        Ultrafine particles of Ulmus davidiana var. japonica induce apoptosis of gastric cancer cells via activation of caspase and endoplasmic reticulum stress

        Small-sized particles are more suitable for targeteddelivery and are therapeutically more effective thanlarge-sized particles. In this study, we investigated theanticancer effects of ultrafine particles of Ulmus davidianavar. japonica (ufUJ) on human gastric cancer cell linesSNU-1, SNU-216, and SNU-484. ufUJ induced apoptosisby the proteolytic activation of caspase-9, caspase-6, andcaspase-3 and cleavage of poly (ADP-ribose) polymerase. The expression levels of the endoplasmic reticulum stressrelatedprotein BiP markedly increased after ufUJ treatment. BiP knockdown decreased ufUJ-induced cell death. ufUJ-induced apoptosis was inhibited by the caspase-3inhibitor z-DEVD-fmk, caspase-6 inhibitor z-VEID-fmk,and caspase-9 inhibitor z-LEHD-fmk, and by siRNAsagainst caspases 3, 6, and 9. Gastric cancer cells did notshow anchorage-independent growth in the presence ofufUJ. However, cells treated with caspase inhibitorsshowed an enhanced colony-forming ability. These findingsmay be helpful in the prevention of gastric cancer andin the development of functional foods.

        토복령 추출물 첨가로 인한 돼지갈비 소스의 품질 특성

        본 연구는 토복령 추출물의 항산화 활성 및 저장성 증진 가능성을 보고자 돼지 갈비소스의 5주간의 품질 변화를 조사하였다. 저장 4주째에 총 균수는 대조군과 0.25% 첨가군에서 0.5%와 0.75% 첨가군에 비해 높은 수치를 나타내어, 토복령 추출물 첨가량이 많아질수록 총 균수는 낮아짐을 알 수 있었다. DPPH 라디컬 소거 활성은 첨가량이 많아질수록 증가하였으며, 토복령 추출물 0.75% 첨가군은 유의적으로 높은 소거 활성을 나타내었다(p<0.05). 또한, 토복령 추출물 첨가량이 증가할수록 TBARS는 낮아졌으며, 0.5%와 0.75% 첨가군은 유의적으로 낮은 수치를 보였다(p<0.05). 본 실험 결과로 토복령 추출물의 항산화 활성을 통한 기능성 향상과 항균 효과에 의한 저장성 강화로 식품 첨가 보존제로서의 개발 가능성이 있을 것으로 사료된다. The present study was designed to study the antioxidant activity and physicochemical and microbiological characteristics of Pork Kalbi sauce added with Smilax china L. extract(0%, 0.25%, 0.5% and 0.75%) for 5 weeks. Total microbial counts were slightly higher in control and 0.25%-added group, therefore, more addition of Smilax china L. extract lowered total microbial counts. Alpha-diphenyl-β-picrylhydrazyl(DPPH) radical scavenging activity was significantly higher in 0.75%-added group compared to those in others(p<0.05). Also, thiobarbituric acid reactive substances(TBARS) was significantly lower in 0.5% and 0.75%-added groups compared to others(p<0.05). These results indicated that Smilax china L. extract addition increased the antioxidant activity of Pork Kalbi sauce and could improve the shelf-life during storage.

        비쭈기 나무(Cleyera japonica) 추출물의 생리활성에 대한 연구

        적 요본 연구에서 비쭈기 나무의 잎 부분에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 비쭈기 나무 잎 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 비쭈기 나무 추출물 처리 시 항산화(IC50, 22.90 ㎎/L) 및항균활성이 매우 우수하게 나타났으며 특히 그람양성세균에 대한 항균활성이 높았다. 또한, 비쭈기 나무 추출물 처리시tyrosinase 활성저해 및 멜라닌 함량 저하(IC50, 101.90 ㎎/L)를보여주었다. 이와 같은 결과로 미루어 볼 때 비쭈기 나무 추출물은 피부미백 소재 등 피부개선 효과를 비롯한 기능성 화장품에활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단된다 In this study, we investigated the applicability of functional materials by examining a variety of physiological activities with the extract of Cleyera japonica leaf. Cleyera japonica extract showed a low cytotoxicity against murine melanoma B16F10 cells. In little or no cytotoxicity at concentrations, we showed that the treatment with Cleyera japonica extract resulted in a significant increase in the DPPH radical scavenging activity (IC50, 22.90 ㎎/L), similar to ascorbic acid (IC50, 18.65 ㎎/L) and anti-microbial activities against Bacillus subtilis, Escherichia coli, and Candida albicans. In particular, anti-microbial activities against Gram-positive bacteria was high. These results suggest that Cleyera japonica extract could be used as a natural preservative. Additionally, Cleyera japonica extract showed the inhibition of tyrosinase activity (IC50, 178.90 ㎎/L), similar to kojic acid (IC50, 89.13 ㎎/L) and decreased melanin content (IC50, 101.90 ㎎/L) higher than the control arbutin level (IC50, 100.65 ㎎/L), especially. Therefore, these results indicate that Cleyera japonica extract may be an effective material for functional cosmetics such as skin whitening materials.

      • 찰옥미 조청을 첨가한 기능성 고추장의 이화학적 특성

        Objective:The present study was designed to examine the change of physicochemical properties of kochujang when waxy corn syrup was added at 0, 10 and 20%. Methods:Physicochemical properties including pH, titratable acidity, salinity, amino-nitrogen and reducing sugar content, viscosity and color values were measured and sensory evaluation was conducted. Result & Conclusion:The salinity was higher and amino-nitrogen and reducing sugar contents were lower in 20% waxy corn syrup-added group than in control. Viscosity increased slightly and L and b values decreased in experimental groups. Sensory evaluation showed that sweetness and viscosity increased whereas saltiness decreased with waxy corn syrup addition. The overall quality value was the highest in 20% waxy corn syrup-added group. The present study showed the possibility of functional kochujang manufacture with waxy corn syrup addition.

      • 느릅나무 근피 추출물의 항산화 활성 및 DNA 손상억제효과

        Objective: The present study was designed to examine the antioxidant activities and the protective effect on DNA damage of the extracts of Ulmus devidiana var, japonica. Methods: The 1,1- diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl scavenging assay and Fe2+ chelating assay were performed for antioxidative effect and φX-174 RF I DNA cleavage assay and intracellular DNA damage assay were used for inhibitory effect of intracellular DNA damage. Results: The EtOAC extracts at 200 ug/ml showed the highest acitivities of 1,1-diphenyl-2- picrylhydrazyl (DPPH) radical and hydroxyl scavenging and Fe2+ chelating, which were 90.82, 65.86, and 52.42%, respectively. The slightly lower activities were found in hot-water with a similar pattern. The significant protective effect of DNA damage against oxidative stress was found from both EtOAC extracts and hot-water extracts. 3,5-bis(t-butyl) phenol, 1,3-benzenediol, Benzeneacetic acid, Aspidinol and Benzoic acid were identified by GC/MS analysis. Conclusion: This study showed that the extracts of Ulmus devidiana var, japonica can be utilized as an effective antioxidants and protective effect of DNA damage against the oxidative stress.

      • 녹용 추출액을 첨가한 양갱의 품질 특성

        Objective: This study was conducted to investigate the rheological and sensory characteristics of yanggeng supplemented by deer antler extract. Methods: The yanggeng samples were prepared by supplementation with different concentrations of deer antler extract (0, 10, 20, and 30%), and Hunter``s color, texture profiles and sensory attributes were examined. Results: In color analysis, lightness of yanggeng samples tended to be decreased and the redness and yellowness values increased when concentrations of deer antler extract supplementation increased. In texture profiles, the cohesiveness and gumminess of samples supplemented by 20% deer antler extract were higher than those of other samples. In sensory evaluation, color scores of 10% deer antler-supplemented yanggeng were higher than those of 20% and 30%-supplemented samples. The overall acceptability of 20% deer antler-supplemented yanggeng showed the highest value among other samples. Conclusion: Based on the data obtained from the present study, it was concluded that the 20% of deer antler extract could be used to produce deer antler extract-supplemented yanggeng without adverse effects on rheological and sensory characteristics.

      • 달맞이꽃 종자유를 첨가한 호상요구르트의 이화학적 변화와 물성 및 관능적 특성에 관한 연구

        본 연구는 β-cyclodextrin 처리를 이용하여 우유에서 cholesterol을 제거하여 호상요구르트를 제조하고 혈중 콜레스테롤 저하 물질인 달맞이꽃 종자유(EPO)의 양을 다르게 첨가하여 호상요구르트의 저장기간 중 이화학적 변화와 물성 및 관능적 특성을 살펴보았으며, 그 결과 pH는 저장기간 15일이 경과함에 따라 모든 군에서 pH의 저하가 일어났고, 각 실험군 간 유의적 차이는 없었다. 마찬가지로 산도도 저장기간 15일이 경과함에 따라 모든 실험군에서 계속적인 증가가 나타났으며, 실험군간 유의적 차이는 없었다. 지방산화도측정결과 TBA가(535nm)는 저장 0일에 0.045에서 저장 15일에 0.196까지 증가하였고, EPO 첨가 호상요구르트는 0.188까지 증가하였으나 실험군간 유의적 차이는 보이지 않았다. 점도는 β-CD를 처리하여 2% EPO를 첨가한 호상요구르트가 저장 0일에 3.7에서 저장 15일에 8.4로 가장 큰 점도 감소를 나타내었고, 저장 15일 동안 모든 군에서 점도감소가 나타났다. 관능검사결과 β-CD처리하여 EPO를 첨가한 호상요구르트의 산패취와 산패한 맛이 control보다 높았지만 저장기간이 지날수록 점차 낮아졌고, 신맛과 쓴맛은 control과 유사하게 나타났다. 결론적으로 EPO를 첨가한 호상요구르트는 이화학적 변화와 물성 및 관능적으로 긍정적인 결과로 나타나 산업화의 가능성을 보였다.
