과학 고등학교 학생을 대상으로 한 DNA 재조합 기술 적용의 탐색

The rapid developments of a biotechnology in 20 century have resulted in enormous changes of our society as well as science. We have been living in the science-based society, and a genetic engineering among the biotechnology especially has affected various areas of our life. However, science high school students, anything but general high school ones, have not had chances of performing experiments for genetic engineering. Therefore, experiments related with recombinant DNA technology, a core of genetic engineering were applied to science high school students for 5 days in their school by their teacher not a professional researcher. All experiment were generally well done and students showed a significant increase of achievements (p<0.05). After doing all experimental procedures, they showed a high interest in a genetic engineering and an usual understanding in procedures of experiments. A little understanding for the experiments may be due to absence of pre-concepts about recombinant DNA and a little experimental experience of teacher. I suggest a proper time for performing the recombinant DNA experiments is after high school students fully obtaining basic concepts of recombinant DNA. Especially, plasmid isolation, DNA electrophoresis and transformation experiments among all procedures were more effective in science motivation and they are thought to be very important and useful within experimental courses for recombinant DNA technology. Therefore, I recommend the three experiments as core courses of recombinant DNA technology.

본 연구의 목적은 국내 여러 기관에 분산 소장 중인 편경 유물을 체계적으로 파악하고, 음향을 측정 분석하여 황종음고의 기본 진동수를 추정하는 것이다. 이를 위하여 국립고궁박물관, 국립국악원, 국립국악고등학교에 소장 중인 241매 유물 경석의 간지별, 율명별 분포를 파악하였다. 보다 타당한 대푯값을 찾기 위하여 17매의 유물 황종 경석에서 측정한 기본 진동수를 군집 분석하였으며, 간지별로 경석을 분류하여 삼분손익법으로 황종음고를 역추정하였다. 경석의 간지와 기록이 일치하는 계축년 간지의 경석 22개로 추정한 황종음고의 기본진동수는 266.9 Hz, 정유년 경석과 갑자년 경석으로 추정한 값은 262.4~262.5 Hz, 20세기에 해당하는 간지인 갑진년, 소화 12년, 소화 13년 경석으로 추정한 값은 258.7~259.3 Hz 등 시대에 따라 변해온 것으로 추정된다. 과거 유물에 대한 접근이 어려웠던 시대에 일부 유물로 황종음고를 추정한 것에 대해서 본 연구는 경석 유물 전수에 대한 음향 분석을 바탕으로 황종음고를 추정한 것에 의의가 있다. This study aimed to figure out the numbers and note distributions and sexagenary cycles of old pyeongyoungs systematically, and estimate the nominal frequency of whangjongeum, the Korean tradition pitch standard. As a total 214 old stones in the National Palace Museum, the National Kukak Center, the Kukak National High School were counted by notes and sexagenaries. The nominal frequencies of 17 old whangjong stones' sounds were categorized by cluster analysis method. Using nominal frequencies of stones according to their sexagenaries and Korean traditional intonation were used to estimate the nominal frequencies of the whangjong. The nominal frequency can be estimated by 22 Keychuk stones as 266.9 Hz, by Cheongyu and Gabja stones as 262.4~262.5 Hz, and by Gabjin, Sowha 12 and Sowha 13 as Estimating by 22 Kyechuk stones which were matched with the records. These results seem to be more reliable, because it is based on the whol samples of old pyeongyoungs, while the former studies have been based on couples of whangjong stones' sounds.

Inspite of students' curosity and social need for genetic engineering there are not actual experiments related with it in biology textbook of high school. A basic laboratory manual of recombinant DNA technology was developed to understand concepts of genetic engineering. The laboratory manual consisted of contents including basic microbial technology to manupulation of DNA. Using developed laboratory manual, it was applied to biology teacher of secondary school, Through activities of many experiments, they showed significant increase in understanding of concepts(p=0.000). Teachers agreeded with the need of experiments of recombinant DNA technology in science high school, However, comparing to scientifically gifted high school students biology, teachers were lower in achievement and interests about experimental activity. It suggests that teacher group need enough explanations and experimental time to understand concepts of recombinant DNA technology.

편경은 기역자형 경석 16개로 이루어진 국악기로 1과 1/3 옥타브의 음역을 가지며, 아악에서 표준 악기의 역할을 한다. 편경의 진동모드를 분석하기 위하여 현재 국립국악원에서 연주에 사용되고 있는 편경과 세종대왕기념관에서 소장 중인 편경 유물의 음향 스펙트럼을 분석하였다. 또한 황종과 청협종에 해당하는 경석의 진동모드형태를 가속도계, TV 홀로그램 및 충격망치를 이용하여 분석하였다. 국립국악원 편경의 16개 경석에서 나타난 기명진동수는 경의 두께에 따라 증가하며, 삼분손익법에 따른 음률에 충실하게 조율되었다고 할 수 있다. 세종대왕기념관 편경의 16개 경석에 적혀있는 제작연도를 나타내는 간지는 서로 다르며, 12율려에 맞게 조율되었다고 하기 어렵다. 세종대왕기념관에 소장된 편경의 진동수는 국립국악원 편경과 최소가지차이 (just noticeable difference) 이상의 차이를 나타냈다. 진동모드형태는 경석의 두께와 무관하게 일정하다. Korean pyeongyeong, a set of sixteen L-shape chime stones covering one and one third octaves, is a standard instrument in the Korean traditional court music. We analyze the vibrational mode frequencies in a pyeongyeong replica which is played at the National Center for Korean Traditional Performance Arts and pyeongyeong remains which are exhibited at King Sejong Memorial Museum. The modal shapes on the Whangjong, the 1st stone and Cheonghyurjong, the 16th stone mapped by scanning accelerometer, TV holography and STAR system. The nominal frequencies in pyeongyeong replica at the National Center for Korean Traditional Performance Arts increase linearly with the thickness of the stones and the tones are tuned in line with the musical scale of Sambunsonik. The sexagenary cycles on the pyeongyeong remains at King Sejong Memorial. which show the Year of product indirectly, are different each other and the tones are not tuned in scale. The relative frequency ratios of each modes on stones differ more than just-noticeable differences from those on the pyeongyeong replica. Modal shapes are same for the two stones regardless of the thickness.
