        Induction of cytochrome P450 (CYP) 1A1 and CYP1B1 by environmental xenobiotic chemicals or endogenous ligands through the activation of the aryl hydrocarbon receptor (AhR) has been implicated in a variety of cellular processes related to cancer, such as transformation and tumorigenesis. Here, we investigated the effects of the adti-diabetes drug metformin on expression of CYP1A1 and CYP1B1 in breast cancer cells under constitutive and inducible conditions. Our results indicated that metformin down- regulated the expression of CYP1A1 and CYP1B1 in breast cancer cells under constitutive and 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD)-induced conditions. Down-regulation of AhR expression was required for metformin-mediated decreases in CYP1A1 and CYP1B1 expression, and the metformin-mediated CYP1A1 reduction is irrelevant to estrogen receptor a(Era) signaling. Furthermore, we found that metformin markedly down-regulated Sp1 protein levels in breast cancer cells, The use of genetic and pharmacological tools revealed that metformin-mediated down-regulation of AhR expression was mediated through the reduction of Sp1 protein. Metformin inhibited en-dogenous AHR Ligand-induced CYP1A1 and CYP1B1 expression by suppressing tryptophan-2,3-dioxygenase (TDO) expression in MCF-7 cells. Finally, metformin inhibits TDO expression through a down-regulation of Sp1 and glucocorticoid receptor(GR) protein levels. Our findings demonstrate that metformin reduces CYP1A1 and CYP1B1 expression in breast cancer cells by down-regulating AhR signaling. Merformin would be able to act as a potential chemopreventive agent against CYP1A1 and CYP1B1-mediated carcinogenesis and development of cancer. ⓒ2014 Elsevier lnc. All rights reserved.

        Piperine is a bioactive component of black pepper, Piper nigrum Linn, commonly used for daily consumption and in traditional medicine. Here, the molecular mechanisms by which piperine exerts antitumor effects in HER2-overexpressing breast cancer cells was investigated. The results showed that piperine strongly inhibited proliferation and induced apoptosis through caspase-3 activation and PARP cleavage. Furthermore, piperine inhibited HER2 gene expression at the transcriptional level. Blockade of ERK1/2 signaling by piperine significantly reduced SREBP-1 and FAS expression. Piperine strongly suppressed EGF-induced MMP-9 expression through inhibition of AP-1 and NF-κB activation by interfering with ERK1/2, p38 MAPK, and Akt signaling pathways resulting in a reduction in migration. Finally, piperine pretreatment enhanced sensitization to paclitaxel killing in HER2-overexpressing breast cancer cells. Our findings suggest that piperine may be a potential agent for the prevention and treatment of human breast cancer with HER2 overexpression. ⓒ2013 Elsevier Ltd. All rights reserved.

        Parenchymal-sparing anatomical hepatectomy based on portal ramification of the right anterior section: A prospective multicenter experience with short-term outcomes

        Backgrounds/Aims: Parenchymal-sparing anatomical hepatectomy (Ps–AH) based on portal ramification of the right anterior section (RAS) is a new technique to avoid unnecessarily transecting too much liver parenchyma, especially in cases of major anatomical hepatectomy. Methods: We prospectively assessed 26 patients with primary hepatic malignancies having undergone major Ps–AH based on portal ramification of the RAS from August 2018 to August 2022 (48 months). The perioperative indications, clinical data, intra-operative index, pathological postoperative specimens, postoperative complications, and follow-up results were retrospectively evaluated. Results: Among the 26 patients analyzed, there was just one case that had intrahepatic cholangiocarcinoma The preoperative level of α– Fetoprotein was 25.2 ng/mL. All cases (100%) had Child–Pugh A liver function preoperatively. The ventral/dorsal RAS was preserved in 19 and 7 patients, respectively. The mean surgical margin was 6.2 mm. The mean surgical time was 228.5 minutes, while the mean blood loss was 255 mL. In pathology, 5 cases (19.2%) had microvascular invasion, and in the group of HCC patients, 92% of all cases had moderate or poor tumor differentiation. Six cases (23.1%) of postoperative complications were graded over III according to the Clavien–Dindo system, including in three patients resistant ascites or intra-abdominal abscess that required intervention. Conclusions: Parenchymal-sparing anatomical hepatectomy based on portal ramification of the RAS to achieve R0-resection was safe and effective, with favorable short-term outcomes. This technique can be used widely in clinical practice.

        One-Pot Preparation of Antibacterial Electrospun Polycaprolactone Membrane Embedded with Gamma Irradiation-Induced Silver Nanoparticles

        In this study, we proposed a straightforward electrospun polycaprolactone (PCL) loaded with silver nanoparticles (SNPs)membrane fabrication process, in which SNPs were directly synthesized from silver nitrate (AgNO3) in PCL–acetone mixtureby gamma irradiation. The insolubility of AgNO3in PCL solution was solved using an auxiliary dimethyl sulfoxide solvent. As a physical approach, gamma rays readily converted silver ions into SNPs without the addition of harmful reductionagents, which reduced the cytotoxicity of the synthesized material. By avoiding some processes such as purification, solventremoval, or redispersion of SNPs, this method was more time-saving compared to other related studies. SNPs formation wasconfirmed by both UV–Visible spectrum (UV–Vis) and X-ray diffraction analysis. Scanning electron microscopy (SEM)revealed that the addition of SNPs significantly reduced the fiber diameter of PCL–Ag membranes compared to that of rawPCL. Uniform spherical-shaped SNPs incorporated in PCL fibers were observed under transmission electron microscopy(TEM). The tensile test showed that the electrospun PCL–Ag membranes exhibited good mechanical characteristics. Moistureeasily penetrated the porous microstructure of PCL–Ag, facilitating wound humidity regulation. Inductively coupledplasma-mass spectroscopy (ICP-MS) was employed to study the release profiles of SNPs at different time intervals. Overall,the PCL–Ag 500 ppm sample exerted excellent antibacterial activity against Pseudomonas aeruginosa and Staphylococcusaureus strains and low in vitro cytotoxicity.

        Identification of Trombiculid Chigger Mites Collected on Rodents from Southern Vietnam and Molecular Detection of Rickettsiaceae Pathogen

        Trombiculid “chigger” mites (Acari) are ectoparasites that feed blood on rodents and another animals. A cross-sectional survey was conducted in 7 ecosystems of southern Vietnam from 2015 to 2016. Chigger mites were identified with morphological characteristics and assayed by polymerase chain reaction for detection of rickettsiaceae. Overall chigger infestation among rodents was 23.38%. The chigger index among infested rodents was 19.37 and a mean abundance of 4.61. A total of 2,770 chigger mites were identified belonging to 6 species, 3 genera, and 1 family, and pooled into 141 pools (10–20 chiggers per pool). Two pools (1.4%) of the chiggers were positive for Orientia tsutsugamushi. Ricketsia spp. was not detected in any pools of chiggers. Further studies are needed including a larger number and diverse hosts, and environmental factors to assess scrub typhus.

        Effects of non-vertical linear motions of a hemispherical-float wave energy converter

        <P><B>Abstract</B></P> <P>The hydraulic power-take-off mechanism (HPTO) is one of the most popular methods in wave energy converter (WECs). However, the conventional HPTO with only one direction motion has some drawbacks which limit its power capture capability. This paper proposes an <I>adjustable slope angle wave energy converter</I> (ASAWEC) and investigates the effect of slope angle on the performance of the proposed wave energy converter to find the optimal slope angle with the purpose to increase the power capture capability as well as energy efficiency. A mathematical model of components from a floating buoy to a hydraulic motor was modeled. A small scale WEC test rig was fabricated to verify the power capture capability and efficiency of the proposed system throughout experiments.</P> <P><B>Highlights</B></P> <P> <UL> <LI> This paper proposed an adjustable moving angle wave energy converter (AMAWEC). </LI> <LI> Mathematical model of AMAWEC was presented and simulated. </LI> <LI> An AMAWEC test rig were fabricated to do the experiment in variety of wave conditions and moving angle in the water tank. </LI> <LI> The effect of moving angle on the performance of a wave energy converter was investigated to find the optimal moving angle in each wave condition. </LI> <LI> Finally, the effectiveness of AMAWEC has been verified by experimental results. </LI> </UL> </P>

        <P><B>Abstract</B></P> <P>The photoelectrochemical properties of CdS-sensitized ZnO nanorods grown on Pt-coated WO<SUB>3</SUB> nanoplates are investigated to evaluate their effectiveness in hydrogen production. WO<SUB>3</SUB> nanonanoplates are synthesized on glass substrates, followed by atomic layer deposition of Pt thin films as the terminal electrode to efficiently collect the photo-carriers generated from the ZnO/CdS absorption layers. Optimization of the fabrication process for the 3D hierarchical structure is performed, and the morphology and its effect on the photoelectrochemical performance of the electrodes are carefully studied using scanning electron microscopy, x-ray diffraction, and measurements of the photocurrent density and photo-conversion efficiencies. The enhanced PEC performance is elucidated by the 3D hierarchical geometry of the electrode. The optimized electrode shows a photocurrent density of ∼ 13 mA cm<SUP>−2</SUP> and a conversion efficiency of ∼8.0% at −0.83 V (<I>vs</I>.         A novel control method to maximize the energy-harvesting capability of an adjustable slope angle wave energy converter

        <P><B>Abstract</B></P> <P>This paper introduces a novel control approach to maximizing the output energy of an adjustable slope angle wave energy converter (ASAWEC) with oil-hydraulic power take-off. Different from typical floating-buoy WECs, the ASAWEC is capable of capturing wave energy from both heave and surge modes of wave motions. For different waves, online determination of the titling angle plays a significant role in optimizing the overall efficiency of the ASAWEC. To enhance this task, the proposed method was developed based on a learning vector quantitative neural network (LVQNN) algorithm. First, the LVQNN-based supervisor controller detects wave conditions and directly produces the optimal titling angles. Second, a so-called efficiency optimization mechanism (EOM) with a secondary controller was designed to regulate automatically the ASAWEC slope angle to the desired value sent from the supervisor controller. A prototype of the ASAWEC was fabricated and a series of simulations and experiments was performed to train the supervisor controller and validate the effectiveness of the proposed control approach with regular waves. The results indicated that the system could reach the optimal angle within 2s and subsequently, the output energy could be maximized. Compared to the performance of a system with a vertically fixed slope angle, an increase of 5% in the overall efficiency was achieved. In addition, simulations of the controlled system were performed with irregular waves to confirm the applicability of the proposed approach in practice.</P> <P><B>Highlights</B></P> <P> <UL> <LI> This paper proposes a novel energy maximization algorithm of a sliding-buoy wave energy converter (SBWEC). </LI> <LI> An efficiency optimization mechanism is designed and integrated into the SBWEC. </LI> <LI> The control logic is based on a learning vector quantitative neural network for classifying the wave information. </LI> <LI> The effectiveness of the proposed approach is verified through both simulations and experiments. </LI> </UL> </P>
