We have cloned a cDNA encoding the VIb subunit of cytochrome c oxidase. The cDNA consists of 488 nucleotides encoding 86 predicted amino acid residues. The sequence was shown to be identical to the coxVIb subunit isolated from a skeletal muscle cDNA library (Taanman, J. W. et al., Gene, 93. 285, 1990) not only in the coding region but also in both 5' and 3' untranslated regions. Hydropathy profile of the thymic VIb protein reveals that the polypeptide is highly hydrophilic, but there are two hydrophobic regions. These hydrophobic regions appear to associate with the inner membrane of mitochondria.
ISOLATION AND CHARACTERIZATION OF A cDNA FROM COLD-INDUCED BRASSICA OLERACEA

A cDNA library was constructed from the cold induced B.o leracea leaf and a putative cold induced cDNA, BOCOR, was isolated by the PCR method. A PCR product was cloned into a Bluescript vector. Nucleotide sequence analysis revealed that BOCOR was consisted of 457 bp nucleotides with deduced amino acids of 142 residues. BOCOR showed a high degree of homology to the cold-regulated winter Brassica napus cDNA (BN115). When PCR was carried out using the reverse-transcribe mRNAs from cold-induced and control plants. the 457 bp DNA was amplified only from the cold-induced transcripts. It will be interesting to test if the DNA encodes a cold-regulated protein. Genomic PCR shows that the BOCOR may contain at least one intron in its open reading frame. In an effort to generate cold-resistant transgenic plants, cloning of the cDNA into a plant expression vector is in progress.

자궁 경부암 환자의 생검 조직으로부터 HPV DNA 존재와 c-myc 증폭을 조사하였다. Southern blot과 in situ hybridization 결과로부터 HPV DNA는 integrated form으로 존재하며, HPV-16/18형은 cervical intraepithelial neoplasia(CIN)에 비해 condyloma에서 더 많이 발견되었고, HPV-16/18, 31/33/35형은 condyloma와 CIN에서 똑같은 빈도로 발견되었다. 또한 자궁경부암 환자 7명 모두에서 c-myc이 증폭되었으며, 통제군에 비해 최고 26배까지 증폭되기도 하였다. 이러한 결과들로부터 자궁경부암 발병에 있어서 HPV와 c-myc이 중요할 역할을 할 것임을 알 수 있다.
형질전환체 담배에서 사람 파필로마 바이러스 HPV-18 E6 유전자의 발현

담배에서 human papil1omavirus-18형 E6(HPV-18 E6) oncogene을 발현시키기 위하여 E6 유전자를 Cauliflower mosaic virus(CaMV)의 35S dual promoter와 tobacco etch virus(TEV) 5'-untranslated region의 조절을 받도록 벡터를 제조한 후, Agrobacterium-mediated leaf disc 방법으로 담배에 도입하였다. Kanamycin 저항성을 갖는 형질전환체를 선별하고, 완전한 식물체로 재생한 형질전환체의 잎조직에 대하여 PCR 방법과 Northem blot, Westem blot 분석을 한 결과, HPV-18 E6 유전자가 형질전환체 담배에서 성공적으로 발현됨을 알 수 있었다. To develop a HPV-18 oral vaccine, HPV-18 E6 gene was cloned into an expression vector, under the control of CaMV 35S dual promoter and 5'UT region of TEV. The constructed vector (pUS-E6) was used to transform tobacco plants by the Agrobacterium-mediated leaf disc method. The transformants were then regenerated to whole transgenic plants. The HPV-18 E6 gene was found to be expressed in the transgenic tobacco plants as determined by PCR, RT-PCR, Northern analysis and Western analysis. These results show a possibility that HPV oral vaccine may be developed by expressing the HPV proteins in tomato or other vegetables.
