보조 형광증가제를 이용한 L-Dopa의 형광분광법적 정량

리간드 증감 유발 형광법을 이용하여 Tb(III)-L-dopa (L-3,4-dihydroxyphenyl alanine) 착이온의 방출세기를 측정함으로써 수용액 중의 L-dopa를 정량하는 방법에 대하여 연구하였다. 들뜸파장, pH, 보조 형광증가제의 선택, Tb(III) 이온의 농도, 보조 형광증가제로 사용된 Lu(III) 이온의 농도 및 방출파장의 방출세기에 대한 영향을 조사하였다. 보조 형광증가제로서 Lu(III) 이온을 첨가하였을때 Tb(III) 이온의 방출세기가 현저히 증가함을 관찰하였고, L-dopa의 검출한계를 낮출 수 있었다. 보조 형광증가제를 첨가하지 않았을 경우에 L-dopa 검정곡선의 직선감응범위는 들뜸파장, pH 및 Tb(Ⅲ) 이온의 농도가 각각 300 nm, 8.0 및 $1.0{\times}10^{-4}$ M였을때, $5.0{\times}10^{-7}$ M~$1.0{\times}10^{-4}$ M였다. 이 조건에서의 검출한계는 $4.0{\times}10^{-8}$ M였다. 보조 형광증가제를 첨가하였을 경우에는 들뜸파장, pH, Tb(III) 이온의 농도, 보조 형광증가제로 사용된 Lu(III) 이온의 농도 및 방출파장이 각각 300 nm, 8.5, $1.0{\times}10^{-5}$ M, $1.0{\times}10^{-5}$ M 및 545 nm였을 때, 직선감응범위가 1.0×$10^{-8}$ M~2.0{\times}10^{-4}$ M였고, 이 때의 검출한계는 $1.0{\times}10^{-9}$ M였다. Methods to determine L-dopa(L-3,4-dihydroxyphenylalanine) in aqueous solution by spectrofluorimetry based upon the ligand sensitized luminescence of Tb(III) ion L-dopa complex have been studied. Tb(III) ion and Lu(III) ion were used as ligand sensitized fluorescencer and co-fluorescence enhancer, respectively. The effects of excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Tb(III) ion L-dopa complex was further increased with addition of Lu(III) ion. The calibration curve for L-dopa was linear over the range from 5.0 ${\times}$ $10^{-7}$ M to 1.0${\times}$ $10^{-4}$ M and the detection limit was 4.0 ${\times}$ $10^{-8}$ M under the optimal experimental conditions of 300 nm, 8.0, 1.0 ${\times}$ $10^{-4}$ M and 545 nm for excitation wavelength, pH, concentration of Tb(III) ion and emission wavelength, respectively. When Lu(III) ion was added to Tb(Ⅲ) ion L-dopa complex, the concentration range of linear response and detection limit were 1.0${\times}$$10^{-8}$ M to 2.0 ${\times}$ $10^{-4}$ M and 1.0 ${\times}$ $10^{-9}$ M, respectively under the optimal experimental conditions of 300 nm, 8.5, 1.0 ${\times}$ $10^{-5}$ M, 1.0 ${\times}$ $10^{-5}$ M, 545 nm for excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength, respectively.

Stability and durability of the artificial reefs(ARs) for wildstock enhancement have been a key issue. We surveyed the stability of 8,740 artificial reefs (80 group AR's) installed at the Gangwon coast of the East Sea (Sea of Japan) through which 1 to 3 typhoons, wave heigh are passing annually. The ARs surveyed were 2 types aging 1996 to 2005 years. The side scan sonar(SSS) and scuba diving surveys confirmed that the ARs stability was age and type-dependent. A case of a confirmation amount to 84.8% ARs remained safe, the rest 15.2% being influenced by a physical parameter of the waters. The representative influences by their circumstances were of capsizing (8.1%), breakage (7.1%). A group of a AR's to 66.3% ARs remained safe, the rest 33.7% being influenced by a physical parameter of the waters. The representative influences by their circumstances were of sink (8.8%), burial (15.0%) and both of two Phenomenon(sink, burial) (10.0%).

Stability and durability of the artificial reefs (ARs) installed for wildstock enhancement .have been a key issue. We surveyed the stability of 4,044 artificial reefs installed at 5 sites (Gangeung, Donghae, Samcheok, Goseong, and Yangyang) along the Gangwon coast of the East Sea (Sea of Japan) through which 1 to 5 typhoons pass annually. The ARs surveyed were 7 types aging 1 to 4 years. The side scan sonar (55S) and scuba diving surveys confirmed that the ARs stability was age and type-dependent. A total of 85.56% ARs remained safe, the rest 14.44% being influenced by physical parameters of the waters such surf. An interesting observation was that the AR stability was apparently influenced by the next year's wave after installation rather than by age. The representative influences by their circumstances were capsized (0.20%), broken (5.79%), sunk (0.27%), and buried (8.18%). By type, dice reef was most stable, while semicircle branch reef was most vulnerable.

목적: 본 연구는 한국형 전산화인지재활 프로그램(CoTras)이 지역사회에 거주하는 뇌졸중 환자의 인지기능과 기본적·수단적 일상생활활동에 미치는 효과를 알아보기 위하여 실시하였다. 연구방법: 본 연구는 2017년 4월부터 동년 7월까지 총 10주 동안 경북 G시에 거주하는 뇌졸중 환자 22명을 대상으로 실시하였다. 한국형 전산화인지재활 프로그램(CoTras)을 중재한 실험군 11명, 전통적인 인지치료를 중재한 대조군 11명으로 나누어 주 2회 30분씩 총 10주간 중재를 실시하였다. 중재 전·후 인지기능을 비교하기 위하여 MMSE-K와 LOTCA를, 기본적·수단적 일상생활활동을 비교하기 위하여 K-MBI와 K-IADL을 각각 평가하였다. 결과: 중재 후 실험군은 대조군에 비하여 통계학적으로 유의하게 인지기능(MMSE-K, LOTCA)과 기본적 일상생활활동(K-MBI)이 향상되었다. 결론: 한국형 전산화 인지재활 프로그램(CoTras)는 지역사회에 거주하는 뇌졸중 환자의 인지기능과 기본적 일상생활활동 향상에 효과적임을 확인할 수 있었다. Objective: The purpose of study was to verify the effect of a Korean computer-based cognitive rehabilitation program(Cotras) for recovering the cognitive function and basic·instrumental activities of daily living in community stroke patients. Methods: This study was conducted at G city from April 2017 to July with community stroke patients. Eleven patients with experimental group given Korean computer-based cognitive rehabilitation program(Cotras) intervention, eleven patients with control group given traditional cognitive therapy for 30 minutes per session two times a week for total ten weeks. Before and after intervention, we compared cognitive function and basic·instrumental activities of daily living we undertook MMSE-K, LOTCA, K-MBI and K-IADL. Result: Experimental group presented significant statistical difference more improve cognitive function(MMSE-K, LOTCA) and basic activities of daily living(K-MBI) compared to control group(p<.05). Conclusion: Korean computer-based cognitive rehabilitation program(Cotras) was improve cognitive function and basic activities of daily living for community stroke patients.
Bacillus subtilis 저온충격 단백질 Csp 유전자 결손이 저온 환경에서의 생장에 미치는 영향

Cold shock proteins (CSPs) are expressed in cold conditions in order to adapt to that cold conditions. Three cold shock proteins CspB, CspC and CspD exist in Bacillus subtilis. To know the effect of cold environment on the cell growth each three cold shock gene was deleted. Growth curves in the conditions of 37℃ and 20℃ were compared to understand cold acclimation of these deleted mutant cells. Also, growing cells in the conditions of 37℃ to the log phase and then cold shock treatment which growing temperature was changed from 37℃ to 15℃ was carried out. The results of cold acclimation showed that both of the growth rate of wild type and Csp deleted mutants at 37℃ is nearly same but the growth rate of mutant cells was reduced compared to the wild type cell. On the other hands, both of wild type and mutant cells show nearly same growth rate on cold shock. These results demonstrate that Csp deleted mutants are not cold sensitive.
