Methylotrophic Yeast, Pichia pastoris에서 사람 락토페린의 발현 및 항균성 연구

사람의 모유에 많이 함유된 human lactoferrin(hLF)은 항균 및 항 바이러스 작용이 있는 것으로 보고되고 있다. 본 연구에서는 hLf를 메탄올자화 효모인 Pichia pastoris에 cloning하고 그 발현을 RT-PCR, Northern blotting, SDS-PAGE및 Western blotting으로 확인하였다. 그 결과 2.1 kb의 hLf 유전자가 P.pastoris의 염색체 DNA로 끼어들어가 안정적으로 hLf를 발현하였다. 이 재조합 P.pastotis로부터 hLf를 포함하는 세포 추출액을 얻어 항균 작용을 연구하였다. 발현된 재조합 hLf는 Staphylococcus aureus, Micrococcus flavus 등의 그람 양성균에 대해 강력한 항균작용을 보일 뿐만 아니라 그람 음성 동물성 병원균인 Pseudomonas fluorescens ID 9631, E. coli ATCC8739, 25922,35 등과 Salmonella typhimurium 114,115 등 다양한 균에 대해서도 강력한 항균작용을 보였다. 이는 재조합 hLf가 생물학적 활성이 있다는 것을 보여준다. The expression and antibacterial. activity of recombinant human lactoferrin (hLf) was studied from methylotrophic yeast Pichia pastoris. The gene encoding hLf, isolated from human breast cDNA library, was subcloned into the expression vector, pPIC3.5K under the control of AOX1 promoter. The gene was integrated into the host chromosome and was identified by Southern blotting. The expression of the integrated gene was investigated by RT-PCR, Northern blotting, SDS-PAGE and Western blotting. Discrete band corresponding to hLf was detected from the SDS-PAGE, which was confirmed by Western blotting. The expression was also confirmed by RT-PCR and Northern blotting. The antibacterial activity of the recombinant hLf (rhLf) was investigated using Staphylococcus aureus ATCC 6538P and Micrococcus flavus ATCC 10240 as test organisms. The rhLf showed strong antibacterial activities against the bacteria. Furthermore, many Gram-negative animal pathogens such as E.coli ATCC8739, 25922, and Salmonella typhimurium 114 and 115, Pseudomonas fluorescens ID 963 I, P. aeruginosa KCCM 11802, and Gram-positive bacteria Bacillus mesentericus were also inhibited in their growth by the rhLf.

The thesis is automated traffic control equipment system aims to improve. Areas where improvement is needed about the existing automated traffic control equipment by applying the latest technology and to improve the things that can be derived. Reflecting the derived improvements, we intend to present a plan for the design and implementation of a new unmanned traffic control device. The main improvements were designed to change the housing material of the unmanned traffic control equipment, simplify the configuration of the equipment, reduce the weight of the equipment, and change the purpose of the software. In order to evaluate the objective performance of the improved unmanned traffic control equipment through this study, it was requested to a public certification authority. The reliability of the equipment was secured through KC certification and durability test. It is intended to present the feasibility of securing the marketability of the unmanned traffic control equipment by comparing and evaluating the construction period and installation cost with the existing unmanned traffic control equipment.

Meta-aramid and polyamic acid were separated and the manufactured films were analyzed for their integration and logarithmic properties. The miscibility of meta-aramid and polyamic acid was analyzed by Fourier transform infrared spectroscop and scanning electron microscopy. Using calorimetric analysis and differential scanning calorimetry, the storage of meta-aramid and polyamic acid, indicated on the right side of the column, was analyzed. It was confirmed that the initial thermal resistance occurs because the polyamic acid is accounted for in the meta-aramid, and the glass transition temperature and persistence phenomenon are explained.
도시철도 신호체계에 따른 폐색규정방식에 관한 상호비교연구

도시철도가 도입된 이후로, 국내 많은 도시철도 운영기관은 전기동차를 기반으로 도시철도 운전규칙을 제정하여 운영해 오고 있다. 하지만 도시철도 유관기관의 규정들이 실질적으로 도시철도 운전규칙과 적용하기에는 아직도 연구 할 부분이 남아있다. 특히 신호체계의 기술발달과 무인운전방식에 호응한 이동폐색에 대한 실제적인 연구가 부족한 현실이다. 각 운영기관들은 비록 본선영업운전 시 이례적인 상황이나 외란 등을 대비한 매뉴얼이나 운영조직 체계는 잘 정비되어 있지만, 신호시스템 발전을 기반으로 한 세밀한 법규정들이 지속적으로 필요한 시기인 것이다. 예를들면 무인운전방식은 경전철 운영에 있어서 미래적인 경향으로 대두 될 것으로 예상되며, 즉 좀더 구체적인 연구가 요구된다. 도시철도운영은 전적으로 도시철도 규정들을 얼마나 잘 효율적으로 내용과 절차들에 맞게 구성하느냐에 달려있다. 본 논문은 도시철도 운영기관의 대부분을 설명하고 있는 기존의 규정들을 수정하고 새롭게 신설 할 조항들을 연구하여 제시하는데 목적이 있다 Since the introduction of the subway, many operating companies in Korea have set up regulations of on a metro railroad designed an electric car. However, they haven’t yet tried to apply for driving regulations practically. In specially, they don’t practically change block system to catch up with developing signaling system and driverless driving. Although there are manuals for what to do and what organizations are as abnormal situations or degraded mode in operating companies, they need continuously to have specific laws based on signaling system to have been upgraded. For example, moving block system will become trend in driverless driving of light-rail commuter route, so, close examination of these problems is required. The subway operation depends wholly on how much well laws of on a metro are set up with practical measures and contents. This paper is intended to discuss the ways of making out or amending new and existing regulations, which account for the most ratio in this field.
