compound purified fromthe medicinal plant Scutellaria baicalensis Georgi and hasbeen reported to possess anti-inflammatory and anti-viralactivities. However, antiplatelet, anticoagulant, and profibrinolyticproperties of baicalin have not been studied. Inthis study, the anticoagulant and antiplatelet activities ofbaicalin were measured by monitoring activated partialthromboplastin-time (aPTT), prothrombin time (PT), theactivities of cell-based thrombin and activated factor X(FXa), platelet aggregation and thrombus formation. Theeffects of baicalin on the expressions of plasminogenactivator inhibitor type 1 (PAI-1) and tissue-type plasminogenactivator (t-PA) were also tested in tumor necrosisfactor-a (TNF-a) activated human umbilical vein endothelialcells (HUVECs). Our data showed that baicalininhibited thrombin-catalyzed fibrin polymerization andplatelet functions, prolonged aPTT and PT significantlyand inhibited the activities and production of thrombin andFXa. Baicalin also prolonged in vivo bleeding time andinhibited TNF-a induced PAI-1 production. Furthermore,PAI-1/t-PA ratio was significantly decreased by baicalin. Collectively, these results indicate that baicalin possessesantithrombotic activities and suggest that the current studycould provide bases for the development of new anticoagulantagents.

        RESEARCH ARTICLE Emodin-6-O-b-D-glucoside down-regulates endothelial protein C receptor shedding

        Endothelial protein C receptor (EPCR) plays animportant role in the protein C anticoagulation pathwayand in the cytoprotective pathway. Previously, EPCR canbe shed from the cell surface, which is mediated by tumornecrosis factor-a converting enzyme (TACE). SolubleEPCR levels are increased in patients with systemicinflammatory diseases. Recently, we reported that a newactive compound, emodin-6-O-b-D-glucoside (EG) fromReynoutria japonica, has anti-inflammatory activities. However, little is known of the effects of EG on EPCRshedding. Here, we investigated this issue by monitoringthe effects of EG on the phorbol-12-myristate 13-acetate(PMA) or the cecal ligation and puncture (CLP)-mediatedEPCR shedding and its underlying mechanisms. Datashowed that EG potently inhibited the PMA and CLPinducedEPCR shedding by suppressing TACE expression. Given these results, EG could be used as a candidatetherapeutic for the treatment of vascular inflammatory diseases.

        족관절 골절의 방사선학적 고찰

        The injuries to the anlke joint complex can cause not only bony fractures but also associated ligament rupture. Understanding of the mechanism of those injuries is important for the radiologist to give more information ot the orthopedist to identify ligament injuries and then to fix the fractures. In 1948, Lauge-Hansen devised a classification of ankle fractures based on the position of the foot and direction of injuring force at the time of injury. On the basis of location and appearance of the fibular fracture, 4 types of injuries are identified ; pronation-abduction (PA), pronation-external rotation (PER), supination-adduction (SA), and supination-external rotation (SER). 60 cases of ankle fractures treated at Kyung Pook National University Hospital from Jan. 1973 to Dec. 1982 were analized an dclassified. The results are as follows : 1. Male was more frequently affected than female and the sex ratio was 4 : 1. 2. The most commonly affected age group was 20's. 3. Right side was more frequently affected than left. 4. A mong 4 types, the most common was SER type (33.3%) and next were PER (30.3%), SA (16.7%), and PA (13.3%). types.

        Ginsenosides Inhibit HMGB1-induced Inflammatory Responses in HUVECs and in Murine Polymicrobial Sepsis

        Asian ginseng is used as a treatment for cardiovascular diseases, ischemia, and cancers. High mobility group box 1 (HMGB1) protein acts as a late mediator of severe vascular inflammatory conditions. However, the effect of ginsenosides from Asian ginseng on HMGB1-induced inflammatory responses has not been studied. We addressed this question by monitoring the effects of ginsenoside treatment on lipopolysaccharide (LPS) and cecal ligation and puncture (CLP)-mediated release of HMGB1, and HMGB1-mediated regulation of proinflammatory responses. Ginsenoside treatment suppressed LPS-mediated release of HMGB1 and HMGB1- mediated cytoskeletal rearrangements. Ginsenosides also inhibited HMGB1-mediated inflammatory responses. In addition, ginsenosides inhibited the production of tumor necrosis factor-α (TNF-α) and activation of protein kinase B (Akt), nuclear factor-κB (NF-κB), and extracellular-regulated kinases (ERK) 1/2 by HMGB1. Ginsenosides also decreased CLP-induced release of HMGB1, production of interleukin (IL) 1β/6, and mortality. These results suggested that ginsenosides may be potential therapeutic agents for treatment of vascular inflammatory diseases through inhibition of the HMGB1 signaling pathway.

        항공승무원의 관계적, 비관계적 속성이기내서비스 품질과 재구매 의도에 미치는 영향

        The airline industry plays a vital role in the development of tourism, whilst air travelers try to seek for the best experience in travelling without compromise. airlines are now trying to satisfy their customer’s special needs. The aim of the study is to analyse the effects of interpersonal and non-interpersonal attributes of flight attendant on in-flight service quality and repurchase intention. To meet the specific objectives of the research, a survey was carried out by means of an self-administered questionnaire to a sample of 320 individuals. To test the hypothesis, it was used descriptive, reliability and validity tests, confirmatory factor analysis and structural equation modeling. The major results of this study are as follows: First, non-interpersonal attribute had a positive effect but not interpersonal attribute on in-flight service quality. Second, in-flight service quality had a positive effect on repurchase intention. The findings of study suggest that the airlines should improve their employees’ performance in delivering excellent service quality to further enhance customer experience. This, in turn, will persuade customers to repeat their patronage of the airline and build positive word-of-mouth recommendations
