        As an endemic species in Korea Adenophora erecta S. Lee, Joongku Lee et S. Kim is distributed in a restricted area of Ulleung island. In this study, we confirmed finding only about 60 individuals in Taeha-ryeung in an area measuring 900 m² in size. These data together with previously collected data indicate that this species is in the “Critically Endangered (CR)” category based on taxon evaluation criteria of the IUCN. We propagated this species using seeds of transplanted plant from a site where this plant was first found. The best seed germination was observed in a batch treated with GA3 (500 mg/L) under the sun light and the best plant growth was observed in a bed of nursery soil and rough-sand soil (1:1). The original habitat of A. erecta in Sukpo-dong was successfully restored by transplantation of the propagated young plants.

      • Decal transfer process using bonding layer for hydrocarbon-based membrane electrode assemblies

        In this study, a hydrocarbon membrane-based membrane electrode assembly (MEA) was fabricated by decal transfer process using bonding layers. Sulfonated poly(arylene ether sulfone) (SPAES) used as a hydrocarbon- based polymer is difficult for applying to the MEA made up of Nafion based electrodes because of their incompatibility and the rigid structure of SPAES. Therefore, we introduce the concept of the bonding layer to improve the low interfacial stability between the SPAES membrane and the Nafion based-electrodes. The bonding layer was synthesized as the SPAES-based structure and dihydroxy diphenyl ether (DHDPE) replaced biphenol (BP) to increase its flexibility through the ether groups. Hexafluoro bisphenol A (6F) was also added to increase the chemical affinity of the electrode.

        솔방울 에틸아세테이트 분획물의 항산화 및 산화적 DNA 손상 억제 활성

        Antioxidant activity and inhibitory effect on oxidative DNA damage of ethyl acetate fractions extracted from Cone of Red Pine (Pinus densiflora) were investigated to find utilization of Cone, by-product of Red Pine, thrown out after berry shatter, as a new natural plant resource. Cone from P. densiflora was extracted with methanol (MeOH) and separated to petroleum ether, ethyl acetate and water fraction. Among them, ethyl acetate fraction was used. The antioxidant activity was conducted by the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, 2, 2'-Azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) radical scavenging assay, Fe 2+ chelating assay and reducing power assay. The inhibitory effect on oxidative DNA damage was determined by DNA cleavage assay using φX-174 RF I plasmid. The results of DPPH and ABTS radical scavenging activity at 200 ㎍/㎖ of extracts were 86.50% and 95.80% respectively, which were similar figures compared with L-ascorbic acid as control. Fe 2+ chelating activity was 77.96% and reducing power was 0.77 at 200 ㎍/㎖. Total phenolic component was 27.29±0.3 ㎎/g and Vitamin C content was 1.84±0.1 ㎎/g. Also ethyl acetate fraction from Cone has inhibitory effect, using φX-174 RF I plasmid on DNA cleavage assay. In conclusion, Cone, by-product of P. densiflora, showed high antioxidant activity and inhibitory effect on oxidative DNA damage. Therefore this study suggests Cone, useless by-product, can be developed as a new natural plant resource with lots of utilization such as an effective antioxidant, natural medicine, food, cosmetics and so on. Key words - Pinus densiflora, DPPH, Reducing power, Fe 2+ chelating 본 연구에서는 솔방울을 새로운 천연 식물자원으로서의 이용 가치를 평가하기 위해 솔방울 ethyl acetate 분획물의 항산화 활성과 DNA 손상억제 효과를 조사하였다. 솔방울 ethyl acetate 분획물의 DPPH, Fe2+ 킬레이팅 및 환원력 활성은 대조군인 L-ascorbic acid와 유사한 활성을 나타내었다. 특히 ABTS 소거 활성의 IC50값은 L-ascorbic acid (10.49 ㎍/㎖)에 비해 솔방울 ethyl acetate 분획물(7.80 ㎍/㎖)이 더욱 높은 활성을 보였다. 또한 총페놀성 화합물과 비타민 C 함량은 각각 27.3 ㎎/g, 1.8 ㎎/g로 나타났다. Hydroxyl radical과 fenton reaction에 의한 산화적 DNA 손상억제 효과는 모든 농도에서 50% 이상의 높은 손상 억제 효과를 나타냈다. 따라서 본 연구결과를 통한 솔방울 ethyl acetate 분획물의 항산화 활성과 산화적 DNA 손상 억제 효과 입증은 다소 부가가치가 낮게 평가되어온 솔방울은 식품이나 천연 의약품 분야에 새로운 천연소재로써의 활용가치가 높을 것으로 판단된다.

        Edge protection using polyacrylonitrile thin-films for hydrocarbon-based membrane electrode assemblies

        A hydrocarbon membrane-based membrane electrode assembly (MEA) is fabricated by attachingpolyacrylonitrile (PAN) films to a sulfonated poly(arylene ether sulfone) (SPAES) membrane to improvethe durability of a proton exchange membrane fuel cell (PEMFC) under cyclic hydration and dehydration. The edge protective PAN film is prepared by bar-coating to be extremely thin (6 1 mm thick) in order tominimize the increase in the MEA resistance. This study focuses on comparing the pristine membrane withthe membrane using the PAN film to identify the effect of the edge protective film. Therefore, durability testwith hydration–dehydration cycles is conducted to evaluate it

      • PlantGPS: A Mobile Application for Collecting Biogeographical Information of Plants and for Their Monitoring

        Cost for plant survey has been dramatically reduced due to the development of mobile technology to obtainpictures with their GPS coordination. We developed a PlantGPS system consisting of an Android applicationspecialized for collecting plant pictures and their locations using mobile devices, a server-side component foruploading data, and a web-based interface for managing and analyzing data. As examples of application of ourPlantGPS system, surveys of “plants in Nangsae (Daksum) Island” and “ferns in Chollipo arboretum” wereconducted. Results showed that our PlantGPS system could be used to quickly survey plant distribution inrestricted area with reasonable time and effort.

        멸종위기 수생식물인 매화마름(Ranunculus kazusensis Makino)의 기내 신초 증식

        본 연구는 자생 멸종위기 수생식물인 매화마름의 기내 증식에 미치는 배양환경(식물생장조절제, AC, sucrose)을 조사하기 위해 수행되었다. 효율적인 캘러스 유도조건을 알아보고자 IAA, NAA, IBA와 2,4-D가 각각 1 mg/L 첨가된 MS배지에 줄기절편을 치상한 후에 캘러스 형성율을 조사한 결과, NAA를 첨가한 배지에서 87%의 캘러스 유도율을 보였으며, IBA가 첨가된 배지에서 78%, 2,4-D가 첨가된 배지는 42%, IAA 첨가 배지는 34%의 순으로 조사되었다. NAA 처리 농도에 따른 캘러스 유도율은 0.5 mg/L의 NAA처리구에서 87.5%로 조사가 되었으나 1 mg/L의 NAA처리구와 유의적으로 차이를 보이지 않았다. 매화마름 캘러스 유래의 다신초 유도는 1.0 mg/L의 BA와 0.5 mg/L NAA 조합처리구에서 절편당 평균 47.6개로 가장 많이 유도되었다. 호르몬이 처리되지 않는 대조구에서는 캘러스 상태로 계속 유지되었다. 2,4-D의 조합처리는 절편당 평균 10개 미만의 다신초가 유도되었다. 매화마름 캘러스 유래의 다신초 신장에 미치는 AC (Activated charcoal)와 sucrose의 농도를 조사한 결과, 엽장(shoot length)은 200 mg/L AC처리구에서 평균 12.6 cm로 가장 긴 반면, 대조구는 평균 8.3 cm로 짧았다. 결과적으로 본 연구는 매화마름의 기내증식에 미치는 몇 가지 요인에 관한 구체적인 결과를 제시하였고, 이러한 결과는 향후 매화마름의 증식 및 보전전략 개발에 중요한 기초자료로 제공 될 것이다. The most suitable conditions for shoot growth after 4 weeks of culture were the MS medium with AC and sucrose. This in vitro propagation protocol will be valuable for conservation and mass propagation of this endangered plant.

When stem segments were cultured on MS media supplemented with 1.0 mg/L IAA, NAA, IBA and 2,4-D, the highest frequency of callus induction was achieved on MS medium supplemented with 1.0 mg/L NAA. Multiple shoot per explant was obtained, the MS medium containing 1.0 mg/L BA and 0.5 mg/L NAA. Additionally, effect of activated charcoal (AC) and sucrose on shoot growth in in vitro culture were examined. The most suitable conditions for shoot growth after 4 weeks of culture were the MS medium with AC and sucrose. This in vitro propagation protocol will be valuable for conservation and mass propagation of this endangered plant.
